    Temporal variation of diversity in Italian durum wheat germplasm

    The aim of this work is to analyse the temporal change of genetic diversity in Italian durum wheat germplasm. The germplasm deployed in this study (158 accessions), belonging to 5 different historical classes, was characterised for its microsatellite and gliadin markers. The level of genetic diversity (He), based on gliadin and SSR markers results – on average – greater in indigenous landraces present in Italy before 1915, with the exception of pure line material which had been selected from landraces (showing highest level of heterozigosity for gliadin markers). Genotypes obtained from crosses or mutagenesis (referring to the 1950–1960 period) along with those resulting from crosses between CIMMYT lines and old materials (1970s and beyond) were also genetically more diverse. Forty-nine percent of indigenous landraces were genetically heterogeneous. Nine out of 53 landrace accessions were able to capture 4 different SSR private alleles. It is speculated that the reduction of allele richness is an indicator of the genetic erosion of the pre-breeding germplasm and it is pointed out that the implementation of appropriate methods of genetic conservation of this germplasm is a priority for breeding and food safet

    Characterization of DNA methylation activity by gene expression analysis in Fragaria Vesca

    DNA methylation represents one of the key processes that play an important role in the transcriptional control of gene expression. The role of cytosine methylation in plant development has been demonstrated. For this purpose gene expression analysis has been widely applied. The present work was directed to demonstrate how nitrate based manuring could induce DNA methylation in Fragaria vesca. Differential display analysis was used for this purpose. Two Fragaria vesca genotypes were analyzed. For each genotype a total of 40 plants were used. Plants were divided into two groups of twenty plants each. The first group was treated with a concentrated solution of calcium nitrate, the second was treated using an organic nitrogen solution at the same molar concentration. Plant material was sampled six times. The first sampling was before treatment, and subsequent sampling was done respectively 1, 3, 7, 15 and 20 days after the first one. For each genotype, leaves and roots were sampled and stored in liquid nitrogen. Genomic DNA was isolated and analyzed by applying isoschizomers to evaluate the extent of DNA methylation. This showed an increase of the DNA methylation in plants treated with calcium nitrate. Total RNA was separately purified from root and leaf and, in order to isolate differentially expressed genome fractions, cDNA was obtained. To generate differentially expressed sequences between different treatments and between sampling times, random and specific primers were applied. 62 differential fragments were isolated and sequenced. All sequences were compared to the database to evaluate similarity with previously isolated genes. Sequence analysis showed that 55% could be classified as ESTs. The other sequences showed homologies with genes related to abiotic stress. In particular it was possible to show a reduction of transcriptomic activity in the sample treated with calcium nitrate. Sequence analysis demonstrated that gene fractions isolated from plants treated with nitrate showed high homology with glycosylase, a gene related with DNA methylation repair action

    Entender cualquier actividad humana, sea social o laboral, exige partir de un buen diagnóstico de los principales rasgos demográficos. En el caso de las parturientas que concurrieron en un determinado momento a la sala de partos, brinda la clave para entender determinadas acciones que se debieron tomar en su momento y que a veces, por no contar con la información correcta se generaron creencias erróneas y, esto a su vez, afectó la conducta humana. El análisis brinda una imagen de las características de una determinada población. La información obtenida por medio de la recolección y procesamiento de datos, es empleada para estudiar las principales características de una determinada población, las cuales permiten explicar hasta un cierto grado, el comportamiento demográfico y social. La falta de datos, representa un problema, tanto a nivel institucional como para el accionar de enfermería, ya que a través de los mismos se podría contar con una base de información que permita un diagnóstico de la situación sociodemográfica de las parturientas, para actuar en acciones de planificación de actividades educativas y asistenciales. Se trabajó en general, con aspectos relacionados con la cantidad de pacientes que concurren a sala de partos, lugar de procedencia, su composición por edad materna, obra social más frecuente, edad estacional del nacido, así como la puntuación de apgar, peso, el tipo de nacimiento y la fecha en que se registraron mayor número de nacimientos.Fil: Greco, Ivana.
