    Caspase-3 is not essential for DNA fragmentation in MCF-7 cells during apoptosis induced by the pyrrolo-1,5-benzoxazepine, PBOX-6

    Effector caspases-3, -6 and -7 are responsible for producing the morphological features associated with apoptosis, such as DNA fragmentation. The present study demonstrates that a member of a novel series of pyrrolo-1,5-benzoxazepines, PBOX-6, induces apoptosis in MCF-7 cells, which lack caspase-3. Apoptosis was accompanied by DNA fragmentation and the activation of caspase-7, but not caspases-3 and -6. Inhibition of caspase-7 activity reduced the extent of apoptosis induced, indicating that activation of caspase-7 is involved in the mechanism by which PBOX-6 induces apoptosis in MCF-7 cells. This study suggests that caspase-3 is not necessarily essential for DNA fragmentation and the morphological changes associated with apoptosis. © 2002 Federation of European Biochemical Societies.     Upon completion of transcription, the newly synthesized messenger RNA (mRNA) translocates out of the nucleus and into the cytoplasm where it gets translated into functional proteins. Decoding the mRNA into an amino acid sequence requires the concerted action of both RNA molecules, including transfer RNAs (tRNAs) and ribosomal RNAs (rRNAS), as well as many cytoplasmic proteins. This chapter outlines the molecular events that underlie the translation of mRNA into protein, highlighting the mechanisms that ensure high fidelity of these events. This chapter also discusses how newly synthesized proteins are targeted to their correct intracellular location, as well as the quality control mechanisms in place that ensure protein integrity inside the cell

    Regulation of Gene Expression: Basic Mechanisms

    The study of genes and the mechanism whereby the information they hold is converted into proteins and enzymes, hormones, and intracellular signaling molecules is the realm of molecular biology. Except for the erythrocyte, all cells in the body have the same DNA complement. One of the most fascinating aspects of this work is the study of the mechanisms that control differential gene expression, both in time and in place, and the consequences if these control mechanisms are disrupted.The goal of this chapter is to introduce the basic concepts involved in regulation of protein-coding genes and how these processes are involved in the causation of human disease. The basic mechanism of gene regulation will be described first, followed by a discussion of a specific gene regulation system to highlight various aspects of the basic mechanism. The chapter will end with a discussion of various ways in which the gene regulatory apparatus can be adapted to suit the needs of different tissues and situations.<br/

    Polyphony and the anxiety of influence in the fiction of Henry James

    James's fiction, especially in the Middle Phase, centres on the figure of the artist and is characterized by, the two interrelated aspects which previous criticism has largely overlooked: the Bakhtinian 'polyphonic' -creation of 'author-thinkers'; and the conflict between ephebes and precursors, for which Harold-Bloom's concept of 'the-anxiety of influence' is the most illuminating model. Polyphony is the narrative mode, and influence is the intra-artistic, theme. These, as the Introduction to the thesis makes clear, are rehearsed in James's inaugural novel, Roderick Hudson. Rowland Mallet is an author-thinker, and his failure is caused by authorial limitations. His monologism -is impaired by his mistaking empathy for the authorial sympathy. Likewise, Hudson's failure does not arise from a mercurial temperament, but from a polyphonic shortcoming: not possessing the power of fiction to contain the fiction of power in, his mentor. And the relationships among the three artists - Gloriani, Hudson and Singleton - perfectly exemplify the Bloomian-theme. It is these two concepts, polyphony and influence, which are the major preoccupation in the Middle Phase; as, the works chosen demonstrate. These are a novella, a novel, and a number of short stories all of which have been unjustifiably neglected. Chapter One, on The Aspern Papers, argues that Tina Bordereau, far from being, the artless victim seen by many critics, actually challenges and defeats the narrator by the very form of her narrative. Her 'realist' discourse undermines his language of 'romance', and shows up its internal unstability. Chapter Two is an extensive study of the critical reception of The Tragic Muse. The most common areas of critical attention have been its contemporary topicality, its relation to previous novels on similar themes, and the possible genealogy of Gabriel Nash. Those have all missed the core of the work. - Chapter Three demonstrates how polyphony and the anxiety of influence make the novel what it really is. Influence arises from the juxtaposition of, and the wrestling between, artistic ephebes and their precursors (Nick and Nash,, Miriam and Madame Carre). The dialogic quality defined by Bakhtin is crucial to the proper, and even-handed, characterization of all, the conflicts in the novel. And since most of James's tales in the eighties and nineties -are about 'masters - and acolytes, the anxiety of influence remains central. Chapter Four is a study of 'The Author of Beltraffiol' and 'The Lesson of the Master'. Again the characters' manipulations are a crucial focus in a way that G6rard Genette's terminology helps to illuminate. The fact that the ephebe is the author-thinker emphasizes the inextricability of the Bakhtinian and the Bloomian in James. Just as polyphony offers a different focus for explicating the poetics of James's fiction; so the ephebal conflict provides the basis for a fresh perception of James's own artistic struggle

    From crops to clinic: the impact of dual azole use on antifungal resistance in Candida and Candida associated yeasts

    Invasive fungal infections caused by pathogenic yeasts are an escalating global health crisis that demands urgent attention within a One Health framework. This review critically examines mounting evidence that widespread agricultural azole fungicide use is a key driver accelerating antifungal resistance in pathogenic yeast. We dissect the shared molecular targets and resistance pathways that underpin dangerous cross-resistance between environmental fungicides and clinical azoles. Traditionally viewed as human commensals, we provide a comprehensive account of the evidenced environmental reservoirs of yeast pathogens, including agricultural soils, wastewater, and the food chain. Ecosystems burdened by persistent azole contamination that create hotspots for resistance evolution and amplification. With antifungal treatment options rapidly diminishing and resistant infections causing rising morbidity and mortality worldwide, we identify vulnerabilities in our shared environment and consider integrated surveillance, stewardship, and environmental interventions to help preserve the efficacy of life-saving antifungals and mitigate the growing threat of fungal disease.<br/

    Yeast Epigenetics: The Inheritance of Histone Modification States

    Saccharomyces cerevisiae (budding yeast) and Schizosaccharomyces pombe (fission yeast) are two of the most recognized and well-studied model systems for epigenetic regulation and the inheritance of chromatin states. Their silent loci serve as a proxy for heterochromatic chromatin in higher eukaryotes, and as such both species have provided a wealth of information on the mechanisms behind the establishment and maintenance of epigenetic states, not only in yeast, but in higher eukaryotes. This review focuses specifically on the role of histone modifications in governing telomeric silencing in S. cerevisiae and centromeric silencing in S. pombe as examples of genetic loci that exemplify epigenetic inheritance. We discuss the recent advancements that for the first time provide a mechanistic understanding of how heterochromatin, dictated by histone modifications specifically, is preserved during S-phase. We also discuss the current state of our understanding of yeast nucleosome dynamics during DNA replication, an essential component in delineating the contribution of histone modifications to epigenetic inheritance.</p
