    Evaluation of lipid coverage and high spatial resolution MALDI-imaging capabilities of oversampling combined with laser post-ionisation

    Matrix-assisted laser desorption/ionisation-mass spectrometry imaging (MALDI-MSI) is a powerful technique for visualising the spatial locations of lipids in biological tissues. However, a major challenge in interpreting the biological significance of local lipid compositions and distributions detected using MALDI-MSI is the difficulty in associating spectra with cellular lipid metabolism within the tissue. By-and-large this is due to the typically limited spatial resolution of MALDI-MSI (30-100 mu m) meaning individual spectra represent the average spectrum acquired from multiple adjacent cells, each potentially possessing a unique lipid composition and biological function. The use of oversampling is one promising approach to decrease the sampling area and improve the spatial resolution in MALDI-MSI, but it can suffer from a dramatically decreased sensitivity. In this work we overcome these challenges through the coupling of oversampling MALDI-MSI with laser post-ionisation (MALDI-2). We demonstrate the ability to acquire rich lipid spectra from pixels as small as 6 mu m, equivalent to or smaller than the size of typical mammalian cells. Coupled with an approach for automated lipid identification, it is shown that MALDI-2 combined with oversampling at 6 mu m pixel size can detect up to three times more lipids and many more lipid classes than even conventional MALDI at 20 mu m resolution in the positive-ion mode. Applying this to mouse kidney and human brain tissue containing active multiple sclerosis lesions, where 74 and 147 unique lipids are identified, respectively, the localisation of lipid signals to individual tubuli within the kidney and lipid droplets with lesion-specific macrophages is demonstrated.</p

    In this paper, arguments and evidence is presented which leads the author to conclude that the structure and imperfections of the financial markets does influence the course of economic growth. In particular, it is concluded that due to a monopoly structure in rural banking, an agricultural credit system which has helped induce a labor saving technology, and private and public flow of funds that impediments to rural development and incentives for rural out migration have been created. Although this is only one of a number of forces operating to cause the post war growth pattern, this financial structure is a key causative element and must be dealt with to initiate a successful rural development strategy.Community/Rural/Urban Development, Financial Economics,

    Procedures of rat in situ skeletal muscle contractile properties

    There are many circumstances where it is desirable to obtain the contractile response of skeletal muscle under physiological circumstances: normal circulation, intact whole muscle, at body temperature. This includes the study of contractile responses like posttetanic potentiation, staircase and fatigue. Furthermore, the consequences of disease, disuse, injury, training and drug treatment can be of interest. This video demonstrates appropriate procedures to set up and use this valuable muscle preparation. To set up this preparation, the animal must be anesthetized, and the medial gastrocnemius muscle is surgically isolated, with the origin intact. Care must be taken to maintain the blood and nerve supplies. A long section of the sciatic nerve is cleared of connective tissue, and severed proximally. All branches of the distal stump that do not innervate the medial gastrocnemius muscle are severed. The distal nerve stump is inserted into a cuff lined with stainless steel stimulating wires. The calcaneus is severed, leaving a small piece of bone still attached to the Achilles tendon. Sonometric crystals and/or electrodes for electromyography can be inserted. Immobilization by metal probes in the femur and tibia prevents movement of the muscle origin. The Achilles tendon is attached to the force transducer and the loosened skin is pulled up at the sides to form a container that is filled with warmed paraffin oil. The oil distributes heat evenly and minimizes evaporative heat loss. A heat lamp is directed on the muscle, and the muscle and rat are allowed to warm up to 37 degrees C. While it is warming, maximal voltage and optimal length can be determined. These are important initial conditions for any experiment on intact whole muscle. The experiment may include determination of standard contractile properties, like the force-frequency relationship, force-length relationship, and force-velocity relationship. With care in surgical isolation, immobilization of the origin of the muscle and alignment of the muscle-tendon unit with the force transducer, and proper data analysis, high quality measurements can be obtained with this muscle preparation.articl

    New capabilities and applications of ambient ionization mass spectrometry for the surface analysis and imaging of lipids

