    A Gα(s) carboxyl-terminal peptide prevents G(s) activation by the A(2A) adenosine receptor

    The molecular mechanisms of interaction between G(s) and the A(2A) adenosine receptor were investigated using synthetic peptides corresponding to various segments of the Galpha(s) carboxyl terminus. Synthetic peptides were tested for their ability to modulate binding of a selective radiolabeled agonist, [(3)H]2-[4-(2-carboxyethyl)phenylethylamino]-5'-N-ethylcarboxam idoade nosine ([(3)H]CGS21680), to A(2A) adenosine receptors in rat striatal membranes. The Galpha(s) peptides stimulated specific binding both in the presence and absence of 100 microM guanosine-5'-O-(3-thiotriphosphate) (GTPgammaS). Three peptides, Galpha(s)(378-394)C(379)A, Galpha(s)(376-394)C(379)A, and Galpha(s)(374-394)C(379)A, were the most effective. In the presence of GTPgammaS, peptide Galpha(s)(374-394)C(379)A increased specific binding in a dose-dependent fashion. However, the peptide did not stabilize the high-affinity state of the A(2A) adenosine receptor for [(3)H]CGS21680. Binding assays with a radiolabeled selective antagonist, [(3)H]5-amino-7-(2-phenylethyl)-2-(2-furyl)pyrazolo[4, 3-e]-1,2,4-triazolo[1,5-c]pyrimidine ([(3)H]SCH58261), showed that the addition of the Galpha(s) peptide modified the slope of the 5'-N-ethylcarboxamidoadenosine (NECA) competition curve, suggesting modulation of receptor affinity states. In the presence of GTPgammaS, the displacement curve was right-shifted, whereas the addition of Galpha(s)(374-394)C(379)A caused a partial left-shift. Both curves were fitted by one-site models. This same Galpha(s) peptide was also able to disrupt G(s)-coupled signal transduction as indicated by inhibition of the A(2A) receptor-stimulated adenylyl cyclase activity without affecting either basal or forskolin-stimulated enzymatic activity in the same membrane preparations. Shorter peptides from Galpha(s) and Galpha(i1/2) carboxyl termini were not effective. NMR spectroscopy showed the strong propensity of peptide Galpha(s)(374-394)C(379)A to assume a compact carboxyl-terminal alpha-helical conformation in solution. Overall, our results point out the conformation requirement of Galpha(s) carboxyl-terminal peptides to modulate agonist binding to rat A(2A) adenosine receptors and disrupt signal transduction

    Truth is in the head. A nod and shake compatibility effect

    Studies from the embodiment perspective on language processing have shown facilitation or interference effects depending on the compatibility between verbal contents, concrete or abstract, and the motion of various parts of the body. The aim of the present study was to test whether such compatibility effects can be found when a higher cognitive process like truth evaluation is accomplished with head movements. Since nodding is a vertical head gesture typically performed with positive and affirmative responses, and shaking is a horizontal head gesture associated with negative and dissenting contents, faster response times can be expected when true information is evaluated by making a vertical head movement and false information by making a horizontal head movement. Three experiments were designed in order to test this motor compatibility effect. In the first experiment a series of very simple sentences were asked to be evaluated as true or false by dragging them vertically and horizontally with the head. It resulted that truth-value was assessed faster when it was compatible with the direction of the head movement, compared to when it was incompatible. In the second experiment participants were asked to evaluate the same sentences as the first experiment but by moving them with the mouse. In the third experiment, a non-evaluative classification task was given, where sentences concerning animals or objects were to be dragged by vertical and horizontal head movements. In the second and third experiment no compatibility effect was observed. Overall results support the hypothesis of an embodiment effect between the abstract processing of truth evaluation and the direction of the two head movements of nodding and shaking. Cultural aspects, cognitive implications, and the limits of these findings are discussed

    Role of hypovitaminosis D in the pathogenesis of obesity-induced insulin resistance

    Obesity and type 2 diabetes have both rapidly increased during the last decades and are continuing to increase at an alarming rate worldwide. Obesity and impaired glucose homeostasis are closely related, and during the last decades of investigation about vitamin D, several clinical and epidemiological studies documented an inverse correlation between circulating vitamin D levels, central adiposity and the development of insulin resistance and diabetes. The insufficient sun exposure and outdoor activities of obese individuals, the storage of vitamin D in adipose tissue, because of its lipophilic properties, and the vitamin D-mediated modulation of adipogenesis, insulin secretion, insulin sensitivity and the immune system, are the main reasons for the close relationship between obesity, glucose homeostasis and hypovitaminosis D. Then objective of this review is to explore the pathophysiological mechanism(s) by which vitamin D modulates glycemic control and insulin sensitivity in obese individuals
