    A School Community Needs Assessment of the Albia Community School District

The purpose of this study was to present an assessment of the educational needs of the Albia Community School District as viewed by parents, voters, and civic leaders in the community, thereby aiding the Board of Education, administration, and faculty in planning the curriculum, facilities, budgets, in-service, etc. Procedure. The opinionnaire method of research was employed to determine the reactions by the parents, voters, and civic leaders. Two methods of administering the opinionnaire were used. One, a group interview technique was used with the civic groups. Secondly, a mailing technique was used to administer the opinionnaire to parents, voters, and the Chamber of Commerce. The results were analyzed by computing the average rating of each objective used in the study and arranging the objectives in order of importance according to the average rating. Thirdly, the objectives were grouped under a major heading and the ratings for the objectives were averaged. That average became the average rating for the major heading. There were sixty objectives used. They were grouped under eighteen major headings. The eighteen major objectives were ranked In order of importance according to their average ratings. Findings. The response to the opinionnaire was good. There were 74.5 percent of the participants that returned the completed opinionnaire. The emphasis of the participants seemed to center on fine arts, use of leisure time, health and safety, and family living. The major objectives' averages ranged from 3.8 to 2.9 on a five point scale where five was the highest possible score. Conclusions. The immediate purpose of the study was to provide a basis for the Albia Community School District to initiate long-range planning of philosophy, goals, and activities, facilities, etc. The study provided that basis by providing a summary of the ratings in order of importance as determined by the participants. The members of the school district feel that emphasis needs to be placed on the fine arts curriculum. In the opinion of the author of the study, the fine arts curriculum in this particular school district needs strengthened. Recommendations. The results of the study should be adopted by the Board of Education and the superintendent or his appointed edministrator should assume leadership for developing an executive committee of a cross-section of the community (including students) to begin a serious study of the curriculum of the Albia Community School District

    An Analysis of the Effects of Prereferral Interventions on the Reduction of Inappropriate Classroom Behaviors

The author identified a number of concerns with traditional special education services which suggest the need to investigate different methods of remediating student problems within the mainstream classroom environment. These included: (a) the educational community's failure to meet the intent of P.L. 94-142, (b) an increasing number of individuals with mild disabilities, (c) the high cost of special education, (d) the lack of objectivity in determining which students are eligible for special education, and (e) the general ineffectivenegs of special education instructional services. The purpose of this study was to determine whether consultants using the Mainstream Assessment Team: A Handbook on Prereferral Intervention (MAT) (Fuchs, Fuchs, Reeder, Gilman, Fernstrom, Bahr, & Moore, 1989) as an operational tool, could successfully assist classroom teachers in reducing inappropriate behaviors of students being considered for special education evaluation. Procedures. Three school psychologists (serving as consultants) received abbreviated instruction in all phases of the MAT. Working with regular education teachers from three different elementary schools, the consultants used MAT techniques in intervening with 14 different students under consideration for special education referral. Findinqs. Compared to gender-matched peers, who served as comparison students, the 14 target students demonstrated a significant reduction in inappropriate behavior, indicating that the MAT can be an effective tool in reducing inappropriate behavior in the mainstream classroom environment. Conclusions. While the MAT proved successful in reducing inappropriate behaviors, additional efforts are necessary to validate its utility as an operational treatment methodology.Specifically, more work is needed in: (a) understanding the dynamics in selecting and training consultants, (b) identifying the types of problems treatable by the MAT, (c) broadening sample sizes and constructing longitudinal studies to strengthen external validity, and (d) socially validating the MAT as a treatment intervention. While this study demonstrated the potential of the MAT as an effective classroom intervention method, until these issues are addressed, its overall usefulness and generalizability in remediating problems without relying on special education services is indeterminable

    Formulation of Biocides Increases Antimicrobial Potency and Mitigates the Enrichment of Nonsusceptible Bacteria in Multispecies Biofilms

