Progress and outlook in studying the substrate specificities of PARPs and related enzymes
Despite decades of research on ADP-ribosyltransferases (ARTs) from the poly(ADP-ribose) polymerase (PARP) family, one key aspect of these enzymes - their substrate specificity - has remained unclear. Here, we briefly discuss the history of this area and, more extensively, the recent breakthroughs, including the identification of protein serine residues as a major substrate of PARP1 and PARP2 in human cells and of cysteine and tyrosine as potential targets of specific PARPs. On the molecular level, the modification of serine residues requires a composite active site formed by PARP1 or PARP2 together with a specificity-determining factor, HPF1; this represents a new paradigm not only for PARPs but generally for post-translational modification (PTM) catalysis. Additionally, we discuss the identification of DNA as a substrate of PARP1, PARP2 and PARP3, and some bacterial ARTs and the discovery of noncanonical RNA capping by several PARP family members. Together, these recent findings shed new light on PARP-mediated catalysis and caution to 'expect the unexpected' when it comes to further potential substrates

Nutritive value of winter wheat for broiler chickens
The nutritive value of Australian winter wheat varieties for broiler chickens was assessed in a series of conventional energy balance studies each of 7-days duration. Across all samples, the mean and standard deviation for apparent metabolisable energy (AME, MJ/kg dry matter) were 14.29±0.36 (n=25) and ranged from 13.68 to 15.02. For individual varieties, means and standard deviations were 14.80±0.03 for Declic (n=2), 14.31±0.33 for Lawson (n=16), 13.97±0.34 for More (n=4) and 14.20±0.09 for Paterson (n=3). In conclusion, winter wheats were consistently high in AME but, nevertheless, were responsive to endo-4,4-xylanase added to the diet. Uplift in AME of wheat due to enzyme supplementation averaged 0.7 MJ/kg (4.9%) and ranged from 0.22 MJ/kg (1.5%) to 1.13 MJ/kg (8.2%).R.J. Hughes, L. Acone, P. Zviedrans and M. Choc

Goal-orientated Functional Rehabilitation using Electrical Stimulation and Iterative Learning Control for Motor Recovery in the Upper Extremity Post-Stroke.
There is increasing evidence that electrical stimulation (ES) combined with task specific training is effective in the recovery of upper extremity dysfunction following stroke. The aim of this study is to develop a rehabilitation system that delivers precisely controlled levels of stimulation to the shoulder, elbow and wrist during goal-oriented activity which utilises everyday real objects. Iterative learning control (ILC) is used to mediate the ES and updates the stimulation signal applied to each muscle group based on the error between the ideal and actual movement in the previous attempt. The control system applies the minimum amount of stimulation required, maximising voluntary effort with a view to facilitating success at each given task. Markerless motion tracking is provided via a Microsoft Kinect, with hand and wrist data measured by an electrogoniometer. Preliminary results show that ES mediated by ILC has successfully facilitated movement across the shoulder, elbow and wrist of chronic stroke patients. Overall, joint error has reduced for all participants with the mean error across all joints showing reductions for all participants.