    Mass spectrometry is arguably the most important technique used in lipid analysis. It provides unparalleled sensitivity and molecular specificity in the detection, quantification and structure elucidation of the diverse range of lipids present in complex biological matrices. The recent and rapid emergence of ambient ionization mass spectrometry presents new and exciting opportunities for the direct detection of lipids from surfaces, including those commonly encountered in lipid research such as tissue sections, cells and thin-layer chromatography plates. This thesis describes the application, optimization and analytical capabilities for two contemporary ambient ionization technologies - namely desorption electrospray ionization (DESI) and liquid extraction surface analysis (LESA) - for the surface and structural analysis of lipids from all three of these sample/substrate classes. Herein the results of ambient ionization analysis are shown to: (i) provide new insight into the distribution (and thus function) of individual lipids within human lens tissue; (ii) monitor and exploit surface chemistry such as the oxidation of unsaturated lipids by atmospheric ozone; and (iii) obtain lipid profiles allowing for the identification of cell types, including analysis of single cells, by coupling LESA-MS with inkjet bio-printing technologies. Results from this thesis serve to highlight the great potential of ambient surface analysis mass spectrometry for lipid research

    Productivity growth and the returns from public investment in R&D in Australian broadacre agriculture

    Investment in R&D has long been regarded as an important source of productivity growth in Australian agriculture. Perhaps because research lags are long, current investment in R&D is monitored closely. Investment in R&D has been flat while productivity growth has remained strong, relative both to other sectors of the Australian economy and to the agricultural sectors of other countries. Such productivity growth, at a time when the decline in terms of trade facing Australian farmers has slowed, may have enhanced the competitiveness of Australian agriculture. The econometric results presented here suggest no evidence of a decline in the returns from research from the 15 to 40 per cent per annum range estimated by Mullen and Cox. In fact the marginal impact of research increases with research over the range of investment levels experienced from 1953 to 2000, a finding which lends support to the view that there is underinvestment in agricultural research. These results were obtained from econometric models which maintain strong assumptions about how investments in research and extension translate into changes in TFP. There are jobs you will have before you 'make it' too. Some you will enjoy. And some - not so much. On my way to becoming a teacher I was a waiter, a landscaper, a tree planter, and a bartender. Some of these jobs I liked and some I hated. But ALL of these jobs brought me to the profession I love and shaped who I have become. At W.R. Myers High School we are proud of being a school where every student can find an avenue for success and in being part of a community where everyone in the Rebel family strives to find the best in themselves and others. But very thankfully, and in true Rebel Spirit, we made the most of the situation and were able to organize and provide our student-athletes with a few opportunities to participate in sport. The Rebels made teams and practiced as much as they possibly could in football, volleyball, cross-country, golf, basketball, and curling. Unfortunately, a few of our traditional sports were not able to get off the ground in the Spring, but hopefully that will only fuel the fire of the up and coming athletes for next season! Myers Concert and Stage Bands were unable to perform this year due to COVID, but we were lucky enough to still be able to play our instruments. Vince Gassi, a composer from Toronto, was commissioned by the W.R. Myers Band Parent Association to write a piece for the Concert Band. The band students met with Mr. Gassi via Zoom in October to talk to him about the piece and the writing process. He completed the piece for us in January and we began working on it. Again the band students met with Mr. Gassi on Zoom for him to listen to us play and give feedback. But unfortunately due to the COVID restrictions, we were unable to premier the piece. Hopefully, next year we will be able to play Tomorrow Shines Soon. The Minister's Youth Council consists of 40 junior and senior high school students (in Alberta) all of which have many diverse interests, perspectives, and backgrounds. Students get the opportunity to share their perspectives on many different topics in education. Over the last couple of years students on this council have worked with the Alberta Government Education Department, MLA's, industry professionals, teachers and school boards to help better education in out province. Specifically, these