    ATM kinase dead: from ataxia telangiectasia syndrome to cancer

    ATM is one of the principal players of the DNA damage response. This protein exerts its role in DNA repair during cell cycle replication, oxidative stress, and DNA damage from endogenous events or exogenous agents. When is activated, ATM phosphorylates multiple substrates that participate in DNA repair, through its phosphoinositide 3-kinase like domain at the 3'end of the protein. The absence of ATM is the cause of a rare autosomal recessive disorder called Ataxia Telangiectasia characterized by cerebellar degeneration, telangiectasia, immunodeficiency, cancer susceptibility, and radiation sensitivity. There is a correlation between the severity of the phenotype and the mutations, depending on the residual activity of the protein. The analysis of patient mutations and mouse models revealed that the presence of inactive ATM, named ATM kinase-dead, is more cancer prone and lethal than its absence. ATM mutations fall into the whole gene sequence, and it is very difficult to predict the resulting effects, except for some frequent mutations. In this regard, is necessary to characterize the mutated protein to assess if it is stable and maintains some residual kinase activity. Moreover, the whole-genome sequencing of cancer patients with somatic or germline mutations has highlighted a high percentage of ATM mutations in the phosphoinositide 3-kinase domain, mostly in cancer cells resistant to classical therapy. The relevant differences between the complete absence of ATM and the presence of the inactive form in in vitro and in vivo models need to be explored in more detail to predict cancer predisposition of A-T patients and to discover new therapies for ATM-associated cancer cells. Founder and director of the Mountain Youth Jamboree, Hubert H. Hayes (1901-1964) auditioned and directed youth to perform in folk dance, music, and folk and ballad singing. The jamboree was held in the Asheville City Auditorium (now known as Thomas Wolfe Auditorium) from 1948 to 1973, and Hayes’ wife, Leona Trantham Hayes (1913-1989) continued to direct the program after his death in 1964.     Activation of telomerase may allow unlimited cell proliferation and immortalization, One of the telomerase protein subunits has a reverse transcriptase (hTERT) activity that is essential for telomerase Function and regulation. In human gliomas, telomerase is frequently associated with malignant tumor progression. In our study, we investigated the expression of hTERT at the cellular level in 34 primary de novo glioblastoma multiforme (GBM) by in situ hybridization (ISH). The expression oh hTERT; in tumor tissue was also assessed by Ri-PCR, In addition, telomerase activity measured by telomeric repeat amplification protocol (TRAP) and telomere length polymorphism assayed by telomere restriction fragment (TRF) Southern blot were investigated. We found that all GEM, including those with negative TRAP reaction, contained abundant amounts of cytoplasmic hTERT mRNA, interestingly, the ISH analysis revealed that the hTERT mRNA was homogeneously expressed by the whole tumor cell population in about 60% of the GBM. In the remaining cases. hTERT was absent in subsets of tumor cells. TRF analysis, which shows that both TRAP-positive and TRAP-negative de novo GBM have elongated telomeres, further supports that telomerase activity is present in all de novo GEM. Correlations with tumor size and extent of necrosis suggest that hTERT reactivation is an early event in GEM development and that telomerase activity may be lost in subpopulations of neoplastic cells during tumor progression.     Calcium Bioavailability From a Calcium-Rich Mineral Water, With Some Observations on Method

    Goals: The study was designed to determine whether high-calcium mineral water is an efficient additional source of dietary calcium, optimizing a method for calcium determination never used for mineral waters. Background: It is generally agreed that an adequate calcium intake is necessary for the acquisition of an ideal peak bone mass and for the maintenance of the bone mineral density in adults, in postmenopausal women, and in the elderly. Mineral waters are calorie free, and some, with high calcium levels, might be significant sources of calcium. Study: The availability of the calcium contained in a high-calcium mineral water was measured in 27 healthy subjects. In 8 subjects the calcium availability of the water was compared with the calcium availability ingested with milk at the same calcium load. Milk and water were labeled extrinsically with 30 mg 44Ca. Fractional absorption from the oral dose was determined from plasma samples using ICP-MS technique. Results: At an ingested calcium load of 3.18 mmol, percentage of absorption for water averaged 22.53 ± 2.53 (mean ± SD) for men, 22.57 ± 2.10 (mean ± SD) for premenopausal women and 21.62 ± 3.12 (mean ± SD) for postmenopausal women. Percentage absorption from milk was 23.15 ± 4.06 (mean ± SD). Discussion: The calcium from the mineral water is thus highly bioavailable, at least as bioavailable as milk calcium, and ICP-MS appears to represent a reliable and reproducible method for calcium absorption from alimentary sources

    This essay reconstructs several images of political concept of freedom in the modern and contemporary tradition. In the first part the Author traces the genealogy of modern political concept of freedom that produces the achievement of sovereignty and the individual rights in the age of revolutions. This political and historical process shows an image of freedom as property of the modern subject which is free because safe, autonomous and independent. In the second part, the Author introduces a different imagine of freedom in accord with the arendtian position of What is freedom? So political freedom can be sketched as an action, a new beginning, an unexpected event; and thus, this different image of freedom is showed as a possible way out from our age of crisis
