    Interactions Between Spermine-Derivatized Tentacle Porphyrins and The Human Telomeric DNA G-Quadruplex

    G-rich DNA sequences have the potential to fold into non-canonical G-Quadruplex (GQ) structures implicated in aging and human diseases, notably cancers. Because stabilization of GQs at telomeres and oncogene promoters may prevent cancer, there is an interest in developing small molecules that selectively target GQs. Herein, we investigate the interactions of meso-tetrakis-(4-carboxysperminephenyl)porphyrin (TCPPSpm4) and its Zn(II) derivative (ZnTCPPSpm4) with human telomeric DNA (Tel22) via UV-Vis, circular dichroism (CD), and fluorescence spectroscopies, resonance light scattering (RLS), and fluorescence resonance energy transfer (FRET) assays. UV-Vis titrations reveal binding constants of 4.7 × 106 and 1.4 × 107 M−1 and binding stoichiometry of 2–4:1 and 10–12:1 for TCPPSpm4 and ZnTCPPSpm4, respectively. High stoichiometry is supported by the Job plot data, CD titrations, and RLS data. FRET melting indicates that TCPPSpm4 stabilizes Tel22 by 36 ± 2 °C at 7.5 eq., and that ZnTCPPSpm4 stabilizes Tel22 by 33 ± 2 °C at ~20 eq.; at least 8 eq. of ZnTCPPSpm4 are required to achieve significant stabilization of Tel22, in agreement with its high binding stoichiometry. FRET competition studies show that both porphyrins are mildly selective for human telomeric GQ vs duplex DNA. Spectroscopic studies, combined, point to end-stacking and porphyrin self-association as major binding modes. This work advances our understanding of ligand interactions with GQ DNA

    Aggregate breakdown of nanoparticulate titania

    Six nanosized titanium dioxide powders synthesized from a sulfate process were investigated. The targeted end-use of this powder was for a de-NOx catalyst honeycomb monolith. Alteration of synthesis parameters had resulted principally in differences in soluble ion level and specific surface area of the powders. The goal of this investigation was to understand the role of synthesis parameters in the aggregation behavior of these powders. Investigation via scanning electron microscopy of the powders revealed three different aggregation iterations at specific length scales. Secondary and higher order aggregate strength was investigated via oscillatory stress rheometry as a means of simulating shear conditions encountered during extrusion. G' and G'' were measured as a function of the applied oscillatory stress. Oscillatory rheometry indicated a strong variation as a function of the sulfate level of the particles in the viscoelastic yield strengths. Powder yield stresses ranged from 3.0 Pa to 24.0 Pa of oscillatory stress. Compaction curves to 750 MPa found strong similarities in extrapolated yield point of stage I and II compaction for each of the powders (at approximately 500 MPa) suggesting that the variation in sulfate was greatest above the primary aggregate level. Scanning electron microscopy of samples at different states of shear in oscillatory rheometry confirmed the variation in the linear elastic region and the viscous flow regime. A technique of this investigation was to approach aggregation via a novel perspective: aggregates are distinguished as being loose open structures that are highly disordered and stochastic in nature. The methodology used was to investigate the shear stresses required to rupture the various aggregation stages encountered and investigate the attempt to realign the now free-flowing constituents comprising the aggregate into a denser configuration. Mercury porosimetry was utilized to measure the pore size of the compact resulting from compaction via dry pressing and tape casting secondary scale aggregates. Mercury porosimetry of tapes cast at 0.85 and 9.09 cm/sec exhibited pore sizes ranging from 200-500 nm suggesting packing of intact micron-sized primary aggregates. Porosimetry further showed that this peak was absent in pressed pellets corroborating arguments of ruptured primary aggregates during compaction to 750 MPa.Ph.D.Includes bibliographical references (p. 166-170)

    El primer número de Conversaciones a Paul Philippot, cuya pluma clara y elegante ha inspirado a más de una generación de conservadores en el mundo entero, y a quien se le recuerda aún por su paso por México en los comienzos del Centro Churubusco (Coordinación Nacional de Conservación del Patrimonio Cultural), a finales de la década de 1960 e inicios de la de 1970. Autor de numerosos escritos que discurren sobre el desarrollo de la disciplina y las implicaciones de las intervenciones de conservación, así como temas específicos tales como la pátina, la limpieza y la reintegración de lagunas, al igual que numerosos textos fundamentales sobre conservación de pintura mural y sobre escultura policromada, y por último, co-autor de la Carta de Venecia, la elección de alguno de sus textos inéditos en español no era tarea sencilla. Al final, se eligió “L’oeuvre d’art, le temps et la restauration”, publicado originalmente en 1995, en la revista Histoire de l’art, a quien agradecemos la posibilidad de replicar aquí el texto original. Este artículo abarca muchos de los temas que han marcado la producción escrita de Paul Philippot.     Reforming institutions for service delivery : a framework for development assistance with an application to the health, nutrition, and population portfolio

    World Development Report 1997: The State in a Changing World (report no. 17300) argued that institutions-the rules of the game that govern production and exchange-shape a country's prospects for sustained market-led growth. The author provides an institutional framework for service delivery, an essential component of state capability. He applies this framework to an evaluation of Bank support for service delivery in the health, nutrition, and population sector. He argues for greater institutional pluralism in the ways the World Bank does business in infrastructure, rural, and social sectors, but cautions against making efficient service delivery an issue of"state versus market."The Bank and its clients face the challenge of fitting menus of"better practice"delivery options to maps of institutional reality. In the health, nutrition, and population sector, the Bank should (1) unbundle and categorize essential health and clinical services according to goods characteristics and (2) integrate country knowledge into operations through upstream assessments of state, political, and social institutions. Overall, the Bank has made progress toward a"goods characteristics"approach, particularly in infrastructure and some rural services-but it has lagged in the social sectors, where support remains largely technocratic. Cross-sector comparisons reveal four generations of support for service delivery. First-generation support focused mainly on physical implementation of projects. Second-generation interventions, which characterized most social service interventions, focused on improving the financial and organizational viability of implementing agencies through technical assistance. Third-generation support was marked by significant unbundling of service delivery activities and clearer links to goods characteristics. In irrigation (1982-94), telecommunications (1980s-present), and transport (1990s), the one-size-fits-all monopoly model gave way to a range of options based on greater private sector and citizen participation in delivery. These included leases, concessions, outsourcing, and contracting as well as building, operating, transfer, and turnover schemes. Fourth-generation interventions are works-in-progress and represent efforts to develop new governance arrangements that systematically combine competition, voice, and hierarchy in the design, delivery, and monitoring of Bank projects. The Bank has a poor track record building country knowledge of institutional endowments that affect service delivery.     The Rhetoric of Widening Participation in Higher Education and its Impact:ending the barriers against disabled people

    This book offers a critical investigation of the exclusion of individuals described as having ‘learning difficulties’ from participation in higher education. Using a postmodernist framework, the author explores the insights and experiences of a theatre group attempting to develop an undergraduate degree programme in the performing arts. In doing so, he provides a theoretical map of insights into discourses of power and knowledge, and makes transparent competing and contradictory discursive practices. Suggesting that ‘learning difficulties’ is a constructed and re-constructed discourse serving normative interests, the author demonstrates that despite the rhetoric of widening participation, individuals are intentionally beset by barriers, silenced and excluded from degree level participation. The author calls for a radical re-think of the notion of ‘learning difficulties’, segregated provision, access to employment in theatre, and critically questions the notion of participation in higher education. This pioneering volume will appeal to students and scholars of inclusive education, (critical) disability studies, cultural studies and the sociology of education
