    Not just for beer: evaluation of spent hops (Humulus lupulus L.) as a source of ecofriendly repellents for insect pests of stored foods

    Spent hops is a waste produced in large amount by the brewing industry. Rhyzopertha dominica and Sitophilus granarius are insects that cause important economic losses of stored foods. In this study, for the first time, spent hops has been evaluated as a source of essential oil (EO) and chemicals with repellent activity against R. dominica and S. granarius. Spent hops EO yield was 0.11 %. The terpenes myrcene, a-humulene, and b-caryophyllene were its main components (47 %). Spent hops EO RD50 values were 0.01 and 0.19 lL cm-2 for R. dominica and S. granarius, respectively. Among the chemicals, myrcene was able to exert the highest repellency against R. dominica (RD50 = 0.27 lM cm-2), while limonene was the most effective compound against S. granarius (RD50 = 0.89 lM cm-2). These results indicate spent hops as an excellent source of EO and chemicals to be utilized as low-cost, eco-friendly insect pests repellents in the protection of stored food

    Aedes albopictus (Diptera Culicidae), volgarmente detta “zanzara tigre asiatica”, si è diffusa dal Sud-Est asiatico in tutto il mondo attraverso il commercio dei copertoni usati, sfruttando la capacità delle sue uova di resistere alla siccità e al freddo. Anche in Italia l'arrivo della specie ha seguito la stessa dinamica. I problemi economico-sanitari riferibili alla specie sono dovuti alla sua attività ecto-parassitaria ed alla trasmissione di numerosi arbovirus e parassiti. Poiché non sono disponibili né vaccini né farmaci contro i virus tramessi da Ae. albopictus, il controllo del vettore resta lo strumento chiave per la prevenzione. Questo tuttavia non risulta di facile gestione a causa della peculiare biologia della zanzara. Le femmine depongono le uova sul bordo di piccoli contenitori scuri contenenti acqua pulita e questo rende la specie tipica degli ambienti fortemente antropizzati dove abbondano le piccole raccolte di acqua in proprietà private. In tale contesto, la lotta larvicida risulta più difficoltosa rispetto a quella condotta contro le larve delle zanzare autoctone che preferiscono deporre sulle grandi raccolte di acqua stagnante. Per questi motivi, il controllo larvale classico (mediante l'uso di organofosforici, regolatori di crescita e Bt) per Ae. albopictus non risulta praticabile. Recentemente, numerosi sforzi sono effettuati nella ricerca di composti naturali efficaci e a basso impatto ambientale. Fra questi si collocano i derivati del neem (Azadirachta indica A. Juss, Meliaceae), in particolare l’olio di neem e il neem-cake, ovvero un sottoprodotto derivante dal processo di estrazione. La presente ricerca ha preso in considerazione (i) l’efficacia larvicida di oli di neem di diversa provenienza e di estratti di neem-cake in vari solventi organici (etanolo, butanolo, etilacetato) e acqua (ii) la deterrenza all’ovideposizione delle stesse sostanze in prove di campo. I risultati hanno indicato che: (i) le frazioni esanica, metanolica, in etilacetato e acquosa di neem-cake hanno indotto, rispetto al controllo, una mortalità dell’80% delle larve di prima età sottoposte a test. Inoltre gli oli di neem di diversa provenienza testati, al pari delle loro frazioni in etilacetato, esplicano, nei confronti di larve di quarta età buona efficacia (LC50 tra 142.28 e 209.73 ppm). Scarsa efficacia ha invece mostrato la frazione butanolica degli stessi; (ii) le frazioni esanica, metanolica e in etilacetato di neem-cake esplicano, rispetto al controllo, buona deterrenza all’ovideposizione anche a basse dosi (a 100 ppm rispettivamente 71.33, 88.59 and 73.49 % di deterrenza) mentre la frazione butanolica e acquosa non sono così efficaci (a 100 ppm rispettivamente 22.72 e 17.06% di deterrenza). In conclusione quindi il neem-cake può essere proposto come insetticida da utilizzare in contesti urbani in quanto efficace, eco-friendly e a basso costo per la lotta a Ae. albopictu

