    An intracellular pH gradient in the anammox bacterium Kuenenia stuttgartiensis as evaluated by (31)P NMR

    The cytoplasm of anaerobic ammonium oxidizing (anammox) bacteria consists of three compartments separated by membranes. It has been suggested that a proton motive force may be generated over the membrane of the innermost compartment, the “anammoxosome”. 31P nuclear magnetic resonance (NMR) spectroscopy was employed to investigate intracellular pH differences in the anammox bacterium Kuenenia stuttgartiensis. With in vivo NMR, spectra were recorded of active, highly concentrated suspensions of K. stuttgartiensis in a wide-bore NMR tube. At different external pH values, two stable and distinct phosphate peaks were apparent in the recorded spectra. These peaks were equivalent with pH values of 7.3 and 6.3 and suggested the presence of a proton motive force over an intracytoplasmic membrane in K.stuttgartiensis. This study provides for the second time—after discovery of acidocalcisome-like compartments in Agrobacterium tumefaciens—evidence for an intracytoplasmic pH gradient in a chemotrophic prokaryotic cell.BiotechnologyApplied Science

    Usability issues with security of electronic mail

    This thesis was submitted for the degree of Doctor of Philosophy and awarded by Brunel University.This thesis shows that human factors can have a large and direct impact on security, not only on the user’s satisfaction, but also on the level of security achieved in practice. The usability issues identified are also extended to include mental models and perceptions as well as traditional user interface issues. These findings were accomplished through three studies using various methodologies to best suit their aims. The research community have issued principles to better align security and usability, so it was first necessary to evaluate their effectiveness. The chosen method for achieving this was through a usability study of the most recent software specifically to use these principles. It was found that the goal of being simultaneously usable and secure was not entirely met, partially through problems identified with the software interface, but largely due to the user’s perceptions and actions whilst using the software. This makes it particularly difficult to design usable and secure software without detailed knowledge of the users attitudes and perceptions, especially if we are not to blame the user for security errors as has occurred in the past. Particular focus was given to e-mail security because it is an area in which there is a massive number of vectors for security threats, and in which it is technologically possible to negate most of these threats, yet this is not occurring. Interviews were used to gain in depth information from the user’s point of view. Data was collected from individual e-mail users from the general public, and organisations. It was found that although the literature had identified various problems with the software and process of e-mail encryption, the majority of problems identified in the interviews stemmed once again from user’s perceptions and attitudes. Use of encryption was virtually nil, although the desire to use encryption to protect privacy was strong. Remembering secure passwords was recurrently found to be problematic, so in an effort to propose a specific method of increasing their usability an empirical experiment was used to examine the memorability of passwords. Specially constructed passwords were tested for their ability to improve memorability, and therefore usability. No statistical significance in the construction patterns was found, but a memory phenomenon whereby users tend to forget their password after a specific period of non-use was discovered. The findings are discussed with reference to the fact that they all draw on a theme of responsibility to maintain good security, both from the perspective of the software developer and the end user. The term Personal Liability and General Use Evaluation (PLaGUE) is introduced to highlight the importance of considering these responsibilities and their effect on the use of security

    Antibacterial and Anti-Inflammatory pH-Responsive Tannic Acid-Carboxylated Agarose Composite Hydrogels for Wound Healing

    pH-sensitive hydrogels play an important role in controlled drug release applications and have the potential to impact the management of wounds. In this study, we report the fabrication of novel carboxylated agarose/tannic acid hydrogel scaffolds cross-linked with zinc ions for the pH-controlled release of tannic acid. The resulting hydrogels exhibited negligible release of tannic acid at neutral and alkaline pH and sustained release at acidic pH, where they also displayed maximum swelling. The hydrogels also displayed favorable antibacterial and anti-inflammatory properties, and a lack of cytotoxicity toward 3T3 fibroblast cell lines. In simulated wound assays, significantly greater cell migration and proliferation was observed for cells exposed to tannic acid hydrogel extracts. In addition, the tannic acid hydrogels were able to suppress NO production in stimulated human macrophages in a concentration-dependent manner, indicating effective anti-inflammatory activity. Taken together, the cytocompatibility, antibacterial, and anti-inflammatory characteristics of these novel pH-sensitive hydrogels make them promising candidates for wound dressings. © 2016 American Chemical Society

    Concept and development of an autonomous wearable micro-fluidic platform for real time pH sweat analysis

    In this work the development of an autonomous, robust and wearable micro-fluidic platform capable of performing on-line analysis of pH in sweat is discussed. Through the means of an optical detection system based on a surface mount light emitting diode (SMD LED) and a light photo sensor as a detector, a wearable system was achieved in which real-time monitoring of sweat pH was performed during 55 minutes of cycling activity. We have shown how through systems engineering, integrating miniaturised electrical components, and by improving the micro-fluidic chip characteristics, the wearability, reliability and performance of the micro-fluidic platform was significantly improved

[email protected] BACKGROUND: Defective pancreatic bicarbonate secretion with low intestinal pH or intestinal inflammation of any origin increase intestinal permeability in cystic fibrosis (CF). METHODS: In this open study, the authors evaluated the effect of a proton-pump inhibitor on intestinal permeability and inflammation in 14 young, pancreatic-insufficient CF patients. Permeability was measured by a three-sugar permeability test before and after 1 year of lansoprazole use, and urinary nitric oxide (NO) oxidation products were assessed before and during that year as a marker of inflammation. RESULTS: After 1 year of lansoprazole use, median urinary recovery percentages changed from 2.5% to 1.7% (P = 0.064), from 24.9% to 24.5% (no significance), and from 10.5% to 11.1% (no significance) for lactulose, mannitol, and L-rhamnose, respectively. Despite the fact that the median urinary excretion ratios decreased from 0.108 to 0.083 (P = 0.03) and from 0.246 to 0.176 (P = 0.016) for lactulose and mannitol and for lactulose and rhamnose, respectively, they both remained increased. Median urinary NO products-to-creatinine ratios were 0.287 for CF patients before lansoprazole and 0.130 for healthy control participants (P = 0.002). Although there was a tendency toward a decrease in the NO products-to-creatinine ratio during treatment, this was not significant at the end point. CONCLUSIONS: Intestinal permeability is considerably increased in CF patients and is partly corrected after the use of a proton-pump inhibitor for 1 year, which may point to a harmful effect of the acid luminal contents on the tight junctional related paracellular permeability pathway. The start and end values for the NO products-to-creatinine ratio in CF patients were not significantly different, but were considerably increased when compared with control participants (P = 0.002)

    Exploring the effect of the pH on the corrosion of multilayer nickel-chromium coatings

    The impact of the pH on the corrosion of microporous nickel-chromium coatings has been explored at localised scale by Scanning Electrochemical Microscopy and validated by potentiodynamic polarisation measurements. Results not only reveal the correlation between both techniques but also enables to identify the different corrosion rate after increasing the electrolyte aggressiveness varying the pH. The Author focuses on the role that legal certainty has played so far in this discussion and suggests that its impact on real life decisions and the functioning of the legal system has been greatly undervalued. The essay’s central argument attempts to demonstrate that, in the interpretation of general clauses and their translation into specific rules, the objective of legal certainty should be pursued and can ultimately be achieved
