The Fiction and the Aesthetic Principles of Flannery O’Connor: A Study in Theory and Practice
Flannery O'Connor, a recent Southern writer, gives evidence in her fiction and in her statements on aesthetics that she has a message vital to modern man. This message, a faith-oriented interpretation of the meaning of man, is conveyed through three basic concepts: Reality, Redemption, and Shook. An understanding of Miss O'Connor's work must focus around this Southern writer's perspective on these key concepts and their treatment in her works. It is the purpose of this paper to clarify and to explain the meaning of these concepts and to show how they function in her work in order that Miss O'Connor's intuition1 of Reality, Redemption, and Shock may stand revealed. CD4+ natural regulatory T Cells prevent experimental cerebral malaria via CTLA-4 when expanded in vivo
Studies in malaria patients indicate that higher frequencies of peripheral blood CD4(+) Foxp3(+) CD25(+) regulatory T (Treg) cells correlate with increased blood parasitemia. This observation implies that Treg cells impair pathogen clearance and thus may be detrimental to the host during infection. In C57BL/6 mice infected with Plasmodium berghei ANKA, depletion of Foxp3(+) cells did not improve parasite control or disease outcome. In contrast, elevating frequencies of natural Treg cells in vivo using IL-2/anti-IL-2 complexes resulted in complete protection against severe disease. This protection was entirely dependent upon Foxp3(+) cells and resulted in lower parasite biomass, impaired antigen-specific CD4(+) T and CD8(+) T cell responses that would normally promote parasite tissue sequestration in this model, and reduced recruitment of conventional T cells to the brain. Furthermore, Foxp3(+) cell-mediated protection was dependent upon CTLA-4 but not IL-10. These data show that T cell-mediated parasite tissue sequestration can be reduced by regulatory T cells in a mouse model of malaria, thereby limiting malaria-induced immune pathology
Activation of human NK cells by Plasmodium-infected red blood cells.
This chapter describes a protocol to assess activation of human NK cells following in vitro stimulation with malaria-infected red blood cells. Activation is assessed by flow cytometry, staining for cell surface expression of CD69 and accumulation of intracellular IFN-γ. Procedures are described for in vitro propagation and purification of Plasmodium falciparum parasites, separation of peripheral blood mononuclear cells from heparinized blood by density centrifugation, in vitro culture of PBMC and for staining and analysis of PBMC by flow cytometry. From Bradford Moor to Silver Dale. The life, work, and legacy of W. Riley, 1866-1961
This thesis presents the first full account of the life and achievements of Bradford-born W. Riley (1866-1961), once internationally known as a popular and prolific Yorkshire author.
Before becoming a famous writer, he was Managing Director of the successful Bradford Optical Lantern Company, Riley Brothers and was also, for 75 years, a Methodist local preacher and an important layman within northern Methodism. He wrote 39 books, published many stories and articles, and was a busy lecturer. Riley located most of his 30 novels in the Yorkshire Dales and has left a legacy of vivid portraits of people and places in the dales that he knew and loved.
This biography of Riley draws upon material never seen hitherto, expanding upon the author's diffident autobiography.
The complete bibliography of his extensive writings includes much new and long-lost material.
In presenting Riley to a new generation, this account places him in context with his contemporaries.
Riley proclaimed his Christianity sympathetically and attractively to his receptive public in much of his output. This thesis includes an insight into the spiritual life, outlook and thinking of a popular and much-respected committed and active Methodist local preacher.
