Angiotensin II induces soluble fms-Like tyrosine kinase-1 release via calcineurin signaling pathway in pregnancy
Maternal endothelial dysfunction in preeclampsia is associated with increased soluble fms-like tyrosine kinase-1 (sFlt-1), a circulating antagonist of vascular endothelial growth factor and placental growth factor. Angiotensin II (Ang II) is a potent vasoconstrictor that increases concomitant with sFlt-1 during pregnancy. Therefore, we speculated that Ang II may promote the expression of sFlt-1 in pregnancy. Here we report that infusion of Ang II significantly increases circulating levels of sFlt-1 in pregnant mice, thereby demonstrating that Ang II is a regulator of sFlt-1 secretion in vivo. Furthermore, Ang II stimulated sFlt-1 production in a dose- and time-dependent manner from human villous explants and cultured trophoblasts but not from endothelial cells, suggesting that trophoblasts are the primary source of sFlt-1 during pregnancy. As expected, Ang II-induced sFlt-1 secretion resulted in the inhibition of endothelial cell migration and in vitro tube formation. In vitro and in vivo studies with losartan, small interfering RNA specific for calcineurin and FK506 demonstrated that Ang II-mediated sFlt-1 release was via Ang II type 1 receptor activation and calcineurin signaling, respectively. These findings reveal a previously unrecognized regulatory role for Ang II on sFlt-1 expression in murine and human pregnancy and suggest that elevated sFlt-1 levels in preeclampsia may be caused by a dysregulation of the local renin/angiotensin system

Hemodynamic regulation of metalloendopeptidases EC3.4; 24.15 and EC3.4; 24.16: expression and function in the vascular endothelium
Hemodynamic forces, namely shear stress and cyclic strain, have been well characterised as modulators of vascular endothelial function, and have been assigned an important role in the maintainence of vascular tone, haemostasis, and regulation of vascular growth and health. They exert their influence in part by effecting changes in the production and release of vasoactive compounds by the endothelium, and by effecting changes in the levels and activity of various enzymes. Thimet oligopeptidase (EC3.4.24.15, EP24.15) and neurolysin (EC3.4.24.16, EP24.15) are closely related zinc metalloendopeptidases that have been shown to be expressed and active in the vascular endothelium. Their substrates include the vasoactive peptides bradykinin and angiotensin I, which have been identified as important regulators of both blood pressure and angiogenic processes. Other related peptidases, namely endothelin converting enzyme (ECE) and angiotensin converting enzyme (ACE), have been shown to be regulated by hemodynamic forces in the vascular endothelium. As such EP24.15 and EP24.16 are likely candidates for regulation by hemodynamic forces.
In this regard we have investigated the effect of cyclic strain on the expression and activty of EP24.15 and EP24.16 in cultured bovine aortic endothelial cells (BAECs). We have shown that exposure to cyclic strain significantly increases the mRNA expression as well as both the cellular and secreted activity of both enzymes. We have demonstrated that up-regulation of both enzymes is dependent on Gi-protein mediated signalling, although with varying Gia/G(3y subunit specificity for either enzyme. Using immunocytochemistry, we have also demonstrated a strain-dependent increase in EP24.15 protein expression within the nucleus and cytoplasm in parallel with an increase in membrane associated EP 24.15
The effects of strain on the ability of BAECs in culture to cleave both Ang I, and BK in an EC24.15/EC24.16 specific manner was also studied. We observed that exposure to cyclic strain induces a significant increase in the EP24.15 specific hydrolysis of both exogenously added BK and Ang I.
