The silence of the administration is an institute of Administrative Law in which at the absence of a response from the public entity is assigned a meaning, usually dismissing the petition. The uruguayan and argentine administrative systems have important similarities and some differences in their regulation. In this paper, both systems are compared, pointing their differences and points in common.El silencio de la administración es un instituto del Derecho Administrativo en el que a la falta de respuesta del ente público se le asigna un significado, por lo general desestimatorio. Toward the development of user-centered neurointegrated lower limb prostheses
The last few years witnessed radical improvements in lower-limb prostheses. Researchers have presented innovative solutions to overcome the limits of the first generation of prostheses, refining specific aspects which could be implemented in future prostheses designs. Each aspect of lower-limb prostheses has been upgraded, but despite these advances, a number of deficiencies remain and the most capable limb prostheses fall far short of the capabilities of the healthy limb. This paper describes the current state of prosthesis technology; identifies a number of deficiencies across the spectrum of lower limb prosthetic components with respect to users' needs; and discusses research opportunities in design and control that would substantially improve functionality concerning each deficiency. In doing so, the authors present a roadmap of patients related issues that should be addressed in order to fulfill the vision of a next-generation, neurally-integrated, highly-functional lower limb prosthesis
Dynamic hydrogen bonding network in the distal pocket of the nitrosyl complex of Pseudomonas aeruginosa cd1 nitrite reductase
cd(1) nitrite reductase (NIR) is a key enzyme in the denitrification process that
reduces nitrite to nitric oxide (NO). It contains a specialized d(1)-heme
cofactor, found only in this class of enzymes, where the substrate, nitrite,
binds and is converted to NO. For a long time, it was believed that NO must be
released from the ferric d(1)-heme to avoid enzyme inhibition by the formation of
ferrous-nitroso complex, which was considered as a dead-end product. However,
recently an enhanced rate of NO dissociation from the ferrous form, not observed
in standard b-type hemes, has been reported and attributed to the unique
d(1)-heme structure (Rinaldo, S.; Arcovito, A.; Brunori, M.; Cutruzzolà, F. J.
Biol. Chem. 2007, 282, 14761-14767). Here, we report on a detailed study of the
spatial and electronic structure of the ferrous d(1)-heme NO complex from
Pseudomonas aeruginosa cd(1) NIR and two mutants Y10F and H369A/H327A in
solution, searching for the unique properties that are responsible for the
relatively fast release. There are three residues at the "distal" side of the
heme (Tyr(10), His(327), and His(369)), and in this work we focus on the
identification and characterization of possible H-bonds they can form with the
NO, thereby affecting the stability of the complex. For this purpose, we have
used high field pulse electron-nuclear double resonance (ENDOR) combined with
density functional theory (DFT) calculations. The DFT calculations were essential
for assigning and interpreting the ENDOR spectra in terms of geometric structure.
We have shown that the NO in the nitrosyl d(1)-heme complex of cd(1) NIR forms
H-bonds with Tyr(10) and His(369), whereas the second conserved histidine,
His(327), appears to be less involved in NO H-bonding. This is in contrast to the
crystal structure that shows that Tyr(10) is removed from the NO. We have also
observed a larger solvent accessibility to the distal pocket in the mutants as
compared to the wild-type. Moreover, it was shown that the H-bonding network
within the active site is dynamic and that a change in the protonation state of
one of the residues does affect the strength and position of the H-bonds formed
by the others. In the Y10F mutant, His(369) is closer to the NO, whereas mutation
of both distal histidines displaces Tyr(10), removing its H-bond. The
implications of the H-bonding network found in terms of the complex stability and
catalysis are discussed
Hyperexcitability and hyperplasticity disrupt cerebellar signal transfer in the IB2 KO mouse model of autism
Autism spectrum disorders (ASDs) are pervasive neurodevelopmental conditions that often involve mutations affecting synaptic mechanisms. Recently, the involvement of cerebellum in ASDs has been suggested, but the underlying functional alterations remained obscure. We investigated single-neuron and microcircuit properties in IB2 (Islet Brain-2) KO mice of either sex. The IB2 gene (chr22q13.3 terminal region) deletion occurs in virtually all cases of Phelan-McDermid syndrome, causing autistic symptoms and a severe delay in motor skill acquisition. IB2 KO granule cells showed a larger NMDA receptor-mediated current and enhanced intrinsic excitability, raising the excitatory/inhibitory balance. Furthermore, the spatial organization of granular layer responses to mossy fibers shifted from a “Mexican hat” to a “stovepipe hat” profile, with stronger excitation in the core and weaker inhibition in the surround. Finally, the size and extension of long-term synaptic plasticity were remarkably increased. These results show for the first time that hyperexcitability and hyperplasticity disrupt signal transfer in the granular layer of IB2 KO mice, supporting cerebellar involvement in the pathogenesis of ASD
