Karanjin, A Promising Bioactive Compound Possessing Anti-cancer Activity against Experimental Model of Non-small Cell Lung Cancer Cells
Aims: The aim of this study is to isolate the Millettia pinnata (Karanj) leaf extract for pure compound with anticancer properties and to study the molecular target of the isolates in non-small cell lung cancer cell lines. Background: In our earlier research Millettia pinnata leaf extract has demonstrated potential anticancer activities. Thus, in pursuit of the bioactive compounds, the most potential active extract from our previous study was purified. Furthermore, the anticancer properties of the isolated compound karanjin was studied and aimed for apopto-sis and restraining growth. Methods: A novel method was developed through column chromatography for isolation and purification of the compound karanjin from leaf chloroform extract. The purified component was then characterised using FTIR, mass spectrometry, and NMR. An MTT-based cytotoxicity assay was used to analyse cell cytotoxicity, whereas fluorescence staining was used for apoptosis and reactive oxygen species inhibition quantification. Furthermore, the real-time PCR assay was used to determine the molecular mechanism of action in cells causing cytotoxicity induced by karanjin dosing. Results: The anticancer activity of karanjin in A549 cell line exhibited prominent activity revealing IC50 value of 4.85 μM. Conferring the predicted molecular pathway study, karanjin restrains the proliferation of cancer cells through apoptosis, which is controlled by extrinsic pathway proteins FAS/FADD/Caspases 8/3/9. Down-regulation of KRAS and dependent gene expression also stopped cell proliferation. Conclusion: Karanjin has been identified as a compound with potential effect in non-small cell lung cancer cells. Molecular mechanism for apoptosis and inhibition of reactive oxygen species induced through H2O2 were observed, concluding karanjin have medicinal and antioxidant properties
3D Hierarchical Boron-Doped Diamond-Multilayered Graphene Nanowalls as an Efficient Supercapacitor Electrode
Synthesis of stable hybrid carbon nanostructure for high-performance supercapacitor electrode with long life-cycle for electronic and energy storage devices is a real challenge. Here, we present a one-step synthesis method to produce conductive boron-doped hybrid carbon nanowalls (HCNWs), where sp(2)-bonded graphene has been integrated with and over a three-dimensional curved wall-like network of sp(3)-bonded diamond. The spectroscopic studies such as X-ray absorption, Raman, and X-ray photoelectrons clearly reveal the coexistence of diamond and graphene in these nanowalls, while the detailed transmission electron microscopy studies confirm the unique microstructure where a diamond nanowall is encased by a multilayered graphene. Interestingly, these HCNWs yield a high double layer capacitance value of 0.43 mF cm(-2) and electrode retention of 98% over 10 000 cycles of charging/discharging in 1 M Na2SO4 electrolyte. A 3-year-old patient in India experiencing headaches and seizures was diagnosed with a fungal infection, initially misidentified as Cladophialophora bantiana. Follow-up sequencing identified the isolate to be Fonsecaea monophora fungus. An evaluation of diode laser as an adjunct to scaling and root planning in the nonsurgical treatment of chronic periodontitis: A clinico-microbiological study
Introduction: Laser is an acronym for "light amplification by stimulated emission of radiation." They are being developed for a variety of uses in periodontal therapy. The diode laser in dentistry is used for treatment of diseased periodontal soft tissue, allowing significant bacterial reduction, and removal of the inflammatory products while creating excellent hemostasis. A flexible fiber optic delivery system allows the clinician easy and safe access around the periodontal pocket. The wavelengths are transmitted through water and are very poorly absorbed in apatite crystals, making them an excellent choice to use in a periodontally involved sulcus that has inflamed tissue and pigmented bacteria. This study was undertaken to evaluate the effectiveness of diode laser as an adjunct to scaling and root planing (SRP) in the nonsurgical treatment of chronic periodontitis. Aim: To evaluate the effectiveness of diode laser on plaque index (PI), gingival index (GI) probing pocket depth (PPD) and clinical attachment level (CAL) in chronic periodontitis patients and to compare the outcome with SRP alone. To evaluate the efficacy of diode laser on plaque microorganisms namely Aggregatibacter actinomycetemcomitans and Prevotella intermedia. Materials and Methods: A total number of 20 generalized chronic periodontitis patients were selected for the study. A randomized split-mouth design was followed to assign quadrants in each to one of the two treatment groups: SRP alone (Group I control group) and diode