    The current investigation aimed to generate data to inform the development of risk assessments of biocide usage. Stabilized domestic drain biofilm microcosms were exposed daily over 6 months to increasing concentrations (0.01% to 1%) of the biocide benzalkonium chloride (BAC) in a simple aqueous solution (BAC-s) or in a complex formulation (BAC-f) representative of a domestic cleaning agent. Biofilms were analyzed by culture, differentiating by bacterial functional group and by BAC or antibiotic susceptibility. Bacterial isolates were identified by 16S rRNA sequencing, and changes in biofilm composition were assessed by high-throughput sequencing. Exposure to BAC-f resulted in significantly larger reductions in levels of viable bacteria than exposure to BAC-s, while bacterial diversity greatly decreased during exposure to both BAC-s and BAC-f, as evidenced by sequencing and viable counts. Increases in the abundance of bacteria exhibiting reduced antibiotic or BAC susceptibility following exposure to BAC at 0.1% were significantly greater for BAC-s than BAC-f. Bacteria with reduced BAC and antibiotic susceptibility were generally suppressed by higher BAC concentrations, and formulation significantly enhanced this effect. Significant decreases in the antimicrobial susceptibility of bacteria isolated from the systems before and after long-term BAC exposure were not detected. In summary, dose-dependent suppression of bacterial viability by BAC was enhanced by formulation. Biocide exposure decreased bacterial diversity and transiently enriched populations of organisms with lower antimicrobial susceptibility, and the effects were subsequently suppressed by exposure to 1% BAC-f, the concentration most closely reflecting deployment in formulated products.IMPORTANCE Assessment of the risks of biocide use has been based mainly on the exposure of axenic cultures of bacteria to biocides in simple aqueous solutions. The current investigation aimed to assess the effects of formulation on the outcome of biocide exposure in multispecies biofilms. Formulation of the cationic biocide BAC significantly increased antimicrobial potency. Bacteria with lower antimicrobial susceptibility whose populations were enriched after low-level biocide exposure were more effectively suppressed by the biocide at in-use concentrations (1% [wt/vol]) in a formulation than in a simple aqueous solution. These observations underline the importance of simulating normal deployment conditions in considering the risks and benefits of biocide use

    Enterobacteriaceae facilitate the anaerobic degradation of glucose by a forest soil

    Anoxic micro zones that occur in soil aggregates of oxic soils may be temporarily extended after rainfall and thus facilitate the anaerobic degradation of organic compounds in soils. The microbial degradation of glucose by anoxic slurries of a forest soil yielded acetate, CO2, H-2, succinate, and ethanol, products indicative of mixed acid fermentation. Prokaryotes involved in this process were identified by time-resolved 16S rRNA gene-targeted stable isotope probing with [C-13-U]-glucose. All labeled phylotypes from the C-13-enriched 16S rRNA gene were most closely related to Rahnella and Ewingella, enterobacterial genera known to catalyze mixed acid fermentation. These results indicate that facultative aerobes, in particular Enterobacteriaceae, (1) can outcompete obligate anaerobes when conditions become anoxic in forest soils and (2) may be involved in the initial decomposition of monosaccharides in anoxic micro zones of aerated forest soils

    Insights into the Fundamental Physiology of the Uncultured Fe-Oxidizing Bacterium Leptothrix ochracea

    Leptothrix ochracea is known for producing large volumes of iron oxyhydroxide sheaths that alter wetland biogeochemistry. For over a century, these delicate structures have fascinated microbiologists and geoscientists. Because L. ochracea still resists long-term in vitro culture, the debate regarding its metabolic classification dates back to 1885. We developed a novel culturing technique for L. ochracea using in situ natural waters and coupled this with single-cell genomics and nanoscale secondary-ion mass spectrophotometry (nanoSIMS) to probe L. ochracea's physiology. In microslide cultures L. ochracea doubled every 5.7 h and had an absolute growth requirement for ferrous iron, the genomic capacity for iron oxidation, and a branched electron transport chain with cytochromes putatively involved in lithotrophic iron oxidation. Additionally, its genome encoded several electron transport chain proteins, including a molybdopterin alternative complex III (ACIII), a cytochrome bd oxidase reductase, and several terminal oxidase genes. L. ochracea contained two key autotrophic proteins in the Calvin-Benson-Bassham cycle, a form II ribulose bisphosphate carboxylase, and a phosphoribulose kinase. L. ochracea also assimilated bicarbonate, although calculations suggest that bicarbonate assimilation is a small fraction of its total carbon assimilation. Finally, L. ochracea's fundamental physiology is a hybrid of those of the chemolithotrophic Gallionella-type iron-oxidizing bacteria and the sheathed, heterotrophic filamentous metal-oxidizing bacteria of the Leptothrix-Sphaerotilus genera. This allows L. ochracea to inhabit a unique niche within the neutrophilic iron seeps. IMPORTANCE Leptothrix ochracea was one of three groups of organisms that Sergei Winogradsky used in the 1880s to develop his hypothesis on chemolithotrophy. L. ochracea continues to resist cultivation and appears to have an absolute requirement for organic-rich waters, suggesting that its true physiology remains unknown. Further, L. ochracea is an ecological engineer; a few L. ochracea cells can generate prodigious volumes of iron oxyhydroxides, changing the ecosystem's geochemistry and ecology. Therefore, to determine L. ochracea's basic physiology, we employed new single-cell techniques to demonstrate that L. ochracea oxidizes iron to generate energy and, despite having predicted genes for autotrophic growth, assimilates a fraction of the total CO2 that autotrophs do. Although not a true chemolithoautotroph, L. ochracea's physiological strategy allows it to be flexible and to extensively colonize iron-rich wetlands