students helped inform a vaping policy from Edmonton Public Schools, assisted in the creation of Bill 1 9 which aims to reduce vaping use in Alberta and helped shape the Ministerial Order on Student Learning. ^ef Keira1' created all of these masterpieces during 'Round 1' of Covid shutdown. That qualified Sayge to go to the National High School Rodeo Finals 2021 in Lincoln, Nebraska. Way to go Sayge!! Kayce Liptak has also had a great showing this year so far. She placed 2nd at Provincials, just behind Sayce Turcato. If you enjoy watching playing a high-intensity, high-contact sport, then this is a game for you! With the Lacrosse league in its infancy in ^ber (this is just the second season of play) the team did a stellar job working on basic skills, learning plays, and Alling as a team unit. They definitely had their ups and downs where the scoreboard was concerned. Our goal at W.R. Myers High School is to pro­vide experiences whether they be educational, recreational, or social which will most enrich the lives of our students. This yearbook attempts to capture and preserve some of these experiences along with the people who helped make them possible. Congratulations to Mrs. Mayer and the students in the Journalism class for all of the hard work which they expended in compiling another outstanding yearbook. Especially though, we extend our apprecia­tion to all of the staff and students who helped make the 1987/88 school year such a memorable one. Because of you, this book is possible; and because of you, we are able to relive the experiences of this past year again and again. School tends to be nostalgic as time goes by. Hopefully, the graduating class will have fond memories of W.R. Myers and the lessons learned will be of value as you progress through life. Among our activities were several fund raisers and community services. We went on a bottle drive, ran a beverage booth at the Christmas dance, held a Caveman dance, and held a wake-a-thon. For services we ran two game tables at the annual community Hallowe'en Party, held a bake sale and donated proceeds to Care and Share, and held a free car wash for the community. Unfortunately, this year the annual conven­tion which was to be held in Thunder Bay was cancelled due to the lack of registrants. However, to make up for this loss we plan­ned a trip to Riverside Waterslide for our own recreation. I would like to conclude by saying thanks to all the dedicated Key Clubbers and that I had a really good experience being the presi­dent of the best Key Club in Western Canada. Myers High School travel club toured Europe. They visited Switzerland, Germany, France, Great Britain, and Holland. Rounding off the year our Concert and Stage Bands combined to form a marching band, contributing to the suc­cess of the annual Taber Rodeo Parade on May 21st. Our thanks go to members of the Myers Music Association, a group of dedicated parents who have assisted throughout the year on trips, with the distribution, care and maintenance of our choir and band uniforms, and, without them, the organization of feeding the masses at our two annual concerts would have been a nightmare. With only four weeks practice under our belts, this year's Stage Band with 50% new members began the season by performing for Premier Getty and members of the Alberta Legislature in September. Other performances this year in­cluded the opening ceremonies for AgExpo '88 in Lethbridge and the Taber Trades Fair at the Agriplex. Over and above the annual Fall and Spring concerts, the Raymond and Spruce Grove exchanges, and a concert for the Grade 5 students at Central, the Stage Band, unknown to Mr. Reid prepared and performed in the farewell supper to Mr. and Mrs. Reid at the Heritage Inn in June. The choir grew in strength this year providing the opportunity once again for three-part harmony: soprano, alto and baritone. Concerts included the Fall and Spring Concerts and the band exchanges. Throughout the Christmas season, the choir performed at Clearview Lodge, Harmony Home, Homestead Manor, and the Taber Hospital. Our contribution to the Kiwanis Carol Festival the L.D.S. Two years ago we were a very new and raw squad whose knowledge and skill level, in this widely spreading sport, was very limited. Today, I'm proud to say, this is not the case. Through hard-work, dedication and commitment our team has excelled as a winning team in the sport. At zone competition, which we hosted, we placed second which entitled us to proceed on to Edmonton to Provincials. This was our first Provincial Competition and I'm proud of our department and efforts while representing our school. As coach of the team, I would like to take this opportunity to express my sincere gratitude to all the girls for their pa­tience, hard-work, enthusiasm and most of all participation on this team. You as individuals have not only grown and improved physically, mentally and emotionally but you have also helped me to learn and grow as