TACC3-ch-TOG track the growing tips of microtubules independently of clathrin and Aurora-A phosphorylation
No full textThe interaction between TACC3 (transforming acidic coiled coil protein 3) and the microtubule polymerase ch-TOG (colonic, hepatic tumor overexpressed gene) is evolutionarily conserved. Loading of TACC3–ch-TOG onto spindle microtubules requires the phosphorylation of TACC3 by Aurora-A kinase and the subsequent interaction of TACC3 with clathrin to form a microtubule binding surface. Whether there is a pool of TACC3–ch-TOG that is independent of clathrin in human cells, and what is the function of this pool, are open questions. Here, we report that TACC3 is recruited to the plus-ends of microtubules by its association with ch-TOG and that this pool is independent of phosphorylation and binding to clathrin. The plus-end binding of TACC3–ch-TOG persists in interphase and we propose that one cellular function of TACC3–ch-TOG is to modulate cell migration. We also describe the distinct subcellular pools of TACC3, ch-TOG and clathrin. TACC3 is often described as a centrosomal protein, but we show that there is no significant population of TACC3 at centrosomes. The delineation of distinct protein pools reveals a simplified view of how these proteins are organized and controlled by post-translational modification

Translation technique and textual studies in the Old Greek and Theodotion versions of Daniel.
No full textThis thesis focuses on two separate, but related areas: the analysis of translation technique and the Greek texts of Daniel. Foremost in the research of Translation Technique (TT) in the Septuagint is the need for a model that is appropriate for the analysis of different ancient languages. In recent years there has been an increasing emphasis on the features of literalism in a translation, but it is argued in this thesis that the focus on literalism is inadequate as a methodology for the analysis of TT. The contention of this thesis is that the analysis of TT should incorporate insights from modem linguistic research. Therefore, the main purpose of this thesis is to develop and apply such a model to the Old Greek (CG) and Theodotion (Th)versions of Daniel. The existence of two complete Greek versions of the book of Daniel that are closely related to the same Vorlage (at least in chapters 1-3 and 7-12), furnish ideal examples for the application of the methodology. Unfortunately, it is no straightforward matter to employ the OG of Daniel, because the available critical edition can no longer be regarded as reliable. The most important witness to the OG version of Daniel is Papyrus 967, and large portions of this manuscript have been published since the appearance of the critical edition of the OG of Daniel in 1954. Therefore, in order to analyze and compare the two Greek texts of Daniel, it is necessary to evaluate all of the variants of Papyrus 967 in order to establish a preliminary critical text of OG. Once a critical text is established the proposed methodology for translation technique is applied to selected passages in the OG and Th versions of Daniel. An analysis and comparison of TT in OG and Th makes it possible to: 1) characterize the TT employed by OG and Th in detail; 2) determine Th's relationship to OG, i.e. is it a revision or independent translation; 3) demonstrate how the Greek texts can be employed effectively for textual criticism of the Hebrew Bible. On the basis of the analysis of Th's text it is also possible to determine Th's relationship to the body of works, which exhibit a close formal correspondence to the Masoretic text, known as Kaige-Theodotion
Beyond translation technique: linguistic approaches to the Septuagint’s translation of the Aramaic verbal system in Old Greek Daniel, Theodotion Daniel, and 2 Esdras
No full textThis thesis applies four distinct linguistic methods to the philological data mined from translation technique of the Aramaic verbal system in Old Greek Daniel, Theodotion Daniel, and 2 Esdras. After briefly setting the present work within the context of previous studies (ch. 1), an exhaustive catalogue of the Aramaic verbal forms and their Greek translations occur, followed by the Greek forms and the Aramaic forms they translate (ch. 2). Chapter 3 applies linguistic typology and typological studies of tense, aspect, and modality (TAM) to the enigmatic Aramaic prefix conjugation (PC). Chapter 4 uses the concepts of language contact to explore Aramaic dialect differences (both lexical and grammatical) between the Aramaic of the Bible and the spoken Aramaic of the translators. Chapter 5 applies the concepts of idiolect as developed in forensic linguistics to Greek verbal syntax in the pluses of Old Greek Daniel, which in turn informs our understanding of the textual problem that is Old Greek Daniel 4-6. Chapter 6 problematizes minority translation equivalents in terms of Relevance Theory, showing how translators’ use aspectual choice functions as a procedural constraint to guide their readers toward intended inferences. The closing chapter (ch. 7) reviews the conclusions of the work and suggests new avenues for research