    Comparative Genomics Reveals the Diversity of Restriction-Modification Systems and DNA Methylation Sites in Listeria monocytogenes

    Listeria monocytogenes is a bacterial pathogen that is found in a wide variety of anthropogenic and natural environments. Genome sequencing technologies are rapidly becoming a powerful tool in facilitating our understanding of how genotype, classification phenotypes, and virulence phenotypes interact to predict the health risks of individual bacterial isolates. Currently, 57 closed L. monocytogenes genomes are publicly available, representing three of the four phylogenetic lineages, and they suggest that L. monocytogenes has high genomic synteny. This study contributes an additional 15 closed L. monocytogenes genomes that were used to determine the associations between the genome and methylome with host invasion magnitude. In contrast to previous findings, large chromosomal inversions and rearrangements were detected in five isolates at the chromosome terminus and within rRNA genes, including a previously undescribed inversion within rRNA-encoding regions. Each isolate's epigenome contained highly diverse methyltransferase recognition sites, even within the same serotype and methylation pattern. Eleven strains contained a single chromosomally encoded methyltransferase, one strain contained two methylation systems (one system on a plasmid), and three strains exhibited no methylation, despite the occurrence of methyltransferase genes. In three isolates a new, unknown DNA modification was observed in addition to diverse methylation patterns, accompanied by a novel methylation system. Neither chromosome rearrangement nor strain-specific patterns of epigenome modification observed within virulence genes were correlated with serotype designation, clonal complex, or in vitro infectivity. These data suggest that genome diversity is larger than previously considered in L. monocytogenes and that as more genomes are sequenced, additional structure and methylation novelty will be observed in this organism. IMPORTANCE: Listeria monocytogenes is the causative agent of listeriosis, a disease which manifests as gastroenteritis, meningoencephalitis, and abortion. Among Salmonella, Escherichia coli, Campylobacter, and Listeria-causing the most prevalent foodborne illnesses-infection by L. monocytogenes carries the highest mortality rate. The ability of L. monocytogenes to regulate its response to various harsh environments enables its persistence and transmission. Small-scale comparisons of L. monocytogenes focusing solely on genome contents reveal a highly syntenic genome yet fail to address the observed diversity in phenotypic regulation. This study provides a large-scale comparison of 302 L. monocytogenes isolates, revealing the importance of the epigenome and restriction-modification systems as major determinants of L. monocytogenes phylogenetic grouping and subsequent phenotypic expression. Further examination of virulence genes of select outbreak strains reveals an unprecedented diversity in methylation statuses despite high degrees of genome conservation

    Britain’s Right to Roam: Redefining the Landowner's Bundle of Sticks

    Jerry L. Anderson is the Richard M. and Anita Calkins Professor of Law, Drake University Law School in Des Moines, Iowa.Britain recently enacted a “right to roam” in the Countryside and Rights of Way Act (CRoW) 2000. At first glance, CRoW appears to be a dramatic curtailment of the landowner’s traditional right to exclude; it opens up all private land classified as “mountain, moor, heath, or down” to the public for hiking and picnicking. Yet, when viewed in the light of history, CRoW may be seen as partially restoring to the commoner rights lost during the enclosure period, when the commons system ended. CRoW also represents a return to a functional rather than spatial form of land ownership, allowing more than one party to have rights in a particular piece of land. The new law highlights some important public values regarding freedom of access that have been all but forgotten in the United States. The law calls into question U.S. Supreme Court precedent that has enshrined the right to exclude as an “essential” stick in the bundle of property rights and serves as a powerful alternative to the Court’s formalistic notion of property rights. Given the differences in its history, culture, and legal system, the United States is unlikely to follow Britain’s lead in enacting a right to roam; nevertheless, the study of CRoW contains valuable lessons for Americans.The author wishes to thank the anonymous donor of the Drake International Research Stipend who generously enabled this research