Cerebellar hyper-plasticity in the IB2 KO Mouse Model of ASD
Autismspectrum disorders (ASD) are pervasive neurodevelopmental disorders
including syndromes with familial conditions. Among these, the
Phelan-McDermid syndrome is associated with the co-deletion of
SHANK3 and IB2 genes at the chromosome 22q terminus. Although
much attention has been devoted to characterize SHANK3 mutations,
very little is known about the role of IB2 in ASD. The IB2 protein is
expressed at synapses and takes part to the NMDA receptor interactome
in the postsynaptic densities. Here we further investigate the synaptic and
circuit alterations in the IB2 KO mice, that already proved to be a reliable
ASD model (Giza et al., 2010). Starting from the previously described
enhancement of NMDA receptor-mediated synaptic currents at the cerebellarmossy
fiber - granule cell synapse of IB2 KO mice, we investigated
the impact of IB2 gene deletion on excitatory/inhibitory balance and
synaptic plasticity in cerebellar cortex microcircuits. In particular, electrophysiological
experiments showed increased granule cell excitability
in IB2 KO compared to WT mice, together with a 3-fold enhanced
NMDA receptor-mediated current. Using voltage sensitive dye imaging
(VSDi), the spatial distribution of excitation (E) and inhibition (I) in the
granular layer was assessed, showing an unbalanced E/I ratio in IB2 KO,
mirrored in the distribution of the enhanced NMDA component of excitation
in IB2 KO mice.Moreover, long-term potentiation (LTP) proved to
be enhanced, while long-term depression (LTD) was reduced, in the IB2
KOcompared toWT. Interestingly, the spatial distribution in center (LTP)
- surround (LTD) structures showed alterations in IB2 KO mice (with
larger center and less deep surround) suggesting a shift from a classic
Bmexican hat^ to a Bstove-pipe^ shape. These data show impressive
cerebellar microcircuit alterations in the IB2 KO model, according to
the growing number of evidence accounting for a major role of the cerebellum
in ASD

Recently, the involvement of cerebellum in ASD has been suggested but the underlying functional alterations remained obscure. Herein, we exploited a combination of whole-cell patch-clamp recordings with voltage sensitive dye imaging (VSDi) in acute cerebellar slices in WT and IB2 KO mice to investigate single-neuron and microcircuit properties. The IB2 gene (chr22q13.3 terminal region) deletion occurs in virtually all cases of Phelan–McDermid syndrome, causing autistic symptoms and a severe delay in motor skill acquisition. The granular layer of these mice revealed severe alterations in synaptic transmission, neuronal excitation and long-term synaptic plasticity. A 2.5-times larger NMDA receptormediated current in IB2 KO granule cells enhanced synaptic plasticity (WT = 20.4 ± 4.2 %, n=12 vs. IB2 KO = 107.7 ± 44.4, n=9; p<0.05) along with the excitatory/inhibitory (E/I) balance (WT = 0.98 ± 0.27, n=6 vs. IB2 KO = 2.78 ± 0.32, n=7; p<0.01). At the same time, the spatial organization of granular layer responses to mossy fiber inputs shifted from a "Mexican hat" to a "stovepipe hat" profile, with stronger excitation in the core (WT = 12.9 ± 1.7 μm vs. IB2 KO = 29.5 ± 4.9 μm, n=5 for both; p<0.01) and limited inhibition in the surround (WT/KO ratio IWT/ KO = 2.83 ± 0.17, n=5). The IB2 KO mouse model therefore configures a complex cerebellar synaptopathy centered on NMDA receptor gain of function, that in several respects resembles alterations also observed in cortical minicolumns. The profound changes of signal processing at the cerebellar input stage unveil a possible new mechanism contributing to the pathogenesis of autistic-like behavior
Elementos del acto administrativo en las decisiones de los colegios de abogados
En el presente artículo el autor analiza las sentencias de los Tribunales de Disciplina de los Colegios Públicos de Abogados a la luz de la teoría del acto administrativo. Luego del análisis de cada elemento, concluye que cuando el acto sancionador reúne todos los elementos propios de los actos administrativos, goza de las características de presunción de legitimidad y ejecutoriedad. En cambio, la ausencia o vicio en alguno de los elementos del acto genera la inexistencia, nulidad o anulabilidad del acto en cuestión.In this article the author analyzes the judgments of the Disciplinary Courts of the Public Bar Associations in the light of the theory of the administrative act. After analyzing each element, he concludes that if the sanctioning act brings together all the elements of administrative acts, it has the characteristics of presumption of legitimacy and enforceability.
