As earlier diagnosis and new treatment modalities increase survival for these women, health care practitioners must address long-term problems secondary to cancer therapy. The development of severe hot flashes, affecting up to 85% of breast cancer survivors, can significantly affect quality of life. Many hot flash treatments are ineffective, have intolerable side effects, or are contraindicated for breast cancer survivors. Research supports the use of acupuncture as an effective, safe treatment for minimizing hot flashes in breast cancer survivors.Purpose: The purpose of this evidence-based practice project was to determine if acupuncture is effective at decreasing the number and severity of hot flashes and improving sleep in breast cancer survivors at an urban hospital. The acceptability of acupuncture as a treatment for hot flashes was also explored.Design: The study design for this project was a quasi-experimental pretest-posttest design. Qualitative data was also obtained by interview after participants completed acupuncture treatments.Methods: Eligible women completed a Pittsburgh Sleep Quality Index (PSQI) and recorded hot flash frequency and severity in a hot flash diary for a week before and after acupuncture treatment. Acceptability of acupuncture was assessed using a Likert Scale (1-10). Participants received twice weekly acupuncture treatments for four weeks. Pre and post acupuncture hot flash frequency and severity, PSQI sleep disturbance score, and acceptability ratings were compared. Participants were interviewed and descriptions of the acupuncture experience examined for themes.Results: Paired t-test revealed significant improvement in night time hot flash frequency (p = 0.04) and severity (p = 0.001). Daytime hot flash severity significantly improved (p < 0.001) although no significant difference was noted in hot flash frequency (p = 0.089). PSQI score was significantly improved following acupuncture (p = 0.021). No significant difference was found between pre and post acceptability scores (p = 0.428). Participants' descriptions of acupuncture encompassed three themes; acupuncture is (1) effective, (2) relaxing, and (3) painful at times. Date of Acceptance: 06/10/2015The spatial uniformity of GaSb- and InAs substrate-based structures containing type II quantum wells was probed by means of large-scale photoluminescence (PL) mapping realized utilizing a Fourier transform infrared spectrometer. The active region was designed and grown in a form of a W-shaped structure with InAs and GaInSb layers for confinement of electrons and holes, respectively. The PL spectra were recorded over the entire 2-in. wafers, and the parameters extracted from each spectrum, such as PL peak energy position, its linewidth and integrated intensity, were collected in a form of two-dimensional spatial maps. Throughout the analysis of these maps, the wafers' homogeneity and precision of the growth procedure were investigated. A very small variation of PL peak energy over the wafer indicates InAs quantum well width fluctuation of only a fraction of a monolayer and hence extraordinary thickness accuracy, a conclusion further supported by high uniformity of both the emission intensity and PL linewidth.Peer reviewe

    Organometallic compounds as anti-cancer agents : interaction with DNA and migration in cells

    De nouveaux composés organométalliques, basés sur le ruthénium, possèdent une activité anticancéreuse aussi efficace que les complexes de platine utilisés jusqu'à présent, mais présentent une neurotoxicité beaucoup plus faible. Le but principal de cette thèse est d'étudier l'affinité de ces composés dérivés du ruthénium (RDC) avec l'ADN. Nous avons étudié trois composés organométalliques différents, et nous sommes posés deux types de questions. Dans un premier temps, nous avons étudié, à l'échelle de la molécule unique, les interactions entre un RDC et une molécule d'ADN double brin. Dans un second temps, nous avons mesuré la cinétique d'entrée des RDC à l'intérieur des cellules. Nous nous sommes servis Förster Resonant Energy Transfer technique pour mesurer le degré d'avancement de la réaction de complexation de l'ADN avec les composés dérivés du ruthénium. Nous avons ensuite réalisé des mesures mécaniques sur une molécule unique, dans le but d'obtenir des informations sur la structure du complexe formé par l'ADN et un RDC. Nous avons ainsi pu observer, selon la nature des ligands porté par le RDC, une intercalation ou une complexation dans un des sillons. Pour obtenir la cinétique d'entrée dans les cellules, nous avons utilisé les propriétés de luminescence des RDC, et les avons détectés au sein de la cellule par microscopie confocale de fluorescence. Nous avons obtenu dans cette thèse le deux résultats suivants : les RDC présentent une forte affinité pour l'ADN et induisent un changement de structure qui dépend des ligands qui les composent, Nous avons observé une augmentation de l'intensité fluorescée par le noyau, et donc une présence de RDC dans le noyau, la plus grande quantité de RDC se trouve localisé autour du noyau.New organometallic derivatives, based on Ruthenium, possess an anti-cancer activity as efficient as platinum complexes (widely used) but exhibit less neurotoxicity. The main goal of this PhD research was an examination of Ruthenium Derived Compounds (RDCs) which display strong affinity to DNA. Three structurally different RDCs were considered in two significant aspects. The first aspect was to understand the interaction between RDC and dsDNA at the single molecule level, its mechanism and strength. The second one concerned the cellular uptake of RDC. We used Förster Resonant Energy Transfer method to obtain the affinity, and Optical Tweezers to obtain the structural information of RDC-DNA complex. We checked the migration of RDC in cells using confocal microscopy. It was found that RDC induces DNA structure changes in vitro by intercalation or groove binding depending on ligands with the highest affinity to DNA. Moreover RDC enters the cell through active transport, however the exact mechanism remains unknown. RDC is stored inside the cell around nucleus and with lower concentration inside

    Praca dotyczy fotografii z powietrza wykonywanej przy pomocy balonu bądź latawca jako narzędzia badawczego w archeologii. Opisano w niej historię powstania i wykorzystywania tej metody, przyrównano ją do innych technik wykonywania zdjęć z lotu ptaka, a także przedstawiono przykłady jej wykorzystania w archeologii. Dodatkowo w dysertacji doktorskiej znalazły się informacje o technice i sposobach wykonywania zdjęć z aparatów fotograficznych podwieszonych pod aerodynami i aerostatami bez napędu i na uwięzi, a także sposobach przetwarzania fotografii technikami komputerowymi. Autor analizuje ponadto przydatność i możliwości takiego sposobu uzyskiwania dokumentacji w badaniach archeologicznych i prezentuje wyniki z przykładowych opracowań. Study concerns aerial photography from kites and balloons as a tool for archaeological research. The author describes the history of the invention and use of this method, compares it to other techniques for taking pictures from the air, and shows examples of its use in archeology. In addition, doctoral dissertation includes information about the technique and how to take pictures with cameras suspended under kites and balloons, as well as methods for processing photographs by computer techniques. The author also analyzes the usefulness and capabilities of this method for obtaining documentation in archaeological research and presents the results of case-studies
