    Transformation of (allo)securinine to (allo)norsecurinine via a molecular editing strategy

    Securinega alkaloids have intrigued chemists since the isolation of securinine in 1956. This family of natural products comprises a securinane subfamily with a piperidine substructure and norsecurinane alkaloids featuring a pyrrolidine core. From a biosynthetic perspective, the piperidine moiety in securinane alkaloids derives from lysine, whereas the pyrrolidine moiety in norsecurinane natural products originates from ornithine, marking an early biogenetic divergence. Herein, we introduce a single-atom deletion strategy that enables the late-stage conversion of securinane to norsecurinane alkaloids. Notably, for the first time, this method enabled the transformation of piperidine-based (allo)securinine into pyrrolidine-based (allo)norsecurinine. Straightforward access to norsecurinine from securinine, which can be readily extracted from the plant Flueggea suffruticosa, abundant across the Korean peninsula, holds promise for synthetic studies of norsecurinine-based oligomeric securinega alkaloids

    In the yeast Saccharomyces cerevisiae, the mineral zinc is essential for growth and metabolism. The effects of zinc depletion on the regulation of the PIS1-encoded phosphatidylinositol synthase were examined. Phosphatidylinositol synthase activity increased when zinc was depleted from the growth medium. Analysis of a zrt1D zrt2D mutant defective in plasma membrane zinc transport indicated that the cytoplasmic levels of zinc were responsible for the regulation of phosphatidylinositol synthase. PIS1 mRNA, its encoded protein Pis1p, and the b-galactosidase activity driven by the PPIS1-lacZ reporter gene were elevated in zinc-depleted cells. This indicated that the increase in phosphatidylinositol synthase activity was due to a transcriptional mechanism. The regulation of PIS1 gene expression by zinc depletion was mediated by the zinc-regulated transcription factor Zap1p. This work advances understanding of phospholipid synthesis regulation by zinc and the transcription control of the PIS1 gene. Mg2+-dependent phosphatidate phosphatase catalyzes the dephosphorylation of phosphatidate to synthesize diacylglycerol and inorganic phosphate. There are at least two genes that encode Mg2+-dependent phosphatidate phosphatase enzymes in yeast. The PAH1 gene encodes an enzyme that is insensitive to N-ethylmaleimide whereas the gene that encodes the N-ethylmaleimide-sensitive enzyme has yet to be identified. The aim of this work was to purify and characterize the N-ethylmaleimide-sensitive Mg2+-dependent phosphatidate phosphatase. To purify the enzyme, a pah1D dpp1D lpp1D triple mutant that is devoid of all known phosphatidate phosphatase enzymes was used. The Mg2+-dependent phosphatidate phosphatase was extracted from the membrane fraction using 1 M NaCl. The salt extracted enzyme was then purified by chromatography with phosphocellulose, Mono Q and Superose 6. The enzyme was enriched 2,250 fold by this procedure. The basic enzymological properties of the purified Mg2+-dependent phosphatidate phosphatase were examined. The enzyme had a pH optimum from 7 to 7.5, and was dependent on Mg2+ ions for activity. The enzyme activity followed surface dilution kinetics using Triton X-100-phosphatidate mixed micelles. The enzyme was inhibited by N-ethylmaleimide, phenylglyoxal and propranolol. In addition, the enzyme activity was regulated by nucleotides, sphingolipids and phospholipids.     Quality characteristics of West African shea butter (Vitellaria paradoxa) and approaches to extend shelf-life

