    Mgat5, a regulator of metabolic homeostasis in mice

    Golgi beta1,6N-acetylglucosaminyltransferase V (Mgat5) produces beta1,6GIcNAc-branched N-glycans on glycoproteins, which increases their affinity for galectins and opposes loss from cell surface to constitutive endocytosis. Oncogenic transformation increases Mgat5 expression, and enhances sensitivities to growth factors, cell motility and tumour metastasis. Therefore, I explored growth and tissue renewal of Mgat5-/- mice, and found that they are hypoglycaemic, resistant to weight-gain, hypersensitive to fasting, display increased oxidative respiration, reduced fecundity and decreased serum leptin. These phenotypes are similar to that of calorie restricted (CR) rodents; however, unlike CR animals, Mgat5-/- mice have accelerated loss of muscle and bone mass and reduced lifespan. CR-induced longevity involve downregulation of the PI3K/Akt/TOR pathway, downstream of many growth factor receptors including EGF, PDGF and IR. In this regard, Mgat5-/- primary MEFs and macrophages are suppressed in MAPK activation in response to EGF, PDGF, IGF, and FCS. However, muscle satellite cells and bone marrow osteoblasts revealed a FCS-induced signalling imbalance, favouring Smad2/3 activation over Erk, which may contribute to reduced growth, self-renewal and longevity of the Mgat5-/- animals. Pten opposes PI3K/Akt signalling and is a potent tumour suppressor. Mgat5 +/- Pten+/- and Mgat5-/- Pten +/- mutant mice showed a small increase in lifespan compared with Pten+/- mice, due to an apparent delay in tumour progression. Primary Pten+/-Mgat5-/- MEFs are normalised for Akt activation and cell spreading, and Pten+/- MEFs have enhanced L-PHA staining, suggesting that Mgat5 structures are regulated by P13K signalling. Aged CR rodents exhibit decreased oxidative damage, implicating either decreased ROS production or elevated antioxidant activity. I demonstrated that Mgat5-/- primary MEFs have decreased glucose uptake and mitochondrial ROS. Unlike WT control, PyMT Mgat5-/- cells fail to down regulate glucose transport, protein synthesis, ROS and activation of Erk and Akt following serum withdrawal. PyMT transformation reveals that Mgat5-deficiency not only impairs positive growth signalling, but also suppresses negative growth regulation, allowing for proliferation and tumourigenicity. Mgat5 participates upstream of PyMT signalling through N-glycan modification of growth factor receptors, but also downstream of Akt signalling through its dependence on glucose uptake for supply of UDP-GlcNAc. Therefore, Mgat5 enhances cellular signalling and growth, and metabolism and longevity in the animal.Ph.D

    Investigating the suitability of customised playlists for childbirth in Ireland and Hong Kong

    The aim of this study was to explore the experience of two couples using customised playlists to support childbirth at the public maternity hospitals in Ireland and in Hong Kong. Two couples participated in a pre-delivery meeting with a music therapist one month before their infant’s due birth date. During this meeting, the couples were assisted in setting up customised playlists and received recommendations on strategies and relaxation techniques to use with the playlists. Data collection was performed through semi-structured interviews with the participants two weeks after the childbirth. The interviews were then examined through Thematic Content Analysis (Braun & Clark, 2006). Three final themes arose included: (1) feasibility of using customised playlists during childbirth; (2) preferred music selection; and (3) perceived benefits of the playlists. The feasibility of using playlists was affected by the adaptability of the playlists to the changeable birth situations and the uncertain feedback and restriction from hospital staff. The suitability of original music selection and the meaning of songs were highlighted. Participants reported that the birthing playlists promoted relaxation and better sleep; provided spiritual support; and diverted attention from discomfort and disturbances

