    Investigations on the covalent modification of thiolase by potential hypolipidemic agents

    This work was designed to test several classes of inhibitors on liver cytoplasmic acetoacetyl-CoA thiolase (EC 2.3.19) and heart mitochondrial p-ketoacyl CoA thiolase (EC 2.3.1.16). These inhibitors are based on a new reactive group for alkylation studies, the thiosulfonate group, which readily modifies enzyme sulphydryl groups. The newly synthesised compounds were bifunctional thiolsulfonates of the general structure, CH SO S(CH2 ) SSO2CH3 (n=2 - 12) and CH (CH ) COCH SSO CH . Methyl methanethiosulfonate, (MMTS) injected thiolase in a time dependant reaction and this was protected by acetoacetyl C0A and acetyl CoA. The thiomethyl group could be removed by reduction. This active site directed thiomethylation allowed the study secondary amino modification (succinic, phthalic, and citraconic anhydrides) which also inactivted thiolase in an active site directed manner. When the enzyme was double modified MMTS and citraconic anhydride, the thiomethyl group could be reduced by excess thiols and the amino group could be unblocked by exposure to acidic pH's. The reduction of thethiomethyl-citraconyl thiolase left the enzyme inactive, I whereas exposure to pH 5 followed by reduction led to the formation of an active enzyme. These results are interpreted as demonstrating a role for an amino group of the enzyme active site. A catalytic mechanism is proposed for the enzyme which incorporates this group.In the second part, the compounds were tested aspotential hypolipidemic agents. This was correlated with their inhibitory effects on the cytoplasmic isoenzyme of I thiolase in vitro. In vitro, the compounds inactivated the enzyme in a psuedo first order kinetic pattern in the order of CH (CH? )' COCH'Cl&gt; n=12&gt; n=10&gt; n=8&gt; n=6&gt;CH (CH )1 SS02Cn &gt; n=5&gt; n=2. In vivo when drugs were injected into groups of newly weaned rats fed on a highcarbohydrate or high fat diet, the compounds had a significant hypocholestrolemic effect and a profound hypotriglyceridemic action. Their hypocholesterolemic was in the order of n=12&gt; CH 3(CH2)14000H2SSO2CH3&gt;CH (CH ) COCHCl&gt; n= 8&gt; n=6, and their hypotriglyceridemic effect was nJ&gt; CH (CH ) COCH SSO CH &gt; CH (CH ) COCH C1&gt; n=12&gt; n=10. Their hypolipidemic action resulted in a mild hypoglyceamia, and their growth rate, protein catabolism (BUN, blood total proteins) was normal. The injections of the drugs did not result in any deposition of atherosclerotic lesions in the aotras. When thiolase activity in isolated liver homogenates was measures after injections of drugs in vivo, the enzyme was inactivated by 80%. This inhibition of enzyme activity does not seem enough to account for the hypolipidemic action in vivo since these effects did not correlate with inhibition of the isolated enzyme. I conclude that the drugs must be exerting their effect by influencing additional unidentified thiol dependant processes.</p

    Drug-eluting stents : Insights into safety and indications

    Currently, more than 4 million patients receive drug-eluting stents worldwide. Despite recent studies and editorials that have stirred controversy and generated tremendous publicity in the lay press related to the safety of drug-eluting stents for the treatment of coronary artery disease, enthusiasm for drug-elutiing stents remains high. Drug-eluting stents decrease the need for repeat revascularization, but exhibit delayed endothelialization compared with bare metal stents. Adverse late vessel wall remodeling and inflammation have also been attributed to the drug/polymer coating. Recent registry analysis data suggesting an increased risk of late or very late thrombosis with drug-eluting stents in routine clinical practice compared with previous clinical trial experience has led to concerns regarding the long-term safety of these devices. Clearly, there are many benefits to drug-eluting stents, which are an excellent and appropriate revascularization treatment for many patients with substantial reduction of restenosis compared with bare-metal stents. There is a need for understanding the benefits and risks of these devices in order to use them optimally and reduce likelihood of thrombosis. This review sheds some light on the risks and benefits of drug-eluting stents, and puts this treatment in perspective compared with alternative revascularization treatment options for obstructive coronary artery disease. The important take-home message is that the risks of drug-eluting stent thrombosis versus bare-metal stent thrombosis may have been overstated by some studies