The potential of the observed effects of cyclic strain on EP24.15 to effect changes in endothelial cell function were also examined. Use of the dual EP24.15/EP24.16 inhibitor, cFP-AAF-pAB, and the EP24.15 specific antisense, FLIP, was seen to significantly attenuate cyclic strain-induced endothelial cell tubule formation and migration. We also found that the effects of FLIP transfection on cyclic strain-induced endothelial cell tubule formation could be largely reversed by addition of exogenous Ang-(l-7). Taken together these results suggest that strain-induced endothelial cell angiogenesis and migration putatively involves EP24.15 cleavage of vasoactive peptide substrates
Fas-induced apoptosis of alveolar epithelial cells requires ANG II generation and receptor interaction
Recent works from this laboratory demonstrated potent inhibition of Fas-induced apoptosis in alveolar epithelial cells (AECs) by the angiotensin-converting enzyme (ACE) inhibitor captopril [B. D. Uhal, C. Gidea, R. Bargout, A. Bifero, O. Ibarra-Sunga, M. Papp, K. Flynn, and G. Filippatos. Am. J. Physiol. 275 ( Lung Cell. Mol. Physiol. 19): L1013–L1017, 1998] and induction of dose-dependent apoptosis in AECs by purified angiotensin (ANG) II [R. Wang, A. Zagariya, O. Ibarra-Sunga, C. Gidea, E. Ang, S. Deshmukh, G. Chaudhary, J. Baraboutis, G. Filippatos and B. D. Uhal. Am. J. Physiol. 276 ( Lung Cell. Mol. Physiol. 20): L885–L889, 1999]. These findings led us to hypothesize that the synthesis and binding of ANG II to its receptor might be involved in the induction of AEC apoptosis by Fas. Apoptosis was induced in the AEC-derived human lung carcinoma cell line A549 or in primary AECs isolated from adult rats with receptor-activating anti-Fas antibodies or purified recombinant Fas ligand, respectively. Apoptosis in response to either Fas activator was inhibited in a dose-dependent manner by the nonthiol ACE inhibitor lisinopril or the nonselective ANG II receptor antagonist saralasin, with maximal inhibitions of 82 and 93% at doses of 0.5 and 5 μg/ml, respectively. In both cell types, activation of Fas caused a significant increase in the abundance of mRNA for angiotensinogen (ANGEN) that was unaffected by saralasin. Transfection with antisense oligonucleotides against ANGEN mRNA inhibited the subsequent induction of Fas-stimulated apoptosis by 70% in A549 cells and 87% in primary AECs (both P < 0.01). Activation of Fas increased the concentration of ANG II in the serum-free extracellular medium 3-fold in primary AECs and 10-fold in A549 cells. Apoptosis in response to either Fas activator was completely abrogated by neutralizing antibodies specific for ANG II ( P < 0.01), but isotype-matched nonimmune immunoglobulins had no significant effect. These data indicate that the induction of AEC apoptosis by Fas requires a functional renin-angiotensin system in the target cell. They also suggest that therapeutic control of AEC apoptosis is feasible through pharmacological manipulation of the local renin-angiotensin system. </jats:p

Masasalamin ang ispiritualidad ng mga Pilipino sa iba’t ibang nakagawian na nila sa kanilang relihiyon. Sa mga relihiyosong aktibidades ng mga Pilipino, pangkaraniwan na ang magkaroon sila ng isang debosyon depende sa kanilang lugar. Isa sa mga prominenteng debosyon sa Pilipinas sa tradisyon ng Romano Katolikong Kristiyano ay ang debosyon sa Sto. Entierro. Tatalakayin ng kasalukuyang pananaliksik ang isang debosyon sa Pampanga kay Apung Mamacalulu. Ito ay isang debosyon na nakaugnay din sa kamalayan at kultura nating mga Pilipino. Nagsagawa ng sarbey ang mananaliksik upang malaman ang mga ginagawa ng mga deboto sa Shrine ni Apung Mamacalulu. Inalam din ng mananaliksik ang mga rason kung bakit sila nagdedebosyon kay Apu at kung ano ang naidudulot nito sa kanilang buhay. Ginawan ng kwalitatibong analisis ang datos. Pinapakita ng mga resulta na ang mga pumpunta sa Apu ay tinitiyak na mahawakan ang imahen, nagsisimba, nagkukumpisal at nagtitirik din ng mga kandila. Sila ay nagdedebosyon para humingi ng tulong at gabay kay Apu pati na rin ang magpasalamat sa kanya sa mga biyayang natatanggap nila. Ang lalaki sa pelikula: Ang toxic masculinity sa mga piling pelikula
Mula pa noon, patuloy na inilalarawan ang mga lalaki sa pelikula bilang marahas, sekswal, matapang, at iba pang katangiang pumapaloob sa isang patriyarkiyang sistema. Sa pagsusuri sa kung paano ipinapakita sa mga pelikula ang toxic masculinity, pinili ng mananaliksi ang mga pelikula kaugnay sa konsepto ng toxic masculinity na a.) batay sa genre romcom, romance drama, action; b.) batay sa protrayal na lalaki ang isa sa pangunahing karakter; at c.) batay sa social function ng karakter na nakabatay sa lipunang kaniyang ginagalawan (pagsusuri ng social roles ng lalaki vis-à-vis pamilya niya, karelasyon, kaibigan, katrabaho, at iba pa). Ang mga pelikulang ito ang ay “Hello, Love, Goodbye” (2019), A Second Chance (2015), at Ang Panday (2017). Sa pagsusuri ng nilalaman ng mga pelikula, ang mga tauhang ginampanan ng mga lalaking artista ay sasailalim sa pagsisiyasat, partikular na nakatuon sa kung paano sila inilarawan at ipinakita sa pamamagitan ng kanilang: pisikal na katangian, katayuang sosyo-ekonomiko, wika, personalidad at pag-uugali, at interpersonal na relasyon. Ang pagbibigay naman ng kahulugan ay nagmula sa nilalaman ng midya na nilapatan ng mga konsepto ng katanyagan (prominence), pagtrato (treatment), at tono (tone). Ang mga natukoy na representasyon ng mga karakter at ang pagtukoy sa katanyagan, pagtrato at tono, ay sinuri sa ilalim ng pagpapakahulugan ni Kupers ng toxic masculinity.