laser as an adjunct to SRP (Group II test group). The assessment of clinical parameters was carried out at baseline, day 30, 90, and 180. The microbiological analysis was done prior to the start of the treatment at baseline, day 15, 30, 90, and 180, respectively. Results: At baseline mean colony count was equal in both groups, however, at all the subsequent time intervals, mean colony count was lower in Group II as compared to Group I. All the groups showed statistically significant reduction in relation to PI, GI, PPD, CAL, and microbial count. Discussion: In this study, an attempt was made to evaluate the adjunctive role of diode lasers on gingival inflammation, pocket probing depth, the level of clinical attachment, and microbial count in relation to SRP using a case-control design. Recently, the use of laser has emerged as an adjunctive procedure in enhancing the clinical outcome of the periodontal treatment. It is one of the most promising new technical modalities for nonsurgical periodontal treatment and has an advantage of reaching sites that cannot be approached by conventional mechanical instrumentation. Conclusion: In the present study, an attempt was made to evalute the adjunctive role of diode lasers on plaque index, gingival index, pocket probing depth, level of clinical attachment and microbial count in relation to scaling and root planning using a case-control design. On the basis of these findings, adjunctive treatment with diode laser at a higher but clinically safe frequency (940nm) at repeated intervals showed a better efficacy in ensuring a better periodontal health as compared to SRP alone
Anxiolytic activity of Nymphaea alba Linn. in mice as experimental models of anxiety
Objective : The aim of the present work was to evaluate the anxiolytic
effect of an ethanolic extract of Nymphaea alba Linn. in mice.
Materials and Methods : The elevated plus maze test (EPMT), light and
dark test (L and DT) and open field test (OFT) were used to assess the
anxiolytic activity of the ethanolic extract of N. alba Linn. in mice.
In addition, aggressive behavior and motor coordination was also
assessed by foot shock induced aggression test (FSIAT) and rota rod
test (RRT). Diazepam 1 mg/kg served as a standard anxiolytic drug,
administered orally. Results : The ethanolic extract of N. alba (100
and 200 mg/kg, p.o.) significantly increased the percentage of time
spent and number of entries in open arm in EPMT. In L and DT, the
extract produced significant increase in time spent, number of crossing
and decrease in the duration of immobility in light box. In OFT, the
extract showed significant increase in number of rearings, assisted
rearings and number of square crossed, all of which are demonstrations
of exploratory behavior. In FSIAT, N. alba extract attenuated
aggressive behavior related to anxiolytic activity, such as number of
vocalization, leaps, rearing, biting/attacks and facing each other in
paired mice. Furthermore, the extract produced skeletal muscle relaxant
effect assessed by RRT. Conclusion : The results of the present study
suggest that an ethanolic extract of N. alba may possess anxiolytic
activity and provide a scientific evidence for its traditional claim
Multimedia in Teacher Education: Perceptions & Uses
Educational systems around the world are under increasing pressure to use the new technologies to teach students the knowledge and skills they need in the 21st century. Education is at the confluence of powerful and rapidly shifting educational, technological and political forces that will shape the structure of educational systems across the globe for the remainder of this century. Many countries are engaged in a number of efforts to effect changes in the teaching/learning process to prepare students for an information and technology based society. Multimedia provide an array of powerful tools that may help in transforming the present isolated, teacher-centred and text-bound classrooms into rich, student-focused, interactive knowledge environments. The schools must embrace the new technologies and appropriate multimedia approach for learning. They must also move toward the goal of transforming the traditional paradigm of learning. Teacher education institutions may either assume a leadership role in the transformation of education or be left behind in the swirl of rapid technological change. For education to reap the full benefits of multimedia in learning, it is essential that pre-service and in-service teachers have basic skills and competencies required for using multimedia. This paper, therefore, explores the latent benefit of using multimedia tools particularly in professional teacher education. The paper also discusses why pre-service teachers need to use multimedia technologies within the context of students' familiar, technology-rich living spaces to develop their own teaching skills and the technology skills of their students.
