    Human reproduction depends on an intact hypothalamic-pituitary-gonadal axis. Hypothalamic gonadotropin-releasing hormone (GnRH) has been recognized as the central regulator of the production and release of the pituitary gonadotropins that, in turn, regulate the gonadal functions and the production of sex steroids. The peculiar development, distribution, and episodic activity of GnRH-producing neurons have solicited an interdisciplinary interest on the etiopathogenesis of several reproductive diseases. Moreover, growing knowledge on a new more complex interplay among integrative centers (hypothalamus and pituitary) and the effector (ovary) open to a new perception of the mechanisms controlling the reproductive axis

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Consensus on women's health aspects of polycystic ovary syndrome (PCOS)

    Polycystic ovary syndrome (PCOS) is the most common endocrine disorder in females with a high prevalence. The etiology of this heterogeneous condition remains obscure and its phenotype expression varies. Two, widely cited, previous ESHRE/ASRM-sponsored PCOS consensus workshops focused on diagnosis (published in 2004) and infertility management (published in 2008). The present third PCOS consensus paper summarizes current knowledge and identifies knowledge gaps regarding various women’s health aspects of PCOS. Relevant topics addressed—all dealt with in a systematic fashion—include adolescence, hirsutism and acne, contraception, menstrual cycle abnormalities, quality of life, ethnicity, pregnancy complications, long-term metabolic and cardiovascular health and finally cancer risk.     Female mammals inactivate one of their two X-chromosomes to compensate for the difference in gene-dosage with males that have just one X-chromosome. X-chromosome inactivation is initiated by the expression of the non-coding RNA Xist, which coats the X-chromosome in cis and triggers gene silencing. In early mouse development the paternal X-chromosome is initially inactivated in all cells of cleavage stage embryos (imprinted X-inactivation) followed by reactivation of the inactivated paternal X-chromosome exclusively in the epiblast precursors of blastocysts, resulting temporarily in the presence of two active X-chromosomes in this specific lineage. Shortly thereafter, epiblast cells randomly inactivate either the maternal or the paternal X-chromosome. XCI is accompanied by the accumulation of histone 3 lysine 27 trimethylation (H3K27me3) marks on the condensed X-chromosome. It is still poorly understood how XCI is regulated during early human development. Here we have investigated lineage development and the distribution of H3K27me3 foci in human embryos derived from an in-vitro model for human implantation. In this system, embryos are co-cultured on decidualized endometrial stromal cells up to day 8, which allows the culture period to be extended for an additional two days. We demonstrate that after the co-culture period, the inner cell masses have relatively high cell numbers and that the GATA4-positive hypoblast lineage and OCT4-positive epiblast cell lineage in these embryos have segregated. H3K27me3 foci were observed in ~25% of the trophectoderm cells and in ~7.5% of the hypoblast cells, but not in epiblast cells. In contrast with day 8 embryos derived from the co-cultures, foci of H3K27me3 were not observed in embryos at day 5 of development derived from regular IVF-cultures. These findings indicate that the dynamics of H3K27me3 accumulation on the X-chromosome in human development is regulated in a lineage specific fashion

While there seems to exist a group of highly educated people who benefit from transnational mobility and networks, for migrants the maintenance of transnational ties to their ‘old homes’ appears to lead to a social mobility trap, and thus to further marginalisation. Yet, the relationship between transnationality and social inequality has so far attracted little systematic exploration. This paper traces the association of transnationality with social inequalities among migrants in Germany. The discussion is led by results from US studies while these, and the few available European studies, have investigated particular groups and used selected indicators in relation to both transnationality and social inequalities. In order to contribute to a better understanding of the relationship, this paper proposes a concept of transnationality as heterogeneity that distinguishes various dimensions of transnationality. In this respect, the analysis concentrates on financial, personal, identity-based and cultural practices. As a marker of heterogeneity, transnationality may be related to the production of social inequalities, understood as differential opportunities to participate in society. These, in turn, are based on the availability of economic, cultural and social capital. Using these concepts, the association between transnationality and social inequalities will be analysed by employing data from the German Socio-Economic Panel Study (SOEP) which contains a relevant sub-sample of migrants as well as a number of transnational items. The analysis focuses on the question whether or not this association is marked by a dualistic pattern as suggested in the current debate. To this end, it investigates how the different dimensions of transnationality are related to the various forms of capital relevant for social inequalities. As it turns out, the analysis identifies no uniform pattern. To the contrary, different dimensions of transnationality are differently associated with the various forms of capital. More often than not, those persons who engage in transnational practices in the various dimensions are those who have higher levels of capital at their disposal.