    Hop, Humulus lupulus L., is an aromatic plant largely utilized in the brewing industry to add flavour and bitterness to beer. As a consequence, a large amount of residual material, named “spent hops”, is produced by the brewing industry. Since only about 15% of the hop constituents end up in the beer, spent hops should be further valorised as a low-cost source of bioactive substances. In this study, hops and spent hops were evaluated as a source of essential oil (EO) with repellent activity against two of the main stored food insect pests. Hops and spent hops EO yields were about 2.4% and 0.1%, respectively. The main components of both the EOs were the terpenes myrcene, α-humulene, and β-caryophyllene. The main chemical difference between hops and spent hops was the myrcene percentage. Area preference method bioassays showed that spent hops EO exerted a strong repellency against the lesser grain borer, Rhyzopertha dominica (F.) (Bostrichidae) and the granary weevil, Sitophilus granarius (L.) (Curculionidae). RD50 values indicated that R. dominica was about 20 time more susceptible to spent hops EO than S. granarius. Among the EO components, myrcene was the strongest repellent against R. dominica while limonene was the most effective compound against S. granarius. However, overall, for its much higher content, myrcene can be considered the compound that mostly contributes to the repellent activity of spent hops EO. These results show that spent hops could be an excellent, virtually costless, source of EO to be utilized as eco-friendly repellent in the protection of stored food from insect pests

    Les plantes aromatiques sécrétrices d'huile essentiale (HE) sont présentées comme une potentielle alternative aux actuels insecticides de synthéses, présents sur le marché, et qui sont à l'origin des dégats écologique majeurs sur la faune, la santé humaine ainsi que sur l'environment. Dans un optique écologique combinée à la valorisation de (Foeniculum vulgare Mill.), récoltées en Kabylie (Algérie) ainsi que de son composé majoritaire, contre deux ravageurs des denrées alimentaires stockées, Sitophilus zeamais et Tribolium confusum. à ces sujet, des bio-essais de répulsivité ont été menés avec deux méthodes différentes et complémentaires. L'une, suivant Tapndojou et al (2005), etudié le seul effect répulsif de l'HE en Petri tandis que l'autre, en pitfall, (germinara et al. 2007) étudie l'effect répulsif du composé majoritaire en présence de l'aliment. L'HE de F.vulgare a été isolée par hydrodistillation en Clevenger modifiée. Les tests avec l'HE, montrent que le taux de répulsivité dépend de l'éspece étudié, de la dose appliquée ainsi que du temps d'exposition. Avec les bio-essais en Petri, à la dose de 1.5% d'HE, on a obtenu 92% et 86% de répulsivité pour T. confusum et S. zeamais respectivement. Les tests en pitfall indiquent que l'effect de l'HE n'est pas du au methyl-chavicol seul, en dépit de son caractére majoritaire, et que le blend est a priori responsable de l'activité répulsive de l'HE. Les resultats obtenus laissent pensere que l'utilisation de l'HE de fenouil pourrait etre donc envisagées comme une persperctive intéressante pour la protection des denrées stockées contre les ravageurs

    Rhyzopertha dominica, Sitophilus zeamais and Tribolium confusum are three of the major food-stuff pests who cause important economic losses of shelved products with special reference to pasta. Due to the long pasta shelf life, insects can easily penetrate into the packaging with consequences that can be economically severe. Eco-friendly strategies to prevent insect attacks to the final packaged product are highly foreseen by pasta companies and essential oils, extracted from aromatic plants, could represent a valid alternative to chemical repellents. In this study, we evaluated the repellent activity of Pistacia lentiscus essential oil (PEO) and its main chemical components by two different bioassay with and without the presence of pasta. Results showed that the whole PEO exerts a broad-range aspecific repellency among the target pests with RD 50 values ranging from 0.010 to 0.037 μL cm −2 . On the contrary, the repellence of PEO components resulted to vary depending on the compound and on the pest species. Among the PEO chemical components, relative median potency analyses indicated that β-caryophyllene was able to exert the highest repellency rates against S. zeamais (RD 50 0.046 μM cm −2 ). The comparison between the two bioassays, with and without pasta, indicated that the two methodologies gave consistent results. Overall, our research firstly showed that, because of their effectiveness as repellents, PEO and its major constituents could represent valid and safe tools against pasta pests

