    The chapter forms part of a volume entitled Aesthetics of Dislocation in French and Francophone Literature and Art: Strategies of Representation, published by Edwin Mellen Press in 2009, co-edited with two other colleagues, Dr Daisy Connon and Dr Gillian Jein. This chapter is concerned with a new articulation of poetic discourse in the writings of the nineteenth-century French poet and journalist Théophile Gautier. Through analysis of his profuse feuilletons, travel writings and art criticism, this chapter argues that Gautier’s prose exhibits and acute perception of the facts determining its construction as text and dramatizes these as a means to explore a range of complex objects of representation which are encountered throughout the topography of the modern urban; the latter include train stations, ports and the Universal Exhibition halls. Taken together, these texts identify the shifting urban environment as a source of poetic renewal. In so doing, they reveal a dramatically more comprehensive and dynamic vision than is permitted by the narrowly aestheticist views by which Gautier’s work is normally characterized

    Fabrication of a corneal-limbal tissue substitute using silk fibroin

    Fibroin extracted from silkworm cocoon silk provides an intriguing and potentially important biomaterial for corneal reconstruction. In the present chapter we outline our methods for producing a composite of two fibroin-based materials that supports the co-cultivation of human limbal epithelial (HLE) cells and human limbal stromal (HLS) cells. The resulting tissue substitute consists of a stratified epithelium overlying a three-dimensional arrangement of extracellular matrix components (principally ‘degummed’ fibroin fibers) and mesenchymal stromal cells. This tissue substitute is currently being evaluated as a tool for reconstructing the corneal limbus and corneal epithelium

    Permafrost Monitoring at Scotty Creek Research Station, 2011-2017 [Northwest Territories, Canada]

    A physically-based one-dimensional sharp-interface model of active layer evolution and permafrost thaw is presented. This computationally efficient, semi-analytical, non-equilibrium solution to soil freeze-thaw problems in partially saturated media is proposed as a component of hydrological models to describe seasonal ground ice, active layer evolution and changes in permafrost temperature and extent. The model is developed and validated against the analytical Stefan solution and a finite volume coupled heat and mass transfer model of freeze-thaw in unsaturated porous media. Unlike analytic models, the interface model provides a non-equilibrium solution to the heat equation while permitting a wide range of temporally variable boundary conditions and supporting the simulation of multiple interfaces between frozen and unfrozen soils. The model is implemented for use in discontinuous permafrost peatlands where soil properties are highly dependent on soil ice content and infiltration capacity is high. It is demonstrated that the model is suitable for the representation of variably saturated active layer and permafrost evolution in cases both with and without a talik

    Changes in culture expanded human amniotic epithelial cells: Implications for potential therapeutic applications

    Human amniotic epithelial cells (hAEC) isolated from term placenta have stem cell-like properties, differentiate into tissue specific cells and reduce lung and liver inflammation and fibrosis following transplantation into disease models established in mice. These features together with their low immunogenicity and immunosuppressive properties make hAEC an attractive source of cells for potential therapeutic applications. However, generation of large cell numbers required for therapies through serial expansion in xenobiotic-free media may be a limiting factor. We investigated if hAEC could be expanded in xenobiotic-free media and if expansion altered their differentiation capacity, immunophenotype, immunosuppressive properties and production of immunomodulatory factors. Serial expansion in xenobiotic-free media was limited with cumulative cell numbers and population doubling times significantly lower than controls maintained in fetal calf serum. The epithelial morphology of primary hAEC changed into mesenchymal-stromal like cells by passage 4-5 (P4-P5) with down regulation of epithelial markers CK7, CD49f, EpCAM and E-cadherin and elevation of mesenchymal-stromal markers CD44, CD105, CD146 and vimentin. The P5 hAEC expanded in xenobiotic-free medium differentiated into osteocyte and alveolar epithelium-like cells, but not chondrocyte, hepatocyte, α- and β-pancreatic-like cells. Expression of HLA Class IA, Class II and co-stimulatory molecules CD80, CD86 and CD40 remained unaltered. The P5 hAEC suppressed mitogen stimulated T cell proliferation, but were less suppressive compared with primary hAEC at higher splenocyte ratios. Primary and P5 hAEC did not secrete the immunosuppressive factors IL-10 and HGF, whereas TGF-β1 and HLA-G were reduced and IL-6 elevated in P5 hAEC. These findings suggest that primary and expanded hAEC may be suitable for different cellular therapeutic applications
