COS-7-based model: methodological approach to study John Cunningham virus replication cycle
John Cunningham virus (JCV) is a human neurotropic polyomavirus whose replication in the Central Nervous System
(SNC) induces the fatal demyelinating disease, progressive multifocal leukoencephalopathy (PML). JCV propagation and
PML investigation have been severely hampered by the lack of an animal model and cell culture systems to propagate
JCV have been very limited in their availability and robustness. We previously confirmed that JCV CY strain efficiently
replicated in COS-7 cells as demonstrated by the progressive increase of viral load by quantitative PCR (Q-PCR) during
the time of transfection and that archetypal regulatory structure was maintained, although two characteristic point
mutations were detected during the viral cycle. This short report is an important extension of our previous efforts in
defining our reliable model culture system able to support a productive JCV infection.
Supernatants collected from transfected cells have been used to infect freshly seeded COS-7 cell line. An infectious viral
progeny was obtained as confirmed by Western blot and immunofluorescence assay. During infection, the archetype
regulatory region was conserved.
Importantly, in this study we developed an improved culture system to obtain a large scale production of JC virus in
order to study the genetic features, the biology and the pathogenic mechanisms of JC virus that induce PML
The Use of Strategic Metaphors in Intercultural Business Communication
This paper contends that the use of strategic metaphors can help deliver the effective intercultural business communication necessary for global success. Using the Renault-Nissan Alliance as an example, the authors argue that an appropriate metaphor can help provide the global glue which captures the essence of the organisation’s activities, encapsulates its strategic intent, incorporates the national and global cultures, and portrays its ethical and business stance. Indeed, as is the case in the Renault-Nissan Alliance, the appropriate use of metaphor allowed the firm to bind a diverse group of stakeholders to a common goal by using the inherent ambiguity and multiplicity of meaning of the metaphor to overcome Asian and Western intercultural differences and at the same time maximise goal congruence.intercultural business communication, strategic metaphors, alliance relationships
IV. Leucaena leucocephala CV. CUNNINGHAM
Nutritive value of Leucaena leucocephala cv. Cunningham foliage was determined during the wet and dry seasons without fertilization nor irrigation. Evaluation was made using castrated wether allocated in individual metabolism cages, using the total fecal collection method. Forage was offered ad libitum (115% of the voluntary intake from the previous day). DM content reached values of 32,1 and 29,4 % while DM digestibility was about 61,3 and 56,9 % for the wet and dry season, respectively. CP content was about 24,9 and 17,8 and CP digestibility was about 68,6 and 60,6 %. OMD had values of 62,5 and 58,9 %, ME content was about 9,23 and 8,56 Mj/kg of DM and DM intake was of 57,5 and 54,2 g/kg P,75 for the wet and dry season, respectively. Nutritive value results indicated that this variety of Leucaena has a suitable potential for ruminant nutrition during both seasons of the year.
Caracterización de la comunidad vegetal en una asociación de gramíneas mejoradas y Leucaena leucocephala cv.: Cunningham Characterization of the plant community in an association of improved grasses and Leucaena leucocephala cv.: Cunningham
In a dairy of the Genetic Enterprise of Matanzas, a work was carried out for five years, with the objective of characterizing the performance of the plant community in an association of improved grasses and Leucaena leucocephala cv Cunningham under commercial conditions. The botanical composition of the pastureland, tree density, dry matter availability and DM, CP, Ca and P contents of the species present in the system were determined. The botanical composition of the pastureland was characterized by a percentage of improved pastures higher than 50%, with predominance of Cynodon nlemfuensis cv. Jamaicano and Panicum maximum cv. Likoni; while leucaena maintained plant density, as an adaptation to the conditions of the system. The total dry matter availability was higher than 3 t/ha/rotation during the experimental stage. It is concluded that in the association of improved grasses and L. leucocephala cv. Cunningham there was high dry matter availability and persistence of the grassland during the five years of evaluation, with values of CP in the improved grasses of 9,6-9,8% and in leucaena of 25%, applying neither irrigation nor chemical fertilizers, which allowed to attenuate the differences between the rainy and dry seasons
Entanglement and quantity in quantum space - About quantum measurement (II)
As a continuation and extension of "quantity in phase space" "quantity in quantum space" is introduced. With that, the disappearing of quantum interference discussed in a previous paper [S. Durr, et al., Nature 395 (1998) 33] is explained in the same spirit as our recent papers [Ren De-Ming, Commun. Theor. Phys.
Sneutrino DM in the NMSSM with inverse seesaw mechanism
In supersymmetric theories like the Next-to-Minimal Supersymmetric Standard Model (NMSSM), the lightest neutralino with bino or singlino as its dominant component is customarily taken as dark matter (DM) candidate. Since light Higgsinos favored by naturalness can strength the couplings of the DM and thus enhance the DM-nucleon scattering rate, the tension between naturalness and DM direct detection results becomes more and more acute with the improved experimental sensitivity. In this work, we extend the NMSSM by inverse seesaw mechanism to generate neutrino mass, and show that in certain parameter space the lightest sneutrino may act as a viable DM candidate, i.e. it can annihilate by multi-channels to get correct relic density and meanwhile satisfy all experimental constraints. The most striking feature of the extension is that the DM-nucleon scattering rate can be naturally below its current experimental bounds regardless of the higgsino mass, and hence it alleviates the tension between naturalness and DM experiments. Other interesting features include that the Higgs phenomenology becomes much richer than that of the original NMSSM due to the relaxed constraints from DM physics and also due to the presence of extra neutrinos, and that the signatures of sparticles at colliders are quite different from those with neutralino as DM candidate.National Natural Science Foundation of China (NNSFC) [11575053]SCI(E)ARTICLE1