Riley's life story is the account of a remarkably successful, self-motivated Victorian. He was a household name in his time, both in Yorkshire and internationally. Immunopathogenesis of human schistosomiasis
Schistosomiasis continues to be a significant cause of parasitic morbidity and mortality worldwide. This review considers the basic features of the pathology and clinical outcomes of hepatointestinal and genitourinary schistosomiasis, presents an overview of the numerous studies on animal models that have clarified many of the immunopathological features, and provides insight into our current understanding of the immunopathogenesis and genetic control of human schistosomiasis. In murine schistosomiasis, pathology is induced by a CD4+ Th2 driven granulomatous response directed against schistosome eggs lodged in the host liver. The Th2 cytokines IL-4 and IL-13 drive this response, whereas IL-10, IL13Rα2, IFN-γ and a subset of regulatory T-cells act to limit schistosome induced pathology. A variety of cell types including hepatic stellate cells, alternatively activated macrophages and regulatory T-cells have also been implicated in the pathogenesis of schistosomiasis. Current knowledge suggests the immunopathogenic mechanisms underlying human schistosomiasis are likely to be similar. The review also considers the future development of anti-pathology schistosome vaccines. As fibrosis is an important feature of many other diseases such as Crohn's disease and sarcoidosis, a comprehensive understanding of the cellular and molecular mechanisms involved in schistosomiasis may also ultimately contribute to the development an effective disease intervention strategy for other granulofibrotic diseases

Yet James Weldon Johnson, the Negro author of the Bamboo Tree lyrics, has been referred to as an esthetic leader of his race, and has been several times compared, as a poet, to John M. Synge. |James Weldon Johnson was born in Jacksonville, Florida, in 1871. Educated at Atlanta University and at Columbia, he spent several years after his graduation collaborating with his brother Rosamund in the writing of librettos and songs for the musical comedy stage. He served at one time as principal of the Jacksonville Colored High School. Alter practicing law for many years he was sent as United States Consul to Venezuela and Nicaragua, where he served for seven, years. Dynamic Imaging of CD8(+) T cells and dendritic cells during infection with Toxoplasma gondii.
To better understand the initiation of CD8(+) T cell responses during infection, the primary response to the intracellular parasite Toxoplasma gondii was characterized using 2-photon microscopy combined with an experimental system that allowed visualization of dendritic cells (DCs) and parasite specific CD8(+) T cells. Infection with T. gondii induced localization of both these populations to the sub-capsular/interfollicular region of the draining lymph node and DCs were required for the expansion of the T cells. Consistent with current models, in the presence of cognate antigen, the average velocity of CD8(+) T cells decreased. Unexpectedly, infection also resulted in modulation of the behavior of non-parasite specific T cells. This TCR-independent process correlated with the re-modeling of the lymph node micro-architecture and changes in expression of CCL21 and CCL3. Infection also resulted in sustained interactions between the DCs and CD8(+) T cells that were visualized only in the presence of cognate antigen and were limited to an early phase in the response. Infected DCs were rare within the lymph node during this time frame; however, DCs presenting the cognate antigen were detected. Together, these data provide novel insights into the earliest interaction between DCs and CD8(+) T cells and suggest that cross presentation by bystander DCs rather than infected DCs is an important route of antigen presentation during toxoplasmosis

Detecting intestinal ischemia using near infrared spectroscopy
Blood supply to the intestine can suddenly be interrupted. Acute mesenteric intestinal ischemia often requires invasive surgery to restore blood supply to the intestine. Early correction of vascular insufficiency is the most important factor in improving patient survival when confronted with acute mesenteric intestinal ischemia. A prolonged loss of blood flow results in irreversible damage to the intestine that can lead to death. It is also imperative that dead segments of the intestines be removed. Several subjective criteria are relied upon to differentiate viable from non-viable tissue, unfortunately, these criteria can lead to an inaccurate assessment. A porcine model of intestinal ischemia was used to determine the efficacy of using near infrared (NIR) spectroscopy to find ischemic segments of the intestine and detect the onset of reperfusion following resolution of vascular occlusion. Nine segments of intestine were identified and six were assigned to three treatment groups; (1) segments undergoing no vascular manipulations, (2) segments undergoing arterial/venous occlusion and (3) segments undergoing arterial/venous occlusion followed by reperfusion. The remaining segments were used as spacers and interposed between each of the ischemia segments. A classification model, using partial least square discriminant analysis, was built on the spectra collected from the segments with no vascular manipulations and the segments that were solely subjected to arterial/venous occlusion. The spectra collected from the intestinal segments that experienced both occlusion and reperfusion were used to test the classification model. The model was able to detect and distinguish ischemic intestinal tissue with a specificity and sensitivity exceeding 80% with an overall classification accuracy of 89%.