well. I had never coached girls before and I learned several valuable lessons: (1) My ego was jolted firmly back into place. (2) I was forced to coach from grass roots for the first time in years. (3) Girls are more emotional. (4) Girls can be tough. (5) Girls are athletic in their own right. I think the girls on the volleyball team learned a lot as well. (1) They learned about discipline. (2) They learned about the work ethic. (3) They learned about commitment. (4) They learned about how to win. 24 Candee Jensen, Lindy Andrus, Karen Steeves, Erin Anderson, Larita Johnson, Sina Platt, and Dee Dee Rombough. (5) They learned about muscles. As a team we started in the cellar and progressed slowly upward. We couldn't beat our way out of a paper bag in September. By November we placed 2nd in the League Post Season Tournament and the McCoy Invita­tional, and capped it all off by winning zones for the first time in 5 years. Pro­vincials were anti climactic for us, but all in all the year turned out to be a good one. 1 thank the girls for their commitment to a valuable ideal. We had fun! The girls were extremely intense while they played. This was a reflec­tion of Mr. Stewart's intense coaching. Each player improved greatly during this rebuilding year, to the point that Myers gave Medicine Hat an extremely close game at the end of the season. Graduating from this year's team are Tracey Baker, Julie Evanson, and Deanna Grant, all of whom will be missed next year. However, with eight returning players Myers is certainly the team to beat next year. Heather Chipman lead the team in scoring with Tracey Baker a close second. Julie Evanson was the leading rebounder and best defensive player. Julie receiv­ed an honorable mention all star at the 4A Girls League meeting. Dana Eldridge was the most improved player, making tremendous improvements in her re­bounding and defense. Tracey Atwood, Deanna Grant and Carolyn Jensen all became good ball handlers and effectively handled presses. The remaining players, Andrea Jensen, Marcy Sebok, and Jackie Cline all became quality subs that actually benefitted the team when they entered the game. Unfortunately, Karen Steeves was plagued with a knee injury that kept her out of action most of the year. BOYS SENIOR TEAM This year's Rebel basketball team was a talented and aggressive team. The record was 24 wins, 13 losses. The team won the St. Mary's Warriors tourney, the MHHS tournament, and finished third at the L.C.I. tournament. The team was led by Grade 12 centre, Larry Johnson. Brad Krizan also had an outstanding season. Grade 12 graduating players are Monte Layton, Cam Francis, Jim Brown, and Larry Johnson. These guys gave their hearts and 100% effort to the program and will be missed. Thanks to my stats: Michelle Fawns, Carla Anderson, and Mandy Shewchuck. Myers students were awesome. We averaged about 500 fans per game and their noise level and enthusiasm was superb. Thanks to the cheerleaders as well - you were great! I hope you will look back on this year with fond memories. I'll treasure this year as I hope you will. J.V. BASKETBALL 1987-88 This year's J.V. Basketball team started off slowly but ended as a contender. The team worked hard and made notable progress in individual, and team play throughout the season. They brought home to Myers the championship trophy from the Medicine Hat Junior Varsity Basketball Tournament; and, by mid­season, the Junior Rebels were competing for a top four-spot in the league. The team had many games that were decided by three or less points, which always made the games exciting to watch. Of these games, the Junior Rebels had a one-point loss to the eventual league tournament champions, L.C.I.; and against the eventual league tournament champions, Medicine Hat, the team suffered losses of only one and two points in regular season play. J would like to thank the wonderful office staff for their countless hours of proofing student names and filling us in on activities. Thank you to all staff for putting up with our "sneaky" candid picture snapping throughout the school year and last but not least Lisa Mstalos for her moments of getting every one back in the game in her "own" crazy way. The year started out with a yearbook Conference in Lethbridge. Ct was a long two days, which set the basis for the next nine months. Only a few of us could attend but by the end of the year you couldn't tell because everyone had mastered the yeartech On-line Program! Our theme this year was music and songs. Our very diverse class presented us with a very diverse mix of music throughout the book, mainly using the titles rather than the entire song. .As Editor of the 2007 yearbook <7 was quite pleased to find a wonderful array of staff and talent. For the first semester we had a rather large class but we made it work and everyone's hard work payed off. Flundreds of hours later we are showcasing an amazing book/ filled with memories talents and dreams of students come and gone.
