Search Engine Optimisation in UK news production
This is an Author's Accepted Manuscript of an article published in Journalism Practice, 5(4), 462 - 477, 2011, copyright Taylor & Francis, available online at: http://www.tandfonline.com/10.1080/17512786.2010.551020.This paper represents an exploratory study into an emerging culture in UK online newsrooms—the practice of Search Engine Optimisation (SEO), which assesses its impact on news production. Comprising a short-term participant observational case study at a national online news publisher, and a series of semi-structured, in-depth interviews with SEO professionals at three further UK media organisations, the author sets out to establish how SEO is operationalised in the newsroom, and what consequences these practices have for online news production. SEO practice is found to be varied and application is not universal. Not all UK news organisations are making the most of SEO even though some publishers take a highly sophisticated approach. Efforts are constrained by time, resources and management support, as well as off-page technical issues. SEO policy is found, in some cases, to inform editorial policy, but there is resistance to the principal of SEO driving decision-making. Several themes are established which call for further research
Necroptosis molecular mechanisms: recent findings regarding novel necroptosis regulators
Necroptosis is a form of programmed necrosis that is mediated by various cytokines and pattern recognition receptors (PRRs). Cells dying by necroptosis show necrotic phenotypes, including swelling and membrane rupture, and release damage-associated molecular patterns (DAMPs), inflammatory cytokines, and chemokines, thereby mediating extreme inflammatory responses. Studies on gene knockout or necroptosis-specific inhibitor treatment in animal models have provided extensive evidence regarding the important roles of necroptosis in inflammatory diseases. The necroptosis signaling pathway is primarily modulated by activation of receptor-interacting protein kinase 3 (RIPK3), which phosphorylates mixed-lineage kinase domain-like protein (MLKL), mediating MLKL oligomerization. In the necroptosis process, these proteins are fine-tuned by posttranslational regulation via phosphorylation, ubiquitination, glycosylation, and protein-protein interactions. Herein, we review recent findings on the molecular regulatory mechanisms of necroptosis.
Antibody-secreting macrophages generated using CpG-free plasmid eliminate tumor cells through antibody-dependent cellular phagocytosis
The non-viral delivery of genes into macrophages, known as hard-to-transfect cells, is a challenge. In this study, the microporation of a CpG-free and small plasmid (pCGfd-GFP) showed high transfection efficiency, sustainable transgene expression, and good cell viability in the transfections of Raw 264.7 and primary bone marrow-derived macrophages. The non-viral method using the pCGfd vector encoding anti-EGFR single-chain Fv fused with Fc (scFv-Fc) generated the macrophages secreting anti-EGFR scFv-Fc. These macrophages effectively phagocytized tumor cells expressing EGFR through the antibody-dependent mechanism, as was proved by experiments using EGFR-knockout tumor cells. Finally, peri-tumoral injections of anti-EGFR scFv-Fc-secreting macrophages were shown to inhibit tumor growth in the xenograft mouse model.
Post-Translational Regulation of ARF: Perspective in Cancer
Tumorigenesis can be induced by various stresses that cause aberrant DNA mutations and unhindered cell proliferation. Under such conditions, normal cells autonomously induce defense mechanisms, thereby stimulating tumor suppressor activation. ARF, encoded by the CDKN2a locus, is one of the most frequently mutated or deleted tumor suppressors in human cancer. The safeguard roles of ARF in tumorigenesis are mainly mediated via the MDM2-p53 axis, which plays a prominent role in tumor suppression. Under normal conditions, low p53 expression is stringently regulated by its target gene, MDM2 E3 ligase, which induces p53 degradation in a ubiquitin-proteasome-dependent manner. Oncogenic signals induced by MYC, RAS, and E2Fs trap MDM2 in the inhibited state by inducing ARF expression as a safeguard measure, thereby activating the tumor-suppressive function of p53. In addition to the MDM2-p53 axis, ARF can also interact with diverse proteins and regulate various cellular functions, such as cellular senescence, apoptosis, and anoikis, in a p53-independent manner. As the evidence indicating ARF as a key tumor suppressor has been accumulated, there is growing evidence that ARF is sophisticatedly fine-tuned by the diverse factors through transcriptional and post-translational regulatory mechanisms. In this review, we mainly focused on how cancer cells employ transcriptional and post-translational regulatory mechanisms to manipulate ARF activities to circumvent the tumor-suppressive function of ARF. Larvicidal composite alginate hydrogel combined with a Pickering emulsion of essential oil
Sulfonated cellulose nanocrystals (S-CNCs) can be used to encapsulate thyme white essential oil (EO) that is volatile and immiscible with water. S-CNCs form a Pickering emulsion (PE) with EOs and the micron-scale PEs are embedded in sodium alginate (SA) to form macroscale hydrogel beads. The incorporation of PEs with SA is confirmed with FTIR, XRD, SEM and confocal microscopic characterizations and the release behavior is monitored to understand the time-dependent biological activity of the EOs. The larvicidal performance of the SA-PE composite hydrogel beads is investigated with Aedes albopictus (Skuse) larvae. The larvicidal activity is higher for SA/PE hydrogel beads prepared at 0.5 % CaCl2 than 0.75 or 1.0 % CaCl2 due to their higher release rate.N
The Intelligent SEO
Práce se zabývá problematikou optimalizace pro vyhledávač Google a s ní spojenou problematikou vyhledávání. Rozebírá nejdůležitější techniky SEO a jejich efekty. Celý teoretický základ tvoří ucelený přehled problematiky SEO, který je zasazen do kontextu ostatních souvisejících disciplín. Práce se opatrně snaží vytyčit směr, kterým se SEO bude vyvíjet v následujících letech. Vymezený teoretický rámec, je následně ověřen na případové studii, která v poslední chvíli umožnila zčásti potvrdit i naznačenou budoucí podobu SEO. Účelem práce bylo rozebíranou problematiku popsat netradičně s menším důrazem na technické detaily a s větším důrazem na pochopení dlouhodobější strategie optimalizací pro vyhledávače. Cílem případové studie bylo prokázat vliv aplikace teoretických poznatků v praxi. Teoretické poznatky v této práci jsou výsledkem syntézy myšlenek užšího spektra autorů, kteří vhodně chápou kontext a smysl SEO. Shodou okolností se autorovi povedlo tuto syntézu myšlenek, ve které vycházel i z jeho vlastních zkušeností, prokázat na události, která uzavírá případovou studii. V práci je prokázán vliv optimalizací pro vyhledávače na nárůst návštěvnosti a obratu internetového obchodu a byl pozorován zpožděný negativní vliv manipulativního jednání za účelem ošálení vyhledávačů.Thesis describes search engine optimization for Google and principles of search. It clears up the most important optimization techniques and its effects. The theoretical part makes a consistent summary of SEO situated in the context of other related disciplines. The thesis cautiously tries predicting the direction of SEO development in next few years. Theoretical scope is verified in a case study that made possible to prove the mentioned future prediction of SEO development. The goals were to describe the topic in a nonconventional way with less focus on technical details and more emphasis on the understanding of long-term SEO strategy in search engines. Main goal of the case study was to prove the impact of theoretical knowledge applied in practice. Theory in this thesis is an outcome of synthesized narrow spectra of ideas of authors who understand the context and the point of SEO in the proper way. By a chance the author managed to prove his synthesis on an occurrence closing the case study. Thesis proves the effects of search engine optimization on e-commerce traffic and sales growth and observes the negative effect of manipulative behavior for the purpose of search engine deception
