No full textDie Züchtung bei Apfel Malus × domestica Borkh. ist aufgrund der langen juvenilen Phase sehr zeitaufwendig. Eine Introgression von Merkmalen in etablierte Sorten ist durch die bestehende Selbstinkompatibilät und den hohen Anteil an Heterozygotie nicht möglich. Für die Einkreuzung von Genen aus Wildarten sind deshalb komplexe Züchtungsprogramme notwendig. Ein solches Programm dauert mehrere Jahrzehnte. Gentechnische Verfahren könnten diesen Prozess beschleunigen. Im Rahmen dieser Arbeit wurden gentechnische Verfahren getestet, um (i) die Krankheitsresistenz durch transgene Ansätze zu erhöhen, (ii) alternative Verfahren zur Selektion von gv-Pflanzen für die Erzeugung cisgener Pflanzen zu testen, (iii) Gene zu identifizieren, mit deren Hilfe es möglich ist, die juvenile Phase des Apfels zu verkürzen, und (iiii) ein Fastbreeding-Verfahren mithilfe von transgenen Apfelpflanzen mit verkürzter juveniler Phase aufzubauen, mit dem es möglich ist, aufwendige Züchtungsprogramme bei Apfel in wenigen Jahren realisieren zu können. Die Ergebnisse der einzelnen Schwerpunkte werden diskutiert.Breeding of apple Malus × domestica Borkh. is time consuming based on the long juvenile phase of apple seedlings. The introgression of selected traits intoestablished cultivars is impossible by traditional breeding based on the existing selfincompatibility and the high level of heterozygosity. Complex breeding programs have to be performed for introducing genes from the wild to the cultivated apple. Such programs take normally several decades. Genetic engineering could help to speed-up the process. The present study was aimed to prove genetic engineering approaches to (i) improve the resistance of apple by using transgenic methods, (ii) to evaluate alternative strategies for the selection of genetically modified plants, which can be used for the production of cisgenic plants, (iii) to identify genes, which can be used to break the juvenile period of apple, and (iiii) to establish a Fast-breeding program, which uses transgenic early-flowering apple plants as crossbreed parents for realizing complex breeding programs within a few years. Chitinase activities, scab resistance, mycorrhization rates and biomass of own-rooted and grafted transgenic apple
No full textThis study investigated the impact of constitutively expressed Trichoderma atroviride genes encoding exochitinase nag70 or endochitinase ech42 in transgenic lines of the apple cultivar Pinova on the symbiosis with arbuscular mycorrhizal fungi (AMF). We compared the exo- and endochitinase activities of leaves and roots from non-transgenic Pinova and the transgenic lines T386 and T389. Local and systemic effects were examined using own-rooted trees and trees grafted onto rootstock M9. Scab susceptibility was also assessed in own-rooted and grafted trees. AMF root colonization was assessed microscopically in the roots of apple trees cultivated in pots with artificial substrate and inoculated with the AMF Glomus intraradices and Glomus mosseae. Own-rooted transgenic lines had significantly higher chitinase activities in their leaves and roots compared to non-transgenic Pinova. Both of the own-rooted transgenic lines showed significantly fewer symptoms of scab infection as well as significantly lower root colonization by AMF. Biomass production was significantly reduced in both own-rooted transgenic lines. Rootstock M9 influenced chitinase activities in the leaves of grafted scions. When grafted onto M9, the leaf chitinase activities of non-transgenic Pinova (M9/Pinova) and transgenic lines (M9/T386 and M9/T389) were not as different as when grown on their own roots. M9/T386 and M9/T389 were only temporarily less infected by scab than M9/Pinova. M9/T386 and M9/T389 did not differ significantly from M9/Pinova in their root chitinase activities, AMF root colonization and biomass

Intention of the treatise is to highlight the richness of his artistic activity, inspirations, ways of artistic statement, benefits and ways of directing as well as the results of a reflection on the performance of above mentioned artist.