    Prevalence of the microsporidian Nosema ceranae in honeybee (Apis mellifera) apiaries in Central Italy

    Nosema ceranae and Nosema apis are microsporidia which play an important role in the epidemiology of honeybee microsporidiosis worldwide. Nosemiasis reduces honeybee population size and causes significant losses in honey production. To the best of our knowledge, limited information is available about the prevalence of nosemiasis in Italy. In this research, we determined the occurrence of Nosema infection in Central Italy. Thirty-eight seemingly healthy apiaries (2 to 4 hives each) were randomly selected and screened from April to September 2014 (n= 11) or from May to September 2015 (n= 27). The apiaries were located in six areas of Central Italy, including Lucca (n= 11), Massa Carrara (n= 9), Pisa (n= 9), Leghorn (n= 7), Florence (n= 1), and Prato (n= 1) provinces. Light microscopy was carried out according to current OIE recommendations to screen the presence of microsporidiosis in adult worker honeybees. Since the morphological characteristics of N. ceranae and N. apis spores are similar and can hardly be distinguished by optical microscopy, all samples were also screened by multiplex polymerase chain reaction (M-PCR) assay based on 16S rRNA-gene-targeted species-specific primers to differentiate N. ceranae from N. apis. Furthermore, PCR-positive samples were also sequenced to confirm the species of amplified Nosema DNA. Notably, Nosema spores were detected in samples from 24 out of 38 (63.2%, 95% CI: 47.8–78.5%) apiaries. Positivity rates in single provinces were 10/11, 8/9, 3/9, 1/7, or 1/1 (n= 2). A full agreement (Cohen’s Kappa = 1) was assessed between microscopy and M-PCR. Based on M-PCR and DNA sequencing results, only N. ceranae was found. Overall, our results highlighted that N. ceranae infection occurs frequently in the cohort of honeybee populations that was examined despite the lack of clinical signs. These findings suggest that colony disease outbreaks might result from environmental factors that lead to higher susceptibility of honeybees to this microsporidian

    Who is polluting Italian honey? A filth test survey

    Honey is largely considered a natural and healthy product. Investigation on foreign matter in honey provides useful information to evaluate honey quality standards in apiary, honey extraction and packaging phases. Indeed, toxic substances (antibiotics, pesticides and heavy metals) as well as foreign matter (e.g. arthropods, hairs) can contaminate honey, acting as allergens or vectors of human pathogens. We used the filth test to evaluate the presence of foreign matter in samples of Italian honeys, mainly from Tuscany. We revealed a high number of carbon particles and other inorganic fragments, followed by fragments of animal origin. This latter included whole small insects, their cuticular fragments, mites and mammal hairs. The kind of contamination allowed us to evidence uncorrected apicultural practices and to suggest the appropriate corrective measures. The filth test method is an excellent and cheap tool to check honey quality, requiring minimal instrumental equipment and giving results that can be interpreted quickly

    Bioactivity of Cinnamomum verum essential oil against Varroa destructor, parasitic mite of Apis mellifera

