Effect of Methionine Hydroxy Analog on Hu Sheep Digestibility, Rumen Fermentation, and Rumen Microbial Community In Vitro
This experiment was conducted to evaluate the effects of a methionine hydroxy analog (MHA) on in vitro gas production, rumen fermentation parameters, and rumen microbiota. Two different MHA, 2-hydroxy-4-(methylthio) butanoic acid isopropyl ester (HMBi) and the calcium salt of the hydroxy analog of methionine (MHA-Ca), were selected for in vitro experiments. The treatments were the Control group (0% of MHA), HMBi group (2%HMBi), and MHA-Ca group (2%MHA-Ca). Dry matter digestibility was measured after 12 h and 24 h of fermentation, and fermentation parameters and microbial composition were analyzed after 24 h. HMBi and MHA-Ca showed increased (p = 0.001) cumulative gas production in 3 h. The total volatile fatty acids, microbial protein (MCP) concentration, acetate, and acetate to propionate ratio in the HMBi and MHA-Ca groups were significantly higher than those in the Control group (p = 0.006, p = 0.002, p = 0.001, p = 0.004), and the NH3-N concentrations in the HMBi and MHA-Ca groups were significantly lower than those in the Control group (p = 0.004). The 16S rRNA sequencing revealed that the HMBi group had a higher (p = 0.039, p = 0.001, p = 0.027) relative abundance of Bacteroidetes, Firmicutes, and Synergistetes and a lower relative abundance of Proteobacteria (p = 0.001) than the Control group. At the genus level, Prevotella abundance was higher (p = 0.001), while Ruminobacter abundance was lower (p = 0.001), in the HMBi and MHA-Ca groups than in the Control group. Spearman’s correlation analysis showed that the relative abundance of Prevotella_1, Streptococcus, and Desulfovibrio was positively correlated with dry matter digestibility, MCP, and fermentation parameters. MHA, thus, significantly increased gas production and altered the rumen fermentation parameters and microbiota composition of sheep
Two-way partial AUC and its properties
Simultaneous control on true positive rate and false positive rate is of significant importance in the performance evaluation of diagnostic tests. Most of the established literature utilizes partial area under receiver operating characteristic (ROC) curve with restrictions only on false positive rate (FPR), called FPR pAUC, as a performance measure. However, its indirect control on true positive rate (TPR) is conceptually and practically misleading. In this paper, a novel and intuitive performance measure, named as two-way pAUC, is proposed, which directly quantifies partial area under ROC curve with explicit restrictions on both TPR and FPR. To estimate two-way pAUC, we devise a nonparametric estimator. Based on the estimator, a bootstrap-assisted testing method for two-way pAUC comparison is established. Moreover, to evaluate possible covariate effects on two-way pAUC, a regression analysis framework is constructed. Asymptotic normalities of the methods are provided. Advantages of the proposed methods are illustrated by simulation and Wisconsin Breast Cancer Data. We encode the methods as a publicly available R package tpAUC. </jats:p
Modeling Population Dynamics and Assessing Ecological Impacts of Lampreys via Sex Ratio Regulation
Regulating lamprey populations is crucial for maintaining ecological equilibrium. However, the unique sex determination process of lampreys is constrained by multiple factors, complicating intuitive analysis of population dynamics and their impact on the natural environment. This study employed a two-species competition mechanism to elucidate the factors influencing sex ratios and their mechanistic effects on lamprey population size. Using the Lotka–Volterra equations, we investigated how sex ratios affect trophic levels both upstream and downstream of lampreys in the food web. A logistic population growth model was applied to assess the impact of sex ratio variations on symbiotic parasitic species, while the Analytic Hierarchy Process (AHP) was utilized to explore the dynamic relationship between sex ratio changes and ecosystem stability. To validate model efficacy, we manipulated temperature and food availability under controlled disturbance conditions, analyzing temporal variations in lamprey population size across different disturbance intensities to evaluate model sensitivity. The findings indicate that the variable sex ratio’s benefit is in facilitating the lampreys’ population’s enhanced adaptation to environmental shifts. The coexisting species exhibit a similar pattern of population alteration as the lampreys, albeit with a minor delay. A definitive link between the quantity of lampreys and the parasitic species is absent. A male ratio of 0.6 optimally contributes to the ecosystem’s equilibrium. Over time, the configuration of our model’s parameters proves to be sensible. This research provides robust theoretical support for developing scientific strategies to regulate lamprey populations
Human epidermal growth factor receptor 2 positive (HER2+) metastatic breast cancer: how the latest results are improving therapeutic options
Human epidermal growth factor receptor 2 positive (HER2+) metastatic breast cancer (MBC) remains an incurable disease, and approximately 25% of patients with HER2+ early breast cancer still relapse after adjuvant trastuzumab-based treatment. HER2 is a validated therapeutic target that remains relevant throughout the disease process. Recently, a number of novel HER2 targeted agents have become available, including lapatinib (a small molecule tyrosine kinase inhibitor of both HER2 and the epidermal growth factor receptor), pertuzumab (a new anti-HER2 monoclonal antibody) and ado-trastuzumab emtansine (T-DM1, a novel antibody-drug conjugate), which provide additional treatment options for patients with HER2+ MBC. The latest clinical trials have demonstrated improved outcome with treatment including pertuzumab or T-DM1 compared with standard HER2 targeted therapy. Here we review the clinical development of approved and investigational targeted agents for the treatment of HER2+ MBC, summarize the latest results of important clinical trials supporting use of these agents in the treatment of HER2+ MBC, and discuss how these results impact therapeutic options in clinical practice.SCI(E)[email protected]
Selective biosorption of thorium (IV) from aqueous solutions by ginkgo leaf.
Low-cost biosorbents (ginkgo leaf, osmanthus leaf, banyan leaf, magnolia leaf, holly leaf, walnut shell, and grapefruit peel) were evaluated in the simultaneous removal of La3+, Ce3+, Pr3+, Nd3+, Sm3+, Eu3+, Gd3+, Yb3+, Lu3+, UO22+, Th4+, Y3+, Co2+, Zn2+, Ni2+, and Sr2+ from aqueous solutions. In single metal systems, all adsorbents exhibited good to excellent adsorption capacities toward lanthanides and actinides. In a simulated multicomponent mixed solution study, higher selectivity and efficiency were observed for Th4+ over other metal cations, with ginkgo leaves providing the highest adsorptivity (81.2%) among the seven biosorbents. Through optimization studies, the selectivity of Th4+ biosorption on ginkgo leaf was found to be highly pH-dependent, with optimum Th4+ removal observed at pH 4. Th4+ adsorption was found to proceed rapidly with an equilibrium time of 120 min and conform to pseudo-second-order kinetics. The Langmuir isotherm model best described Th4+ biosorption, with a maximum monolayer adsorption capacity of 103.8 mg g-1. Thermodynamic calculations indicated that Th4+ biosorption was spontaneous and endothermic. Furthermore, the physical and chemical properties of the adsorbent were determined by scanning electron microscopy, Brunauer-Emmett-Teller, X-ray powder diffraction, and Fourier transform infrared analysis. The biosorption of Th from a real sample (monazite mineral) was studied and an efficiency of 90.4% was achieved from nitric acid at pH 4 using ginkgo leaves
