    CTL ELISPOT AssayCytotoxic T-Cells

    No full text
Enzyme-linked immune absorbent spot (Elispot) is a quantitative method for measuring relevant parameters of T cell activation. The sensitivity of Elispot allows the detection of low-frequency antigen-specific T cells that secrete cytokines and effector molecules, such as granzyme B and perforin. Cytotoxic T cell (CTL) studies have taken advantage with this high-throughput technology by providing insights into quantity and immune kinetics. Accuracy, sensitivity, reproducibility, and robustness of Elispot resulted in a wide range of applications in research as well as in diagnostic field. Actually, CTL monitoring by Elispot is a gold standard for the evaluation of antigen-specific T cell immunity in clinical trials and vaccine candidates where the ability to detect rare antigen-specific T cells is of relevance for immune diagnostic. The most utilized Elispot assay is the interferon-gamma (IFN-γ) test, a marker for CD8(+) CTL activation, but Elispot can also be used to distinguish different subsets of activated T cells by using other cytokines such as T-helper (Th) 1-type cells (characterized by the production of IFN-γ, IL-2, IL-6, IL-12, IL-21, and TNF-α), Th2 (producing cytokines like IL-4, IL-5, IL-10, and IL-13), and Th17 (IL-17) cells. The reliability of Elispot-generated data, by the evaluation of T cell frequency recognizing individual antigen/peptide, is the core of this method currently applied widely to investigate specific immune responses in cancer, infections, allergies, and autoimmune diseases. The Elispot assay is competing with other methods measuring single-cell cytokine production, e.g., intracellular cytokine by FACS or Miltenyi cytokine secretion assay. Other types of lymphocyte frequency and function assays include limiting dilution assay (LDA), cytotoxic T cell assay (CTL), and tetramer staining. Compared with respect to sensitivity the Elispot assay is outranking other methods to define frequency of antigen-specific lymphocytes. The method described herein would like to offer helpful and clear protocols for researchers that apply Elispot. IFN-γ and perforin Elispot assays are described

    The Elephant In-Between

Within an area that presents edges with very different characteristics, surveilled and controlled to various extents, the proposal will focus on designing an intervention that will exercise control over the user through the qualities and through the use of the designed space. In addition, because the site presents the zone of the lagoon where an environmental regeneration was attempted, my proposal will make use of the new “nature” that has been designed.Considering these characteristics, as a function of the building I propose a sanatorium, a place fully controlled, where the human is contained, and removed from his usual environment. The graduation proposal project focuses mostly on understanding the extent of social control that a designed space can exercise on the user. In addition, it serves as a commentary on the current society that we live in, always watched and monitored, congested in chaotic cities that keep growing haphazardly to accommodate more and more inhabitantsArchitecture, Urbanism and Building Sciences | Borders and Territorie

Case study of “Magazinul Victoria”: Analysis of an interbellum monument, its decay and decline

    No full text
This study investigates the interwar development of a new typology of building- the department store, focusing on the case study of “Mgazinul Victoria”, first of its kind in the city of Bucharest, Romania. Rising to fame during the first half of the 20th century, surviving Second World War, and thriving during the Communist period (1947-1989), earning the title of protected monument from the Patrimony of Romania, today the building shows clear signs of economic decline and architectural decay. In this paper I explain and contextualize the important role that the store had in society, how the attitudes changed and how this change is reflected in the architecture of the store. Furthermore, by analyzing the architecture of the building I classify it within the Art Deco movement and argument that this classification together with its location within the city support the premise that the addition of the department store typology is representative for the public wish of alignment with the Western European countries, its cultures, formal urban structures, and architectural aesthetics. Additionally, the thesis illustrates the extensive decay and attempts to understand why is it present.AR2A011Architectural History ThesisArchitecture, Urbanism and Building Science

    Training the sages of integrity

    No full text
Drawing on his experience in the field and his familiarity with theatrical techniques, Marian Popescu offers us a stimulating reflection on what he ends up calling academic "integrative ethics" and on the role that "wise men of integrity" should play in its implementation. The author develops his proposal by understanding the historical flaws and advances in the field of character education. The training, combining cognitive devices and communicative and dramatic skills of these experts, mediators and referents appears, in his view, to be the key element in the current fight for integrity

THE SOCIAL COMPONENT OF THE COMMUNITY JUSTICE REFORM

The European Convention on Human Rights established an unprecedented system ofprotection of individual rights and freedoms applicable in the present, in almost all Europeancountries. The creation of a Permanent Court in 1988, was motivated essentially by the increasing the number ofindividual complaints the settlement of which was not possible under the old procedure. The number ofindividual complaints continued to increase on an accelerated rhythm and therefore the number ofcases in which settlement was required has increased considerably. The growing number of individual applications impose Community Justice System permanent reformand consideration of issues raised in the findings of this paper.community justice reform, European citizenship, European Court of Human Rights.

The Development of Touristic Services through Individual and Organizational Learning. Study Case: Romania and Spain

    No full text
The present research aims to establish and to design the development strategies for services in small and medium-sized enterprises (SMEs) that activate in the tourism field of activity (hotels and other accommodation establishments, restaurants, passenger transport, travel agencies, cultural tourism agencies). Organizational learning is focused on the learning process in the organization which has direct influence on the goods and services produced. Learning activities, carried out individually, are not easily transferred at the organizational level. Moreover, in order to better highlight the weaknesses and the strengths of touristic services management approaches and to define recommendations, our research theme is developed as comparative study: similarities and differences within SMEs touristic services and learning organization practices in Romania and other European Union’s country members (the example of Spain was considered).services, tourism, small and medium-sized enterprises (SMEs), organizational learning, human resources