    Kertas kerja ini bertujuan menganalisis watak-watak dan ciri-ciri perwatakan wanita dalam cerpen-cerpen terpilih daripada Kumpulan Cerpen: Meneruskan Perjalanan karya Sasterawan Negara Zurinah Hassan. Cerpen-cerpen terpilih ini menyaksikan kehadiran watak-watak wanita Melayu Islam yang berprinsip agama dan resam Melayu di samping berfikiran dan berazam maju. Mereka terdiri daripada pelajar, pendidik, penulis, pegawai dan sebagainya. Jika ditinjau kajian-kajian lepas kebiasaanya karya-karya sebegini akan diteliti dan dikaji menggunakan kerangka analisis Feminisme Barat. Namun mutakhir ini terdapat beberapa orang sarjana Arab dan Melayu melalui kajian mereka menegaskan kerangka analisis Feminisme Barat ini bercanggah dan bertentangan dengan akidah dan ajaran Islam. Bertitik tolak daripada premis ini maka kertas kerja ini akan menganalisis cerpen-cerpen terpilih Zurinah Hassan berasaskan konsep dan kerangka sastera Islam “al-Adab al-Islami” yang diketengahkan oleh Muhammad Qutb dan seterusnya didokong dan diperteguhkan oleh kritikawan-kritikawan dan sasterawan-sasterawan Arab seperti Najib al-Kilani, Abdul Rahman Ra’fat Basha, Ali al-Tantawi, Imaduddin al-Khalil, Walid Qassab dan lain-lain lagi. Konsep sastera al-Adab al-Islami ini melihat sastera sebagai “ekspresi indah tentang alam, insan dan kehidupan berdasarkan pandangan sarwa Islam, akidah dan nilai-nilai moral keagamaan”

    Self-Portraiture and the Rise of "Arab" Art: Tarek Al-Ghoussein, Hassan Musa and Walid Raad.

    This dissertation offers three monographic case studies of contemporary artists from the Arab world, Tarek Al-Ghoussein (Palestine/Kuwait), Hassan Musa (Sudan/France) and Walid Raad (Lebanon/US), to challenge the prevalent reading and curatorial framing of non-EuroAmerican contemporary art through generalized notions of identity. I focus on self-portraiture, arguing that the artists use it to complicate assumptions about identity. By including Arab artists of diverse national, geographic, religious and diasporic experiences, I start with the premise that the term “Arab” embodies a broad spectrum of identities. Additionally, I outline a history of the international exhibitions and biennials that have contributed to the rise of art from the Middle East and North Africa. I highlight the role of scholars of African art in this process, for by insisting on the diversity of the term “African,” they included Arab North African artists. I position Al-Ghoussein, Musa and Raad within art historical trajectories that illustrate their engagements with western art history while also referencing their collective histories. Al-Ghoussein stages photographs that integrate the genres of self-portraiture, landscape and performance in an interrogation of Palestinian identity, both his as an exile as well as the terrorist connotations prevalent in the media. The themes that emerge-belonging, longing, land and isolation-find resonance in post-1948 Palestinian literature and art. Musa’s paintings illustrate “ArtAfricanism,” a term he coined to critique the art world’s promotion of African art that reflects a narrow conception of African identity. Identifying as “Arab, African and Western,” he produces artworks that reference the display and performance of essentialized identities. Raad references formative theories about photography and historical avant-garde artists to question notions of truth, institutional authority and authenticity. He does this in two multimedia projects about 1) the Lebanese Civil War (1975-1990) and 2) the burgeoning art industry in the Middle East and Arabian Gulf. Al-Ghoussein, Musa and Raad each uniquely approach the project of self-portraiture, tempering the representation of identity with strategies that stymie simplistic readings. By elucidating these artists’ engagements with western art history, this dissertation argues for the expansion of curatorial and scholarly approaches to artists from outside the EuroAmerican context.PhDHistory of ArtUniversity of Michigan, Horace H. These mixers can be either actively or passively driven. The current work focuses on split and recombine micro-mixers which can be employed as heat exchangers, reactors, simple mixers, or all of that combined. In this work, a comprehensive examination of different structures, along with a newly proposed one will be presented. In order to highlight the performance of the proposed mixer under creeping flow conditions, the various designs were compared against each other in terms of pressure drop and mixing quality for Reynolds numbers ranging between 0.001 up to 10. For this reason, a finite element solver, Ansys Polyflow, was used to calculate the pressure drop, Fanning friction factor, concentration profile, and segregation scales. Results show that the newly proposed mixer exhibits a superior performance by means of a better mixing quality and a lower energy consumption rate
