    Genome-wide association analysis of agronomical and physiological traits linked to drought tolerance in a diverse potatoes (Solanum tuberosum) panel

    Potatoes is often considered a drought-sensitive crop and its sustainable production is threatened due to frequent drought episodes. Drought tolerance is a complex trait of increasing importance in potatoes therefore its yield is threatened. The differential response of several physiological and agronomical traits was evaluated in a diverse potatoes panel under contrasting water regimes to identify quantitative trait loci (QTL) associated with drought tolerance using genome-wide association analysis with 4859 high-quality SNP markers. Phenotypic data were collected from multiple environments and years. QTL with diverse linkage disequilibrium blocks for proline concentration, water consumption and yield were detected on chromosomes 1, 4 and 10. These QTL detected were associated with known gene functions. The current study provides insights into the putative genes that controlled the responses to the drought tolerance in potatoes at physiological and agronomical levels.     Use of chemicals to maintain quality potato minimally processed products

    La demanda por el agregado de valor a los productos de origen agropecuario ha generado interés sobre la preservación de productos mínimamente procesados (PMP). La vida útil de los PMP es disminuida por la reacción de pardeamiento enzimático, la cual causa una pérdida en la calidad del alimento. En el presente trabajo se estudió el efecto de la adición de compuestos químicos sobre el contenido de ácido ascórbico, fenoles totales, la actividad de la enzima PPO y el color de los PMP de papa (Solanum tuberosum L.). Los tratamientos fueron combinaciones de diferentes concentraciones de metabisulfito de sodio (MS), ácido ascórbico (AA) y ácido cítrico (AC). El ensayo se realizó en dos variedades de papa, Spunta y Newen INTA. Los tratamientos con 1,5% de AA y 1% AC mantuvieron la luminosidad y aumentaron la concentración de ácido ascórbico y el contenido de fenoles totales en el producto durante los primeros 10 días de almacenamiento. Es posible disminuir la concentración de MS si se lo combina con AA para mantener los parámetros de calidad en papas cortadas refrigeradas.Demand for value addition to the products of agricultural origin has generated interest in the preservation of minimally processed products (PMP). The shelf life of the PMP is decreased by the enzymatic browning reaction, which causes a loss in the quality of food. In this study the effect of chemical addition on the content of ascorbic acid, total phenolics, the PPO enzyme activity and colour of PMP potato (Solanum tuberosum L.) was studied. Treatments were combinations of different concentrations of sodium metabisulfite (MS), ascorbic acid (AA) and citric acid (CA). The assay was performed in two potato varieties, Newen INTA and Spunta. Treatments with 1.5% AA and 1% AC kept the brightness and increased the concentration of ascorbic acid and total phenolic content in the product during the first 10 days of storage.     Controlar efectivamente a Phytophthora infestans, deviene de seleccionar genotipos con capacidad de transmitir resistencia. Con el objeto de caracterizar progenitores de papa en base a aptitud combinatoria y heterosis para resistencia a P. infestans, se cruzaron seis variedades (Libertas, Jaspe, Chotañawi, Pollerita, Robusta e INRA 92T.114.76). Quince familias de tubérculos de segunda generación clonal, obtenidas, fueron inoculadas con P. infestans en Balcarce (Buenos Aires, Argentina) y evaluadas bajo diseño en bloques completos aleatorizados con dos repeticiones. Se midió área bajo la curva del progreso de la enfermedad (AUDPC). Se estimó Aptitud Combinatoria (AC) general y específica, heterosis media, heterobeltiosis, heterosis específica (Hs) y heredabilidad en sentido amplio (H2) y estricto (h2). La AC fue significativa. El progenitor Robusta disminuyó la enfermedad estimada por AUDPC, el valor negativo grande de AC específica señaló mejor híbrido a Robusta x Chotañawi. Un tercio de las cruzas expresaron aditividad en todos los niveles de heterosis. Los valores de Hs manifestaron equivalencia con el porcentaje de disminución del AUDPC. Las heredabilidades (H2=0,63 y h2=0,54) indicaron que la selección por bajo AUDPC puede ser efectiva. El progenitor Robusta y la cruza Robusta x Chotañawi, conformaron los genotipos superiores recomendados para transferir resistencia a P. infestans.The efficient control of Phytopthora infestans results from the selection of genotypes with the capacity of transferring resistance. One third of the crosses expressed significant additive effects for all levels of heterosis. Hs values showed equivalence with the percent reduction of AUDPC.     Mapeo asociativo y análisis de desequilibrio de ligamiento para la identificación de genes de tolerancia a estrés abiótico en papa tetraploide

