    Activation of HER2 Signaling Pathway Modulates AR Functions and Prostate Cancer Progression

    前列腺癌對於雄性素去除治療法產生耐受性使其在治療上十分棘手,其中,雄性素受體持續活化可促進前列腺癌惡化及擺脫雄性素去除所造成的治療效果,正常狀態下雄性素受體受到雄性素作用調控,但是在雄性素去除治療法失效的情況下,前列腺癌細胞產生許多改變造成雄性素受體可藉由其他方式活化,在目前的研究中HER2被認為是其中一個的可能因子。HER2對於乳癌的惡化以及對tamoxifen治療產生耐受性上扮演重要的角色,然而,在前列腺癌惡化過程中HER2所扮演的角色仍然並不清楚。在此研究中,我們利用帶有PSA-reporter的22Rv1前列腺癌細胞株分析HER2促成前列腺癌惡化的訊息傳遞機制,我們發現HER2的表現量以及活化程度的提升與細胞的生長及細胞群落形成能力上升相關,此外,利用chemical genetic的方式找出PI3K、Rac1、p38以及IKKα受到 HER2 的調控,HER2 藉由PI3K-Rac1-p38-IKKα訊息途徑調控雄性素受體的活 性,另一方面,抑制這個訊息途徑將會造成前列腺癌細胞進行細胞凋亡,除此之 外,p38 的表現量與活化狀態會隨著HER2 表現量改變而同時提升,我們更近一 步的在組織免疫染色上證明HER2 表現量會隨著前列腺癌惡化而提升。綜合我們 的實驗結果,我們發現HER2 影響雄性素受體活性的訊息途徑可能是透過 PI3K-Rac1-p38-IKKα而不是藉由Akt 或Erk1/2。對於HER2 調控雄性素受體訊息 途徑的了解,將有助於提供治療目標,進而治療惡化的前列腺癌或提升治療效果。Escaping from androgen ablation therapy makes prostate cancer intractable. The androgen receptor (AR) remains active and supportive to the scenario of PCa development. Deregulations of normal constraints on hormone action activate AR in androgen ablation therapy. Among these deregulation mechanisms, HER2 is considered as one factor providing outlaw pathway to activate AR. HER2 is well studied in breast cancer progression and its tamoxifen resistance, but role of HER2 in prostate cancer progression remains unclear. Here we investigated the molecular basis downstream of HER2 signaling regarding prostate cancer progression. In 22Rv1 reporter cell lines, higher growth rate and colony forming ability correlate with increase of expression and activation level of HER2. We also showed that HER2-specific inhibitor AG825 decreases AR-activated transcription. Analysis on signaling molecules downstream of HER2 for involvement in AR-activated transcription revealed a complete inhibition of AR activity by targeting to PI3K, Rac1, p38α/β and IKKα IKKα. Moreover, inhibitions on these proteins decline the growth rate and colony forming by inducing apoptosis. On the other hand, increase of p38 levels and activities correlate with the HER2 expression level. Futhermore, immnohistological staining in clinical tissue array demonstrated HER2 expression levels alone with prostatic tumor progression stages. Our data suggest that HER2 overexpression promotes PCa progression might through a PI3K-Rac1-p38α/β-IKKα pathway and is independent of Akt and Erk1/2.     Révélation du rôle des cellules dendritiques conventionnelles de type 1 dans l'orchestration de la fibrose au sein du microenvironnement tumoral

    Dendritic cells (DCs) are essential for developing long-lasting immune responses against pathogens while promoting immune tolerance. In the context of chronic diseases, a small proportion of T cells, known as progenitor exhausted T cells, is maintained and colocalizes with antigen-presenting cells (APCs), forming niches where conventional type 1 dendritic cells (cDC1) support them. Moreover, cDC1 boosts the effector functions of CD8⁺ T cells and is essential for a proper anti-tumor immune response. One critical component of the tumor microenvironment (TME) is the accumulation of the extracellular matrix (ECM) components, particularly collagen, produced by cancer-associated fibroblasts (CAFs), which leads to fibrosis and is linked to poor prognosis. The role of cDC1 in preventing tumor fibrosis remains unknown. Here, we use the XCR1-DTR-Venus mice model to selectively deplete XCR1+ cDC1 and its impact on tumor-infiltrating T-cell distribution, migration capacity, and tumor fibrosis. With flow cytometry and intravital imaging, depletion of cDC1 leads to the expansion of tumor-specific CD8+ T cells with altered distribution and impaired motility within the tumor. With immunostaining, we observe that cDC1 removal leads to the accumulation of tumor fibrosis. By re-introducing splenic cDC1 in tumor-bearing mice, we show that fibrosis is reduced and the motility of exhausted T cells is restored inside the tumor. Our findings uncover an anti-fibrotic role of cDC1 in the TME and have the potential to be extended to chronic inflammatory diseases where dysregulated fibrosis contributes to the underlying pathology

    The symbiotic system in the tumor microenvironment that supports tumor progression