    The honeybee, Apis mellifera (Hymenoptera Apidae) has a great economic and ecological importance. In fact, beside in the beehive products, A. mellifera plays a major role in the pollination of wild angiosperm and crops species, essential for ecosystem functioning and human food production. Varroa destructor (Acari Mesostigmata Varroidae), an ectoparasitic mite, is the most devastating parasite of the Western honeybees and causes substantial losses of colonies worldwide. Currently, the control of this pest mainly relies on chemicals such as oxalic acid, pyrethroids, and organophosphates. However, the extensive and repeated use of acaricides have caused the development of resistance in V. destructor populations, causing a reduction of their efficacy and are toxic to bees. Moreover, these products may leave residues in the hive, contaminating honey and beeswax. For these reasons, the use of alternative strategies for the control of V. destructor are required. Essential oils of aromatic plant are known to have repellent, insecticidal and acaricidal properties [1] and could represent a very valuable low-cost, environmental-friendly alternative to synthetic products [2]. Cinnamomum verum (Lauraceae) J. Presl is an evergreen tropical tree originated in Sri Lanka, traditionally widely used to flavour foods. C. verum bark yields an essential oil containing cinnamaldehyde and eugenol as main constituents that showed important biological activities, such as peripheral vasodilatory, antitumor, antifungal, cytotoxic and antimutagenic as well as insecticidal and insect repellent activities. In this work, cinnamon bark essential oil was extracted, chemically analysed and tested to evaluate its toxic/repellent activity against V. destructor in both laboratory and apiary. Results of laboratory tests showed that C. verum EO is able to cause, at the concentration of 2%, 24h after the treatment, the detachment of 50% of the mites present on adult bees without causing significant mortality of the bees. In the field, C. verum EO was able to reduce V. destructor population up to about 70%. This study indicated that C. verum could be a promising repellent against V. destructor used as an alternative to chemical treatment in integrated pest management strategies in bee colonies

    Gli oli essenziali (EO) di piante aromatiche sono considerati tra le alternative più promettenti ai repellenti e agli insetticidi di sintesi per la protezione delle derrate alimentari dagli attacchi degli insetti dannosi. Tuttavia, uno dei maggiori limiti al loro utilizzo è che l’efficacia varia non solo in base alla specie di insetto contro la quale vengono usati ma anche in relazione ad una serie di fattori quali: varietà, luogo di coltura della pianta, periodo di raccolta ecc. Tale variabilità è ritenuta dipendere dalle singole sostanze attive che li compongono e che contribuiscono all’azione tossica o repellente complessiva dell’olio essenziale. Dal punto di vista applicativo è infatti molto utile una valutazione dell’efficacia degli oli essenziali sulla base della loro composizione chimica. Tuttavia, sebbene sia generalmente ritenuto che il contributo delle singole componenti degli oli essenziali sia di tipo additivo alcune evidenze indicano che possa essere anche di tipo sinergico o antagonista, rendendo difficilmente prevedibile l’efficacia di un olio. Il finocchio (Foeniculum vulgare Mill.) è una pianta aromatica, coltivata in tutto il mondo, ben nota per le sue proprietà antimicrobiche, acaricide, farmacologiche e antiossidanti che presenta diversi chemotipi distinguibili sulla base del contenuto relativo nell’OE di due sostanze: (E)-anetolo ed estragolo. Diversi studi hanno, inoltre, dimostrato la sua efficacia come insetticida e repellente contro gli insetti. In questo studio è stata valutata la repellenza di EO estratti da acheni di due diversi chemotipi, anethole- e estragole-type, di F. vulgare, contro Rhyzopertha dominica, Sitophilus zeamais e Tribolium confusum, tre dei principali insetti dannosi dei cereali in post-raccolta. Oltre agli EO, è stata saggiata anche la repellenza dei loro principali componenti chimici: (E)-anetolo, estragolo, limonene e fencone ed è stato valutato l'effetto di co-repellenza di (E)-anetolo ed estragolo. I saggi biologici hanno indicato che l’estragole-type è il chemotipo più repellente contro le tre specie d’insetti. Coerentemente, l’analisi della potenza mediana relativa ha mostrato che l'estragolo è significativamente più repellente dell’(E)-anetolo. Il calcolo del coefficiente di co-repellenza (CRC) indica che l’effetto congiunto di (E)-anetolo ed estragolo negli OE di finocchio è di tipo sinergico. I risultati ottenuti indicano in conclusione che l'EO di finocchio “estragole-type” è promettente per lo sviluppo di repellenti a basso costo ed ecocompatibili per la protezione in post-raccolta delle granaglie e sottolineano l'importanza di una standardizzazione degli oli essenziali basata sulla loro bioattività oltre che sulla composizione chimica