    Shea butter is a versatile plant fat extracted from kernels of shea nuts, seeds of shea trees (Vitellaria paradoxa). Shea butter has long been used in sub-Saharan Africa for medicinal, culinary, and other applications and serves as a cocoa butter equivalent in the manufacture of chocolate as well as an ingredient in cosmetics. Shea butter, rich in unsaturated fatty acids undergoes hydrolytic and oxidative degradation during post-harvest processing and storage, resulting in inconsistent and degraded quality and limited shelf-life. The objective of this study was to assess important quality characteristics of shea butter. Seven West African shea butters were analyzed to measure physicochemical parameters by wet chemical tests and to measure chemical composition by gas chromatographic analysis. Physical properties were consistent among samples and within the range of typical shea butter. The samples also shared similar chemical compositions, showing palmitic (3.36-4.44 % of total fatty acids), stearic (39.74-44.62 %), oleic (40.71-44.48 %), and linoleic acids (5.73-6.41 %) as the major fatty acids and α-amyrin having anti-inflammatory property (57.26-64.37 % of total sterols and triterpenes) as the major unsaponifiable matter. Moisture, insoluble impurities, free fatty acids, and peroxide values were needed to be controlled. Free fatty acid level was the most variable parameter, ranged from 1.07 to 8.56 %. Peroxide value was low enough except the one which was as high as 15.32 mEq/kg. Total unsaponifiable matters were measured lower (2.21-4.18 %) compared to the previous studies (4-11 %) but still higher than many other plant oils and fats (~2 %). This study also aimed at identifying the protective effect of selected synthetic (BHT) and natural (rosmarinic and gallic acids) antioxidants on shea butter from oxidation. Peroxide value, conjugated dienes, thiobarbituric acid reactive substances, and the amount of major fatty acids were measured as oxidative parameters at 0, 72 and 144 hours while the control and samples with 0.02 % of antioxidants were stored at 90 °C with air flow. The antioxidants were significantly effective in protecting shea butter from oxidation and no significant difference in the effect of synthetic and natural antioxidants was found.M.S.Includes bibliographical referencesby Hee Seung Nah

    Developing a best practice framework for clinical competency education in the traditional East-Asian medicine curriculum

    Background While clinical competency is crucial for traditional East-Asian medical education, available studies on the educational system for fostering clinical performance are scarce. This study aimed to review the educational system, curriculum, facilities, and management of current traditional East-Asian medicine in a well-established university of Korea and develop a Best Practice Framework (BPF) of clinical competency education. Methods The clinical competency education system in Pusan National University School of Korean Medicine was systematically described through 5 steps of governance of the educational system, competency of the graduates, educational resources, assessment strategies and tools, and gaps in the curriculum. We also reviewed the experiences in education and the points to be improved. Results The Office of Traditional Korean Medicine Education governs the development, implementation, and evaluation of the educational curriculum for cultivating students&apos; clinical competency. Medical students have undertaken 39 modules of clinical biomedicine and 21 of traditional medicine during the clinical clerkship courses in an affiliated hospital, Clinical Skill Practice Center, clinical research center, practice lab for medical herb, and other locations. After training, 15 modules of simulated clinical training using standardized patients, students&apos; clinical competency are evaluated by a Clinical Performance Test using a Clinical Performance Examination (CPX) and an Objective Structured Clinical Examination (OSCE) for biomedical and traditional medical skills. Conclusions A clinical competency framework is required for a qualified physician of traditional East-Asian medicine. This study reviewed the current well-organized educational system of Korean traditional medicine in detail, which can be used for the BPF of competency-based clinical education. We expect the current study to be a representative reference for establishing an educational system of traditional medicine such as acupuncture and medical herbs in other countries.N

    Advances in proteomic technologies have led to the creation of large-scale proteome databases that can be used to elucidate invaluable information on the dynamics of the metabolic, signaling and regulatory networks and to aid understanding of physiological changes. In particular, proteomics can have practical applications, for example, through the identification of proteins that may be potential targets for the biotechnology industry, and through the extension of our understanding of the physiological action of these proteins. In this review, we describe proteomic approaches for the discovery of targets that have potential biotechnological applications. These targets include promoters, chaperones, soluble fusion partners, anchoring motifs, and excretion fusion partners. In addition, we discuss the potential applications of proteomic techniques for the design of future bioprocesses and the optimization of existing ones. Successful applications of proteomic information have proven to have enormous value for both scientific and practical applications. (C) 2009 Elsevier B.V.
