    Who am I?: questioning identity in three short stories by Liliana Heker

    This thesis focuses on the question of identity in three short stories by the Argentine writer Liliana Heker (1943-); "La fiesta ajena" (The Stolen Party), "Berkeley o Mariana del universo" (Bishop Berkeley or Mariana of the Universe), and "Georgina Requeni o la elegida" (Georgina Requeni or The Chosen One). Heker's texts are founded on the assumption that a sense of identity arises from the mediation of social objective discourses (who the world tells me I am) and the subjective concept of self (who I think I am). The author began publishing her works in 1966, the same year that Argentina came under the military rule of Juan Carlos Ongania. The synchronicity of Heker's foray into literature and the beginning of a period of military governments whose repression and violence spanned three decades is symbolic of Heker's function as literary historiographer. Through the examination of moments of epiphany that alter one's identity, slightly or tremendously, Heker enunciates the protagonists' identity against the backdrop of the collective social consciousness and lays bare cultural assumptions of class, power, and gender that are crucial for understanding Argentine society. Heker's belief that "toda polemica es enriquecedora en la medida que permite la confrontation de dos sistemas de ideas" (all polemic is enriching in that it allows for the confrontation of two systems of ideas) is the reason she creates historically anchored texts that question the role of the dialectic of social discourses in the formation of identity.Arts, Faculty ofFrench, Hispanic, and Italian Studies, Department ofGraduat

    A highly focused antigen receptor repertoire characterizes γδ T cells that are poised to make IL-17 rapidly in naive animals

    Interleukin (IL)-17 plays a key role in immunity. In acute infections, a rapid IL-17 response must be induced without prior antigen exposure, and γδ T cells are the major initial IL-17 producers. In fact, some γδ T cells make IL-17 within hours after an immune challenge. These cells appear to acquire the ability to respond to IL-1 and IL-23 and to make IL-17 naturally in naïve animals. They are known as the natural Tγδ17 (nTγδ17) cells. The rapidity of the nTγδ17 response, and the apparent lack of explicit T cell receptor (TCR) engagement for its induction have led to the view that this is a cytokine (IL-1, IL-23)-mediated response. However, pharmacological inhibition or genetic defects in TCR signaling drastically reduce the nTγδ17 response and/or their presence. To better understand antigen recognition in this rapid IL-17 response, we analyzed the antigen receptor repertoire of IL-1R+/IL-23R+ γδ T cells, a proxy for nTγδ17 cells in naïve animals, using a barcode-enabled high throughput single-cell TCR sequence analysis. We found that regardless of their anatomical origin, these cells have a highly focused TCR repertoire. In particular, the TCR sequences have limited V gene combinations, little or no junctional diversity and much reduced or no N region diversity. In contrast, IL-23R- cells at mucosal sites similar to most of the splenic γδ T cells and small intestine epithelial γδ lymphocytes expressed diverse TCRs. This remarkable commonality and restricted repertoire of IL-1R+/IL-23R+ γδ T cells underscores the importance of antigen recognition in their establishment/function

    An overview of the characteristics of the novel avian influenza A H7N9 virus in humans

    The novel avian influenza A H7N9 virus which caused the first human infection in Shanghai, China; was reported on the 31st of March 2013 before spreading rapidly to other Chinese provinces and municipal cities. This is the first time the low pathogenic avian influenza A virus has caused human infections and deaths; with cases of severe respiratory disease with pneumonia being reported. There were 440 confirmed cases with 122 fatalities by 16 May 2014; with a fatality risk of approximately 28%. The median age of patients was 61 years with a male-to-female ratio of 2.4:1. The main source of infection was identified as exposure to poultry and there is so far no definitive evidence of sustained person-to-person transmission. The neuraminidase inhibitors (NI), namely oseltamivir, zanamivir and peramivir; have shown good efficacy in the management of the novel H7N9 virus. Treatment is recommended for all hospitalized patients, and for confirmed and probable outpatient cases; and should ideally be initiated within 48 hours of the onset of illness for the best outcome. Phylogenetic analysis found that the novel H7N9 virus is avian in origin and evolved from multiple reassortments of at least four origins. Indeed the novel H7N9 virus acquired human adaptation via mutations in its eight RNA gene segments. Enhanced surveillance and effective global control are essential to prevent pandemic outbreaks of the novel H7N9 virus

    Fermentation conditions that affect clavulanic acid production in Streptomyces clavuligerus: a systematic review

    The β-lactamase inhibitor, clavulanic acid is frequently used in combination with β-lactam antibiotics to treat a wide spectrum of infectious diseases. Clavulanic acid prevents drug resistance by pathogens against these β-lactam antibiotics by preventing the degradation of the β-lactam ring, thus ensuring eradication of these harmful microorganisms from the host. This systematic review provides an overview on the fermentation conditions that affect the production of clavulanic acid in the firstly described producer, Streptomyces clavuligerus. A thorough search was conducted using predefined terms in several electronic databases (PubMed, Medline, ScienceDirect, EBSCO), from database inception to June 30th 2015. Studies must involve wild-type Streptomyces clavuligerus, and full texts needed to be available. A total of 29 eligible articles were identified. Based on the literature, several factors were identified that could affect the production of clavulanic acid in S. clavuligerus. The addition of glycerol or other vegetable oils (e.g. olive oil, corn oil) could potentially affect clavulanic acid production. Furthermore, some amino acids such as arginine and ornithine, could serve as potential precursors to increase clavulanic acid yield. The comparison of different fermentation systems revealed that fed-batch fermentation yields higher amounts of clavulanic acid as compared to batch fermentation, probably due to the maintenance of substrates and constant monitoring of certain entities (such as pH, oxygen availability, etc.). Overall, these findings provide vital knowledge and insight that could assist media optimization and fermentation design for clavulanic acid production in S. clavuligerus

