    Evidence for microtubule-based organelle transport in the pollen tube

    The role of microtubules in organelle movement during pollen tube growth is still unclear, although microtubule motor proteins have been identified. To understand the specific role of microtubules in supporting the movement of organelles, we have recently started two related research lines. The aim is to identify and characterise functionally novel microtubule motors and to verify whether microtubules can effectively support the movement of pollen tube organelles. Collected evidences suggest that pollen tube microtubules have the proper protein machinery to move organelles and that microtubules support the translocation of pollen tube organelles under in vitro conditions. These data re-evaluate the role of microtubules in organelle movement and indicate that microtubules contribute to the translocation of organelles in the pollen tube

    Dynein related polypeptides during pollen tube growth

    Nicotiana tabacum pollen tubes contain two high molecular weight polypeptides (about 400 kDa) which are specifically expressed during pollen germination and pollen tube growth. The high molecular weight doublet resembles the dynein heavy chains in some biochemical properties. Sedimentation profiles of pollen tube extracts show that the high molecular weight bands have sedimentation coefficients of 22 S and 12 S respectively. ATPase assay of sedimentation fractions shows an activity ten times higher when stimulated by the presence of bovine brain microtubules in fractions containing the 22 S high molecular weight polypeptide. Both these high molecular weight polypeptides can bind microtubules in an ATP-dependent fashion. A mouse antiserum (Dy-l) to a synthetic peptide reproducing the sequence of the most conserved ATP-binding site among dynein heavy chains recognized the two high molecular weight polypeptides. Therefore these polypeptides have epitopes immunologically related to the ATP binding sites of dynein heavy chains. immunofluorescence studies using the Dy-l antibody showed punctate structures, with a characteristic distribution pattern depending on the tube length. Biochemical evidence confirmed the presence of DHC related bands in the pollen tube membrane fraction

    The ancientness of viticulture and the mythology of wine have given the grapevine a specially favorable and beneficial position among cultivated fruit plants.It is thought that cultivation of the grapevine (Vitis vinifera L.) began during the Neolithic era (6000–5000 BC) in the region known as Transcaucasia corresponding to the eastern shores of the Black Sea. However archeological finds of grape seeds indicate that either the grapevine or at least its progenitor, Vitis sylvestris, was already distributed throughout much of Europe during the Atlantic and Sub-Boreal paleoclimatic peri-ods (7500–2500 years ago). Quite recently archeological grape seeds, dating according to radiocarbon method, between 4350 and 3950 years, were discovered in Spain (Walker 1985)