    La papa (Solanun tuberosum spp. tuberosum, 2n = 4x = 48) es el cuarto cultivo alimenticio más importante del mundo. A nivel mundial, el cultivo es sembrado en 19 millones de hectáreas y su producción anual es de alrededor de 325 x 106 toneladas. En la Argentina, se siembran aproximadamente 75.500 mil hectáreas anuales, de las cuales 32 mil hectáreas se encuentran en el Sudeste de la provincia de Buenos Aires. La papa es un cultivo descripto como susceptible a la sequía, limitando las áreas de producción a zonas húmedas o bien irrigadas. Entre las especies presentes en el cultivo de papa existe gran variabilidad genotípica respecto a la tolerancia al estrés hídrico. El mapeo asociativo aprovecha la diversidad existente en el germoplasma estudiado para identificar alelos y loci vinculados a un rasgo de interés como la tolerancia a estrés hídrico por sequía sin la necesidad de construir poblaciones de mapeo. En este trabajo se evaluó la estructura poblacional y la diversidad genética en un panel de mapeo asociativo vinculado al germoplasma del Programa de Mejoramiento Genético de Papa de la EEA INTA-Balcarce que incluyó genotipos pertenecientes a Solanun tuberosum spp. tuberosum, S. tuberosum spp. andígena, S. tarijense y S. chacoense mediante dos aproximaciones: utilizando marcadores moleculares (SNPs) y por medio de marcadores fenotípicos de fácil medición. En complementación, se calcularon predictores lineales insesgados (BLUPs) para rasgos fenotípicos asociados a la tolerancia al estrés por sequía en ambientes con regímenes hídricos contrastantes. Finalmente, a partir de los valores BLUPs se realizó un análisis por mapeo asociativo para evaluar la asociación de éstos con la variación genotípica evidenciada por el genotipado de la población mediante marcadores tipo SNPs. Se utilizó como cofactores fijos la estructura poblacional y la matriz de parentesco. La estructura de la población estudiada mediante el uso por marcadores tipo SNPs se asoció con el origen del germoplasma evaluado. Los rasgos fenotípicos de fácil medición mostraron capacidad preliminar para agrupar poblaciones de papa de diverso origen, grupo taxonómico y nivel de ploidía. En general las variables fenotípicas evaluadas en ambientes con regímenes hídricos contrastantes mostraron respuesta diferencial frente al estrés. Los BLUPs permitieron generar un ordenamiento de genotipos con buena respuesta frente al estrés hídrico por sequía. Mediante el análisis molecular se observó desequilibrio de ligamiento en cada uno de los cromosomas de la papa. Posteriormente, se encontraron asociaciones significativas entre los marcadores moleculares y los fenotípicos asociados al rendimiento, concentración de prolina y consumo de agua. Los marcadores asociados a la concentración de prolina y consumo de agua se vincularon con genes candidatos de respuesta a estrés. Los análisis conducidos en la presente tesis permitieron (i) evidenciar la variabilidad genética en la población de papa utilizada, (ii) validar la capacidad selectiva de los predictores insesgados tipo BLUPs de los rasgos fenotípicos evaluados para la tolerancia a estrés hídrico por sequía y por último (iii) proponer posibles QTLs asociados a la tolerancia a estrés por sequía.Fil: Tagliotti, Martin Enrique.     Temporal and spatial dynamics of Tomato spotted wilt virus and its vector in a potato crop in Argentina

    The nature of spatial and temporal dynamics of Tomato spotted wilt virus (TSWV) and its vector in a potato crop cv. Innovator without insecticide application is analysed. Seed tuber was analysed for the presence of TSWV as a source of initial inoculum. The presence of plants with symptoms of TSWV was evaluated by visual observation and DAS-ELISA analysis to confirm the virus infection. Thrips species were collected from leaves and inflorescences and identified under stereomicroscope. The distribution of symptomatic plants and thrips species was recorded five times at 14 days intervals. The initial seed tuber infection was of 1.1%. Disease incidence was 0% at 29 days after planting (DAP), 0.2% at 43 DAP, 2.2% at 56 DAP, 11.6% at 70 DAP and 14.6% at 84 DAP. The progress of the disease was adequately described by a Logistic model [y=0.15/(1+1205372.93 × exp (−0.22 × DAP))]. Thrips vector species identified as resident in the crop during the whole cycle were Thrips tabaci (n=423), Frankliniella occidentalis (n=141) and as occasional species, F. schultzei (n=34) and F. gemina (n=5) were found. At 43 and 56 DAP a random distribution pattern was observed and the thrips species found were T. tabaci (n=188) and F. occidentalis (n=105). An aggregated pattern was determined at 70 and 84 DAP. Spatial patterns of the disease spread suggest a polycyclic epidemic with TSWV secondary spread in the potato crop. Multiple control measures were deduced from these epidemiological results like virus testing in tubers, removal of external virus infection sources and thrips control.EEA BalcarceFil: Salvalaggio, Andrea Eugenia.     Ascorbic Acid and Citric Acid Treatments Increase the Shelf Life of Fresh-Cut Potato: Cultivar Effect

