    Differential Phosphorylation of Smad1 Integrates BMP and Neurotrophin Pathways through Erk/Dusp in Axon Development

    SummarySensory axon development requires concerted actions of growth factors for the precise control of axonal outgrowth and target innervation. How developing sensory neurons integrate different cues is poorly understood. We demonstrate here that Smad1 activation is required for neurotrophin-mediated sensory axon growth in vitro and in vivo. Through differential phosphorylation, Smad1 exerts transcriptional selectivity to regulate the expression and activity of Erk1 and Erk2—two key neurotrophin effectors. Specifically, bone morphogenetic proteins (BMPs) signal through carboxy-terminal phosphorylation of Smad1 (pSmad1C) to induce Erk1/2 transcription for enhanced neurotrophin responsiveness. Meanwhile, neurotrophin signaling results in linker phosphorylation of Smad1 (pSmad1L), which in turn upregulates an Erk-specific dual-specificity phosphatase, Dusp6, leading to reduced pErk1/2 and constituting a negative-feedback loop for the prevention of axon overgrowth. Together, the BMP and neurotrophin pathways form a tightly regulated signaling network with a balanced ratio of Erk1/2 and pErk1/2 to direct the precise connections between sensory neurons and peripheral targets

    Objective To learn current status of health management services in the elderly individuals living in rural areas of Beijing,so as to provide evidence for healthcare improvement.Methods The elderly residents aged 60 or above from several rural districts of Beijing were selected by using multistage random sampling method in January 2013.The health management programme those participants obtained and their satisfactory scale were evaluated.Results A total of 719 of 1 023 (71.3%) individuals had health check-up over the past years.Only 4 subjects (0.4%) obtained mental health management,and 35 (4.9%) finished follow-up studies for chronic diseases.The percent of the elderly who received physical examination,profiles of medical record,health education,medical consultation,health evaluation,cancer screening or vaccination showed statistically significant difference of regional distribution (x2 values were 15.7,39.4,30.5,25.2,24.1,18.4 and 19.3,respectively; all P<0.05).Sites for health check-up,profiles of medical record and health education were also found to have statistically significant regional distribution difference (all P<0.05).In ordinal logistic regression analysis,age,occupation,health insurance,self health assessment and health medical affected satisfaction of the respondents with health management.Conclusions Our investigation shows a gap between current health management programme and national standard requirement for equal public health services,and the elderly's satisfaction with health management needs to be improved

    Epigenetic regulation of sensory axon regeneration after spinal cord injury

    Axon regeneration is hindered by a decline of intrinsic axon growth capability in mature neurons. Reversing this decline is associated with the induction of a large repertoire of regeneration-associated genes (RAGs), but the underlying regulatory mechanisms of the transcriptional changes are largely unknown. Here, we establish a correlation between diminished axon growth potential and histone 4 (H4) hypoacetylation. When neurons are triggered into a growth state, as in the conditioning lesion paradigm, H4 acetylation is restored, and RAG transcription is initiated. We have identified a set of target genes of Smad1, a proregenerative transcription factor, in conditioned DRG neurons. We also show that, during the epigenetic reprogramming process, histone-modifying enzymes work together with Smad1 to facilitate transcriptional regulation of RAGs. Importantly, targeted pharmacological modulation of the activity of histone-modifying enzymes, such as histone deacetylases, leads to induction of multiple RAGs and promotion of sensory axon regeneration in a mouse model of spinal cord injury. Our findings suggest epigenetic modulation as a potential therapeutic strategy to enhance axon regeneration. </p

    Skin-Stiffener Separation in T-Stiffened Composite Specimens in Postbuckling Condition

    An experimental and numerical investigation was conducted to study the skin-stiffener separation of single T-shape stiffener specimens in postbuckling condition. Three specimens were manufactured with a centrally located Teflon insert, and were loaded in compression until collapse. Deformation patterns and separation evolution were monitored during the tests. To measure the full-field displacements and the strain distributions of the specimens, a digital image correlation (DIC) system was used. Skin-stiffener separation was observed and measured with an ultrasound system. Finite-element (FE) analyses were conducted to capture interlaminar damage mechanism based on the virtual crack closure technique. The numerical analysis accurately predicted the postbuckling deformation and the skin-stiffener separation behavior. The close correlation between the experimental and numerical results allows for further exploitation of the strength reserve in the postbuckling region and wider design options for the next generation of composite aircraft designs.</p

    A general multi-relational classification approach using feature generation and selection

    Multi-relational classification is an important data mining task, since much real world data is organized in multiple relations. The major challenges come from, firstly, the large high dimensional search spaces due to many attributes in multiple relations and, secondly, the high computational cost in feature selection and classifier construction due to the high complexity in the structure of multiple relations. The existing approaches mainly use the inductive logic programming (ILP) techniques to derive hypotheses or extract features for classification. However, those methods often are slow and sometimes cannot provide enough information to build effective classifiers. In this paper, we develop a general approach for accurate and fast multi-relational classification using feature generation and selection. Moreover, we propose a novel similarity-based feature selection method for multi-relational classification. An extensive performance study on several benchmark data sets indicates that our approach is accurate, fast and highly scalable. ? 2010 Springer-Verlag.EI

    Finite Element Methods and Their Convergence for Elliptic and Parabolic Interface Problems

    In this paper, we consider the finite element methods for solving second order elliptic and parabolic interface problems in two-dimensional convex polygonal domains. Nearly the same optimal L 2 -norm and energy-norm error estimates as for regular problems are obtained when the interfaces are of arbitrary shape but are smooth, though the regularities of the solutions are low on the whole domain.