    Strain Alleviation in an Isomorphous Series of Lanthanide 2-Nitroterephthalates [Ln2(TPNO2)3(H2O)2]·2H2O (Ln = Pr – Lu, except Pm)

    An extended series of trivalent lanthanide 2-nitroterephthalates, [Ln2(TPNO2)3(H2O)2]·2H2O, (Ln = Pr through Lu, except Pm) were synthesized hydrothermally from Ln2O3 and 2-nitroterephthalic acid (H2TPNO2) at 170 °C in Teflon lined Parr steel autoclaves, and were characterized via single crystal X-ray diffraction, powder X-ray diffraction, FT-IR spectroscopy, elemental analyses, and thermogravimetric analyses. All [Ln2(TPNO2)3(H2O)2]·2H2O coordination polymers are isomorphous, crystallizing in the monoclinic crystal system with space group C2/c. The metal centers in all networks possess the coordination number 8, while forming a three-dimensional extended lattice. Two metal centers form Ln2O14 entities, comprising crystallographically identical LnO8 polyhedra, connected via edge-sharing, utilizing two carboxylate O-atoms. These Ln2O14 units are separated along the a- and b-axes by individual 2-nitroterephthalate linkers, while being closely connected along the c-axis via two carboxylate groups on each side. Compared to small inorganic anions, the rather flexible 2-nitroterephthalate seems to allow for the unobstructed decrease in size of the LnO8 polyhedra as Ln3+ ionic radii decrease towards the heavier Ln elements. Hence, the structural parameters of the crystal lattice adjust gradually without noticeable strain buildup along the series resulting in isomorphous arrangements for all networks.     Weathering uncertainty traditional knowledge for climate change assessment and adaptation

    This UNESCO report looks into the damaging effects of climate change on Indigenous cultures.When considering climate change, indigenous peoples and marginalized populations warrant particular attention. Impacts on their territories and communities are anticipated to be both early and severe due to their location in vulnerable environments, including small islands, high-altitude zones, desert margins and the circumpolar Arctic. Indeed, climate change poses a direct threat to many indigenous societies due to their continuing reliance upon resource-based livelihoods. Heightened exposure to negative impacts, however, is not the only reason for specific attention and concern. As many indigenous societies are socially and culturally distinct from mainstream society, decisions, policies and actions undertaken by the majority, even if well-intended, may prove inadequate, ill-adapted, and even inappropriate. There is therefore a need to understand the specific vulnerabilities, concerns, adaptation capacities and longer-term aspirations of indigenous peoples and marginalized communities throughout the world.     Assessing the Radiation Response of Lung Cancer with Different Gene Mutations Using Genetically Engineered Mice

    Purpose: Non-small cell lung cancers (NSCLC) are a heterogeneous group of carcinomas harboring a variety of different gene mutations. We have utilized two distinct genetically engineered mouse models of human NSCLC (adenocarcinoma) to investigate how genetic factors within tumor parenchymal cells influence the in vivo tumor growth delay after one or two fractions of radiation therapy (RT). Methods and Materials: Primary lung adenocarcinomas were generated in vivo in mice by intra-nasal delivery of an adenovirus expressing Cre-recombinase. Lung cancers expressed oncogenic K-rasG12D and were also deficient in one of two tumor suppressor genes: p53 or Ink4a/ARF. Mice received no radiation treatment or whole lung irradiation in a single fraction (11.6 Gy) or in two 7.3 Gy fractions (14.6 Gy total) separated by 24 hours. In each case, the biologically effective dose (BED) equaled 25 Gy10. Response to RT was assessed by micro-CT two weeks after treatment. Quantitative reverse transcription-polymerase chain reaction (qRT-PCR) and immunohistochemical staining were performed to assess the integrity of the p53 pathway, the G1 cell-cycle checkpoint, and apoptosis. Results: Tumor growth rates prior to RT were similar for the two genetic variants of lung adenocarcinoma. Lung cancers with wild-type p53 (LSL-Kras; Ink4a/ARFFL/FL mice) responded better to two daily fractions of 7.3 Gy compared to a single fraction of 11.6 Gy (P=0.002). There was no statistically significant difference in the response of lung cancers deficient in p53 (LSL-Kras; p53FL/FL mice) to a single fraction (11.6 Gy) compared to 7.3 Gy x 2 (P=0.23). Expression of the p53 target genes p21 and PUMA were higher and BrdU uptake was lower after RT in tumors with wild-type p53. Conclusions: Using an in vivo model of malignant lung cancer in mice, we demonstrate that the response of primary lung cancers to one or two fractions of RT can be influenced by specific gene mutations