    Potato cultivars were processed as minimally processed products (MPP) and treated with different concentrations of ascorbic (AA) and citric acids (CA) in order to investigate the effect of these treatments on the overall quality of MPP from ‘Innovator’, ‘Newen INTA’ and ‘Spunta’ cultivars. Colour changes, PPO activity and bioactive compounds of fresh-cut potato differed amongst potato cultivars and treatments. ‘Innovator’ had the least colour changes and low enzymatic activity. ‘Newen INTA’ highly susceptible to browning was the one that presented the highest values of polyphenol content and antioxidant capacity. Treatments applied kept microbiological counts and global appearance during 10 days of storage. Considering the different susceptibility of cultivars to enzymatic browning, the results obtained provide crucial information to select the best treatment to keep the overall quality of MPP of these important cultivars of Southeastern Buenos Aires, Argentina, as a useful tool for the industry.Fil: Garcia Procaccini, Luz Milagros.     Identificación de factores genéticos asociados a la resistencia a Tizón tardío (Phytophthora infestans Mont. de Bary) en papa tetraploide, mediante el uso de mapeo asociativo con SNPs

    Tesis para obtener el grado de Doctor en Ciencias Agrarias, área Mejoramiento Genético, de la Universidad Nacional de Mar del Plata, en 2019.La papa (Solanum tuberosum ssp. tuberosum L.) es el cuarto alimento más consumido en el mundo, luego del maíz, el trigo y el arroz. El tizón tardío de la papa, enfermedad causada por el oomycete Phytophthora infestans (Mont.) de Bary, se encuentra en todas las zonas de producción de papa de Argentina. Los objetivos del mejoramiento genético de este cultivo, han sido la incorporación de resistencias a factores bióticos y abióticos adversos, el desarrollo de cualidades para el procesamiento industrial y su combinación con alto rendimiento y calidad comercial. En este sentido, el mapeo asociativo utiliza la diversidad existente en el germoplasma estudiado para identificar alelos y loci responsables de la variación observada en la manifestación de un carácter fenotípico sin la necesidad de construir poblaciones de mapeo biparentales. A pesar de que la base genética de la papa cultivada es considerada estrecha, posee un alto número de especies emparentadas (de diferente ploidía y origen) con gran variabilidad para numerosos caracteres de interés agronómico. Se analizó la diversidad genética y la estructura de una población formada por 231 genotipos de la colección de germoplasma del Programa de Mejoramiento genético de papa de la EEA INTA Balcarce compuesta por variedades de S. tuberosum ssp. tuberosum de distintos orígenes, genotipos de S. tuberosum ssp. andigena y especies silvestres de papa mediante la utilización de marcadores moleculares (SNPs). El análisis de estructura dividió a la población en grupos bien definidos relacionados con origen, ploidía, taxonomía y programa de mejoramiento de cada genotipo. Este resultado fue confirmado por un dendrograma realizado a partir de la distancia genética de Nei para la población completa. Los índices de diversidad confirmaron la presencia de variabilidad genética en la población. El análisis de pedigrí dentro de las subpoblaciones de S. tuberosum ssp. tuberosum identificó los genotipos parentales más frecuentes y aquellos que realizaron la mayor contribución a cada subpoblación. Katahdin estuvo presente como el parental más frecuente en las líneas materna y paterna de todas las subpoblaciones analizadas de S. tuberosum ssp. tuberosum, pero sus contribuciones a la diferenciación genética fueron bajas. Por otro lado, se determinó el comportamiento a campo de los genotipos frente a P. infestans y se estimó el rendimiento luego de someter las plantas a la infección con el patógeno. Las condiciones meteorológicas permitieron que la enfermedad estuviera presente en todos los años de ambas localidades. Se encontró variabilidad para los tres parámetros fenotípicos evaluados, lo cual se vio reflejado en los valores de las medias ajustadas para cada genotipo en cada localidad. El análisis de componentes principales dividió a los genotipos con valores altos de RAUDPC de aquellos con valores bajos para la variable, y de igual forma lo realizó para la variable PESO TOTAL, en ambas localidades. De esta manera, se pudo obtener un grupo de genotipos con buen comportamiento para ambas variables. Los valores de heredabilidad obtenidos para la variable RAUDPC son medios a altos y coinciden con aquellos reportados en la bibliografía. Sin embargo, para la variable PESO TOTAL resultaron más bajos que los reportados. Finalmente, dicha información se utilizó para realizar mapeo por asociación. El mismo se efectuó sobre 187 genotipos del panel analizado, los cuales contaron con información fenotípica y caracterización molecular con SNPs. Se encontraron ocho asociaciones significativas utilizando la metodología de Bonferroni, con valores de -log (p) que oscilan entre 4,61 y 5,42 y nueve asociaciones significativas empleando Permutaciones, con valores de - log(p) entre 4,5 y 6,42. Algunos de los SNPs detectados, se encuentran adyacentes a genes que codifican enzimas participantes de vías metabólicas que se activan en respuesta a la interacción con patógenos.Cultivated potato (Solanun tuberosum ssp. tuberosum L.) is the fourth most consumed food in the world, after corn, wheat and rice. Potato late blight is a disease caused by Phytophthora infestans (Mont.) de Bary. It is found in all areas of potato production in Argentina. Potato breeding objectives have been the incorporation of resistance or tolerance to adverse biotic and abiotic factors, the development of better qualities for the industrial processing and and combine these characters with high yield and commercial quality. According to this, association mapping uses the existing diversity in the studied germplasmto identify alleles and loci responsible for the variation observed of a phenotypic character without the need to build biparental mapping populations. Although the genetic base of the cultivated potato is considered narrow, it has a high number of related species (of different ploidy and origin) with great variability for many characters of agronomic interest. The genetic diversity and the genetic structure of a population formed by 231 genotypes of the germplasm collection of the EEA INTA Balcarce Potato Breeding Program were analyzed using molecular markers (SNPs). The population was composed by genotypes of S. tuberosum ssp. tuberosum of different origins, genotypes of S. tuberosum ssp. andigena and wild potato species. The structure analysis divided the population into well-defined groups related to their origin, ploidy, taxonomy and breeding program. This result was confirmed by a dendrogram made from the Nei genetic distance for the whole population. The diversity indices confirmed the presence of genetic variability in the population. Pedigree analysis within subpopulations of S. tuberosum ssp. tuberosum identified the most frequent parental genotypes and those that made the greatest contribution to each subpopulation. Katahdin was the most frequent parental in the maternal and paternal path of all the subpopulations analyzed of S. tuberosum ssp. tuberosum, but their contributions to genetic differentiation were very low. On the other hand, an assessment of the genotype’s behavior and yield under infection with P. infestans, was made under field conditions. The environmental conditions allowed the presence of the disease in all the years of both localities. Phenotipic variability was found for the three parameters evaluated, which was reflected in the values of the adjusted means for each genotype in each locality. Principal components analysis divided the genotypes with high values of RAUDPC from those with low values for the variable, and in the same way it was done for the variable TOTAL WEIGHT, in both locations. In this way, it was possible to identify a group of genotypes with desirable values for both variables. The heritability values obtained for the variable RAUDPC are medium to high and coincide with those reported in the bibliography. However, the heritability for TOTAL WEIGHT was lower than that reported. Finally, this information was used to perform mapping by association. The same was carried out on 187 genotypes of the panel analyzed, which had phenotypic information and molecular characterization with SNPs. Eight significant associations were found using the Bonferroni methodology, with values of -log (p) ranging between 4.61 and 5.42 and eleven significant associations using Permutations, with values of -log (p) between 4.5 and 6.42.     Discriminant analysis of principal components and pedigree assessment of genetic diversity and population structure in a tetraploid potato panel using SNPs