    Il a été découvert que cibler le métabolisme de la glutamine améliore la réponse immunitaire anti-tumorale. Cependant, le principal producteur de glutamine dans le microenvironnement tumoral (TME) et la voie de signalisation sous-jacente qui régule la production de glutamine restent inconnus. Dans cette étude, j'ai découvert que le liquide interstitiel tumoral contient des niveaux plus élevés de glutamine par rapport à celui du sérum dans des modèles de tumeurs murins et des patients atteints de carcinome hépatocellulaire. En explorant le TME dans un modèle de mélanome de souris, j'ai révélé que les fibroblastes associés au cancer (CAF) sont les principaux producteurs de glutamine, qui accentue les phénotypes pro-tumoraux et immunosuppresseurs des macrophages associés aux tumeurs (TAM) via une reprogrammation métabolique. Mécaniquement, les lipides produits par les cellules cancéreuses, en particulier le palmitate, stimulent la spécialisation transcriptomique et fonctionnelle des CAF vers des CAF inflammatoires par un processus de détection des lipides qui dépend du TLR4. Ce processus de détection des lipides dépendant du TLR4 active la voie de signalisation Syk-NF-KB pour déclencher la sécrétion d'IL-6 afin d'orchestrer la polarisation inflammatoire des CAF. À la suite de cette spécialisation fonctionnelle induite par les cellules cancéreuses, les CAF augmentent la production de glutamine pour soutenir le phénotype pro-tumoral dans les TAM. Collectivement, nos résultats dévoilent une symbiose métabolique inexplorée entre les cellules cancéreuses, les CAF et les TAM, qui joue un rôle central dans l'orientation de la nature suppressive des TAM pour favoriser la progression tumorale. -- Targeting glutamine metabolism has been discovered to enhance anti-tumor immune response. However, the main producer of glutamine in the tumor microenvironment and the underlying signaling pathway that regulates glutamine production remain unclear. In this study, I found that tumor interstitial fluid contained higher levels of glutamine compared with that in serum in both murine tumor models and hepatocellular carcinoma patients. By exploring the TME in the mice melanoma model, I further revealed that cancer-associated fibroblasts (CAFs) are the main producer of glutamine that in tum supports pro-tumoral and immunosuppressive phenotypes in tumor-associated macrophages (TAMs) via metabolic reprogramming. Mechanistically, cancer cell-produced lipids, particularly palmitate, stimulate transcriptomic and functional specialization of CAFs toward to inflammatory CAFs via a TLR4-dependent lipid sensing process. This TLR4-dependent lipid-sensing process activates Syk-NF-K13 signaling pathway to trigger IL-6 autocrine for orchestrating inflammatory CAF polarization. As a result of this cancer cell-instructed functional specialization, CAFs increase glutamine production to support pro-tumorigenic phenotype in TAMs. Taken together, our findings unveil an unexplored metabolic symbiosis among cancer cells, CAFs and TAMs, that plays a pivota} role on guiding suppressive nature in TAMs for promoting tumor progression

    Uncoupling protein 2 reprograms the tumor microenvironment to support the anti-tumor immune cycfle

    Plusieurs stratégies ciblant le système immunitaire présentent de bons résultats thérapeutiques dans différentes formes de cancer. Cependant, chez une partie importante des patients, ces traitements ne sont toujours pas efficaces à cause d'un manque d'infiltration des lymphocytes T dans les tumeurs. Il est donc essentiel de comprendre le mécanisme derrière le recrutement des cellules T dans les tumeurs, pour permettre à un nombre plus important de patients de bénéficier de ces traitements. Pour ce faire, nous avons analysé les données provenant de la banque de donnée TCGA de 472 patients atteints de mélanome. Nous avons séparé les données des patients ayant un fort taux d'infiltration lymphocytaire de celles des patients avec un taux plus faible et nous avons comparé leur profil d'expression génétique. Nous avons identifié une corrélation positive entre les niveaux d'expression d'ARNm de la protéine « mitochondrial uncoupling protein 2 » (UCP2) et les réponses antitumorales spécifiques aux lymphocytes T ainsi qu'une augmentation de la survie des patients. Pour mieux comprendre ce mécanisme, nous avons induit la surexpression de l'UCP2 dans des cellules de mélanome. Nos résultats révèlent que la surexpression de cette protéine altère la composition en cytokines dans le milieu et normalise la néovascularisation tumorale in vivo. Des niveaux d'expression de l'UCP2 importants augmentent également l'infiltration des lymphocytes T CD8+ dans le microenvironnement tumoral (TME) en modifiant la production de CXCL10 dépendante de l'IRF5. De plus, les lymphocytes T CD8+ infiltrants les tumeurs sécrètent la chimiokine CCL5 afin de recruter les cellules dendritiques conventionnelles de type 1 (cDCl), la sous-population principale de cellules dendritiques (DC) qui activent de façon préférentielle les cellules T CD8+. L'UCP2 déclenche alors une transition pro inflammatoire grâce à l'engagement d'un cycle immunitaire antitumoral dépendant de l'axe cDCl-lymphocyte T CD8+ dans le TME. Etonnamment, nous avons aussi démontré que l'induction de l'expression de l'UCP2 dans des cellules de mélanome par approche génétique ou par l'administration de rosiglitazone, un agoniste PPAR anti-diabétique approuvé par la FDA, permettait de sensibiliser au traitement visant à bloquer PD-1 des mélanomes qui résistaient auparavant à ce type de thérapie. Nos données montrent donc, que cibler l'UCP2 dans des cellules de mélanome pourrait être une stratégie potentielle pour augmenter l'efficacité des réponses antitumorales aux immunothérapies dans les tumeurs non-infiltrées par les lymphocytes T
