Permutation inference for a class of mixture models
In statistical surveys, people are often asked to express evaluations on several topics or to make an ordered arrangement in a list of objects (items, services, sentences, etc.); thus, the analysis of ratings and rankings is receiving a growing interest in many fields. In this framework, we develop a testing procedure for a class of mixture models with covariates (defined as cub models), proposed by Piccolo (2003) and D'Elia and Piccolo (2005) and generally developed in a parametric context. Instead, we propose a nonparametric solution to perform inference on cub models, specifically on the coefficients of the covariates. A simulation study proves that this approach is more appropriate in some specific data settings, mostly for small sample sizes
Piccolo Regulates the Dynamic Assembly of Presynaptic F-Actin
Filamentous (F)-actin is a known regulator of the synaptic vesicle (SV) cycle, with roles in SV mobilization, fusion, and endocytosis. However, the molecular pathways that regulate its dynamic assembly within presynaptic boutons remain unclear. In this study, we have used shRNA-mediated knockdown to demonstrate that Piccolo, a multidomain protein of the active zone cytomatrix, is a key regulator of presynaptic F-actin assembly. Boutons lacking Piccolo exhibit enhanced activity-dependent Synapsin1a dispersion and SV exocytosis, and reduced F-actin polymerization and CaMKII recruitment. These phenotypes are rescued by stabilizing F-actin filaments and mimicked by knocking down Profilin2, another regulator of presynaptic F-actin assembly. Importantly, we find that mice with a targeted deletion of exon 14 from thePclogene, reported to lack >95% of Piccolo, continue to express multiple Piccolo isoforms. Furthermore, neurons cultured from these mice exhibit no defects in presynaptic F-actin assembly due to the expression of these isoforms at presynaptic boutons. These data reveal that Piccolo regulates neurotransmitter release by facilitating activity-dependent F-actin assembly and the dynamic recruitment of key signaling molecules into presynaptic boutons, and highlight the need for new genetic models with which to study Piccolo loss of function.</jats:p
Teaching the Piccolo: A Survey of Selected College Flute Teachers
"The piccolo is an auxiliary member of the flute family. A diversity of opinion exists among college flute teachers as to the importance and method of piccolo study as part of a flute curriculum. This author conducted a study to outline some current pedagogical trends among college flute teachers. This document presents information gathered via electronic survey from college flute instructors at schools accredited by the National Association of Schools of Music. The survey questions pertained to background information of respondents, studio size, studio requirements, instruments and pedagogical material, and pedagogy. Landscape of Exception as Spatial and Social Interaction Between High-Quality Agricultural Production and Immigrant Labour Exploitation
This Chapter analyses the spatial and social interaction phenomena between high-quality agricultural production and immigrant labour exploita- tion that produce the landscape of exception, a particular declination of the Agambenian (2005) “state of exception” concept. The landscape of exception construction mechanism is generated within South-Eastern Sicily through the productive system of greenhouses, finalised to the vegetables production. Green- houses, in particular, represent an effective tool for spatial manipulation over the landscape and social control of migrant workers. In relation to these considerations, this work reflects on ethical challenges and dilemmas of planning, highlighting (both explicit and latent) conflicts and power inequalities in the landscapes of exception, where issues of social justice, environmental sustainability and suspension of norms are strictly intertwined
From the Metropolitan Areas to the Post-metropolitan Dimension
This chapter introduces a description of the two main areas that will be discussed throughout the book: the metropolitan region of Palermo and South-Eastern Sicily. The chapter starts with a socio-economic analysis describing the characteristics of demography, employment and income in these two parts of Sicily, by comparing them to the situation of other areas in Italy. A spatial analysis of housing and land use complements the socio-economic outlook. Each section describes the most consolidated trends in these areas, but also discusses the most innovative changes and the challenges Sicily has been facing over the years
