Performance of gravel aggregates in superpave mixes with 100/95 angularity
The current aggregate requirement for aggregate angularity that the NYSDOT requires for their Superpave Hot Mix Asphalt (HMA) is 100/98. With this requirement, many of the gravel quarries in New York State cannot meet that specification. As a result, quarries that are further away from the job site have to transport the aggregates which in turn increases the cost of construction. This paper investigates a multitude of test methods to determine whether the NYSDOT can further lower their Superpave aggregate angularity requirement. Epigenetic regulation and transcription factor programming enhances neurogenesis in neural stem cells
In this thesis, we questioned how neuronal and glial phenotypes become specialized. Epigenetic chromatin modifiers and transcription factors were investigated on their roles in programming and maintaining neural lineage restriction. A relatively homogeneous population of cells was generated by deriving immortalizing neural clones from embryonic rat forebrains. Three phenotypes; neuronal, glial and multipotential (GE6, GE2, CTX8), provided contrasting lineages to probe the factors responsible for shaping cell fate. One particular clone, GE6, differentiated into a functional inhibitory like interneuron. Gene expression analysis showed several genes such as Ascl1, Dlx1, Dlx5, may be responsible for the interneuronal phenotype. Epigenetic regulation through histone modifications is believed to be an essential component within the developing nervous system, ultimately affecting cell fate. Testing chromatin signatures on specific neural genes with permissive and repressive histone “marks” shows that chromatin state in undifferentiated precursors correlates with current and predicts downstream gene expression. These results suggest that cell fate may already be predetermined. Furthermore, ChIP sequencing reveals global differences between the representative clones. Extrinsic growth factors, such as BMP2 promotes the neuronal and glial phenotypes in the multipotential cell CTX8. BMP2 asserts its phenotypic response in part by regulating global acetylation enrichment in specific neural gene networks, providing a mechanism to promote and maintain cell fate. Directly altering chromatin marks using a histone deacetylase inhibitor, valproic acid (VPA), globally acetylates the chromatin of CTX8 cells and enhances neurogenesis. VPA treatment was found to also maintain or increase acetylation in specific neuronal genes, such as Ascl1. In addition, several microRNAs thought to play a role in neurogenesis were also epigenetically regulated after VPA treatment. Finally, through the combination of gene expression and epigenetic analyses, direct programming through exogenous expression of Ascl1, Dlx1 and Dlx5 enhanced neurogenesis in CTX8. Gene expression and epigenetic signature mapping provides us with a deeper understanding of how lineage restriction occurs. Learning the programming rules will assist in directing homogeneous populations of neuronal cells to further probe the mechanisms of neurodegenerative diseases.Ph. Heritability and Linkage Analysis of Appendicitis Utilizing Age at Onset
Appendicitis usually afflicts the young, but there is a large tail in the distribution of onset age. The genetics of this disease are still not well understood. A heritability analysis and genome wide linkage analysis of a large twin dataset was undertaken. Treating age of onset of appendicitis as a censored survival trait revealed a heritability of 0.21, and found evidence of linkage to Chromosome 1p37.3. Higher U (1)-gerbe connections in geometric prequantization
We promote geometric prequantization to higher geometry (higher stacks), where a prequantization is given by a higher principal connection (a higher gerbe with connection). We show fairly generally how there is canonically a tower of higher gauge groupoids and Courant groupoids assigned to a higher prequantization, and establish the corresponding Atiyah sequence as an integrated Kostant-Souriau infinity-group extension of higher Hamiltonian symplectomorphisms by higher quantomorphisms. We also exhibit the infinity-group cocycle which classifies this extension and discuss how its restrictions along Hamiltonian infinity-actions yield higher Heisenberg cocycles. In the special case of higher differential geometry over smooth manifolds, we find the L-infinity-algebra extension of Hamiltonian vector fields - which is the higher Poisson bracket of local observables - and show that it is equivalent to the construction proposed by the second author in n-plectic geometry. Finally, we indicate a list of examples of applications of higher prequantization in the extended geometric quantization of local quantum field theories and specifically in string geometry
Common greenbrier (Smilax rotundifolia L.) as a model for understanding fungal community organization in the phyllosphere
Fungi asymptomatically infect all terrestrial vegetation, but the structure and assembly of these fungal communities are poorly understood. Smilax rotundifolia, a common woody vine of the northeastern United States, was used as a model to study endophytic (internal colonizing) and epiphytic (surface colonizing) fungal communities, from the perspectives of niche-based influences, spatial variation, and evolutionary relationships. Wild greenbrier plants were sampled in New Jersey (USA) in late winter at a single site, and again in mid-summer, in a multi-site, multi-tissue, sampling effort. Fungal communities of the plant surface and interior were made up of mostly different species. Correlative relationships were found between some fungal species' abundances, but none were seen between species that were strongly restricted to the surface and those strongly restricted to the interior. The summer, multi-site study, revealed that the strongest factor determining fungal community composition was surface vs. interior habitat, followed by tissue/organ type, and lastly general geographic location. The effect of season was also studied by comparing the single-site winter dataset against the data from the same site sampled in summer. Season had a major influence on some fungal species but not others, similar to influence of tissue type. Also, in a very novel finding, it was found that certain endophytes showed statistically different abundances, depending on the distance from the stem base at which a sample was taken. The optimum height differed for different fungal species. At the scale of a single stand of plants, it was found that epiphytic, but not endophytic, samples showed a significant correlation between spatial proximity and fungal community similarity. Finally, the phylogenetic relationships were studied between congeneric fungal species that were common on the plant. Two-gene phylogenies were constructed using Smilax-derived isolates, along with downloaded sequences of well-defined species in the same genera. These congeneric species were found to be only distantly related (i.e., they were widely separated within the known phylogenies of their genus). The final chapter is a literature review, bringing insight from the present dissertation research to identify important unanswered questions. Exploiting Knowledge across Networks through Reputation Management.
The emerging paradigm of network competition is increasingly in evidence across many industrial sectors and provides further support for the idea that ‘supply chains compete, not companies’. It can be argued that network competition requires a much greater focus on managing the interfaces that connect the individual players in that network and exchanging and leveraging knowledge across the network. This paper sets out to establish a framework whereby the critical interfaces and the knowledge sharing benefits can be identified and how the strength of the relationships at those interfaces can become the basis for building organisational reputation and create an environment more conducive to co-operation and knowledge sharing. Finally, the paper analyses the potential impact of reputational risks in influencing the perception of stakeholders about the organisation.
Whilst the idea of value-adding networks based on closely connected providers of capabilities and resources is appealing, it should be recognised that, if not properly managed, the actions of the stakeholders in those networks can impact the risk profile of the business significantly—particularly reputational risk. The more that organisations become part of complex global networks, the more dependent they become upon the other network members for knowledge and other resources. Because of this dependency there is always the danger that the reputation of the focal firm can be damaged by the actions of other network members, hence reducing the likelihood of future collaborative working and knowledge exploitation.
Using examples drawn from a variety of industries, the paper highlights the potential for reputational risk if the critical network interfaces are not closely managed. It will be argued that by actively managing relationships with stakeholders in the network the risk to the organisation's reputation can be mitigated and the sharing of knowledge simultaneously enhanced
