Expression of a gene-encoded FtsZ-based minimal machinery to drive synthetic cell division
No full textThe Christophe Danelon lab is involved in the long-term effort to construct an autonomous minimal cell using a bottom-up approach. Our goal is to achieve self-maintenance, selfreproduction, and evolution of a liposome compartment containing a minimal genome and a cell-free gene expression system. Self-reproduction requires splitting of the mother compartment into two daughter cells. The project described in this dissertation is part of the group’s attempts to create a minimal division unit for the synthetic cell. The development of a gene-driven, controllable, content-preserving liposome division strategy is an ongoing challenging task. Here, we reconstituted some of the organizational mechanisms for division of Escherichia coli in a cell-free system. In E. coli, cytokinesis is mediated by a multiprotein complex that forms a contractile ring-like structure at the division site. The ring is composed of the cytosolic filament-forming protein FtsZ, as well as its membrane anchoring proteins FtsA and ZipA. The Min system assists in the ring localization at mid-cell by oscillating from pole to pole. Using liposomes as a synthetic compartment and PURE system for cell-free gene expression, we reconstituted membrane-bound cytoskeletal structures and oscillating gradients of Min proteins for liposome constriction and dynamic organization of FtsZ filaments.BN/Christophe Danelon La

No full textRomeo e Giulietta, Renzo e Lucia: due coppie letterarie tanto diverse quanto emblematiche che l’autore mette a confronto muovendo dalla constatazione che Manzoni, pur ammirando e ben conoscendo il teatro shakespeariano, non riprende nel romanzo spunti apprezzabili dalla tragedia ‘veronese’. Nella costruzione narrativa dei Promessi sposi, infatti, l’amour-passion è volutamente omesso dallo scrittore milanese che intende presentare un modello di cristiana serenità matrimoniale, radicalmente diverso da quello erotico sentimentale shakespeariano. L’analisi comparativa delle due coppie, tuttavia, rivela inattesi rovesciamenti e singolari analogie che portano a considerare, secondo un’originale angolatura, la presunta latitanza del Romeo and Juliet nei Promessi sposi.Romeo and Juliet, Renzo and Lucia: two literary pairs so different as emblematic that the author compares moving from the observation that Manzoni, while admiring and having good knowledge of shakespearean theatre, does not draw in the novel appreciable ideas from the ‘veronese’ tragedy. In the narrative construction of the Promessi sposi, in fact, l’amour-passion is intentionally omitted because the writer intends to give a model of christian conjugal serenity, radically different from the shakespearean erotic sentimental one. The comparative analysis of two pairs, however, shows unexpected reversals and peculiar analogies that lead to consider, according to an original angle, the supposed absence of Romeo and Juliet in the Promessi sposi

Roadmap to Building a Cell: An Evolutionary Approach
No full textLaboratory synthesis of an elementary biological cell from isolated components may aid in understanding of the fundamental principles of life and will provide a platform for a range of bioengineering and medical applications. In essence, building a cell consists in the integration of cellular modules into system’s level functionalities satisfying a definition of life. To achieve this goal, we propose in this perspective to undertake a semi-rational, system’s level evolutionary approach. The strategy would require iterative cycles of genetic integration of functional modules, diversification of hereditary information, compartmentalized gene expression, selection/screening, and possibly, assistance from open-ended evolution. We explore the underlying challenges to each of these steps and discuss possible solutions toward the bottom-up construction of an artificial living cell.BN/Christophe Danelon La