    The reported narrow genetic base of cultivated potato (Solanum tuberosum) can be expanded by the introgression of many related species with large genetic diversity. The analysis of the genetic structure of a potato population is important to broaden the genetic base of breeding programs by the identification of different genetic pools. A panel composed by 231 diverse genotypes was characterized using single nucleotide polymorphism (SNP) markers of the Illumina Infinium Potato SNP Array V2 to identify population structure and assess genetic diversity using discriminant analysis of principal components (DAPC) and pedigree analysis. Results revealed the presence of five clusters within the populations dif ferentiated principally by ploidy, taxonomy, origin and breeding program. The information obtained in this work could be readily used as a guide for parental introduction in new breed ing programs that want to maximize variability by combination of contrasting variability sources such as those presented here.EEA BalcarceFil: Deperi, Sofía Irene.     Solanum tarijense reaction to P. infestans and the role of plant defense molecules

    Late blight is the main disease of potatoes around the world. Because of the difficulties in applying effective control methods, the introduction of resistant cultivars represents a safe strategy. The potato species Solanum tarijense represents an attractive resistance source as its adaptation to long days is promissory, producing tubers of good size and aspect. Deposition of structural compounds like lignin and callose were described as a non‐specific resistance mechanism. In this work, we measured polyphenoloxidases (PPO) and peroxidases (POX) activities and the accumulation of phenols, lignin and callose and their correlation with the resistance levels of S. tarijense. Clones Oka 6320.9 and Oka 5632.11 showed low infection rates and these were correlated with a higher accumulation of phenols, lignin and callose and a strong induction of PPO and POX activities. However, in highly infected clones, a lower or no accumulation of these compounds was observed. These results demonstrate a correlation between the amount of defence molecules and the resistance levels according to the detached‐leaf assay.