    Crazy Rich Asians是2018年广受瞩目的美国浪漫爱情喜剧片之一。它突破了以往的格局,成为首部全亚裔阵容的好莱坞电影。这部改编自美籍华裔作家关凯文的同名小说的电影不仅在北美区创下近十年的新高,小说的销售量也达到数百万册,至今已译入二十多种语言。小说描写了美籍亚裔女生朱瑞秋与新加坡最富有家族的继承人杨尼克的爱情故事,从而勾勒出亚洲上流社会人士的生活面貌与形象,同时也向世界展现了独特的新加坡文化。小说中所呈现的新加坡社会课题发人深省,但更值得关注的是,这是一部跨国、跨语言、跨文化及跨媒介的文学作品,牵涉了无数个文本相互交织在一起,使之成为互文理论下极佳的研究对象。有鉴于此,本论文主要以互文性的理论框架为切入点,探讨小说与作者、原文读者、译者、译文读者以及社会外在环境等因素之间的多重互文关系,从而进入小说的翻译研究。 Crazy Rich Asians was one of the most highly anticipated Hollywood romantic comedy films in 2018. It gathered overwhelming expectations before its premiere as the world awaited the first major studio film which featured an all-Asian cast, breaking through long-held stereotypes in the Hollywood scene. The movie, adapted from the Singaporean-American author Kevin Kwan’s best-selling novel, became the highest-grossing romantic comedy in the United States in the recent decade. The novel, with 1.5 million copies sold in 2018, has also been translated into over 20 different languages worldwide. The story follows an Asian-American woman, Rachel Chu, as she accompanies her boyfriend, Nicholas Young, to his childhood best friend's wedding in Singapore, where she discovered that he came from the wealthiest family in Singapore. The novel sheds light on the luxurious and extravagant lifestyles of the extremely rich in Asia - particularly in Singapore - while portraying its unique culture, of which both aspects are foreign to the West. The novel spans across various countries, languages, cultures and mediums, producing a rich source text intertwined with other literary and non-literary texts. Thus, it is significant and relevant to the analysis of intertextuality in translation studies. Using the intertextuality theoretical framework, this paper attempts to study the intertextual relationships between the text, author, translator, readers of both source and target languages and the culture at large, before exploring how these relationships influenced the translation processes by both the author and translator.Bachelor of Arts in Chines

    Στόχος της παρούσας μελέτης ήταν η διερεύνηση της εμπειρίας δύο ζευγαριών που χρησιμοποίησαν προσαρμοσμένες λίστες αναπαραγωγής [playlists] για την υποστήριξη του τοκετού σε δημόσια μαιευτήρια στην Ιρλανδία και στο Χονγκ Κονγκ. Τα δύο ζευγάρια συμμετείχαν σε μία συνάντηση με τη μουσικοθεραπεύτρια ένα μήνα πριν από την ημερομηνία γέννησης του βρέφους τους. Κατά τη διάρκεια αυτής της συνάντησης, τα ζευγάρια βοηθήθηκαν στη δημιουργία εξατομικευμένων λιστών τραγουδιών και τους προτάθηκαν στρατηγικές και τεχνικές χαλάρωσης τις οποίες μπορούσαν να χρησιμοποιούν με αυτές τις λίστες. Η συλλογή δεδομένων έγινε μέσω ημι-δομημένων συνεντεύξεων με τους συμμετέχοντες, δύο εβδομάδες μετά τον τοκετό. Στη συνέχεια οι συνεντεύξεις αναλύθηκαν βάσει της Θεματικής Ανάλυσης Περιεχομένου (Braun & Clarke, 2006). Οι τρεις τελικές κατηγορίες που προέκυψαν περιλαμβάνουν (1) την εφικτότητα χρήσης προσαρμοσμένων λιστών αναπαραγωγής κατά τον τοκετό, (2) την προτιμώμενη επιλογή μουσικής, και (3) τα αντιλαμβανόμενα οφέλη από τη χρήση των λιστών. Η εφικτότητα της χρήσης των λιστών αναπαραγωγής επηρεάστηκε από την προσαρμοστικότητα των λιστών στις μεταβαλλόμενες συνθήκες της γέννας και στην αβέβαιη ανατροφοδότηση και τον περιορισμό από το προσωπικό του νοσοκομείου. Επισημάνθηκε η καταλληλότητα των αρχικών μουσικών επιλογών και το νόημα των τραγουδιών.     Translation of humour in literature

    The translation of humour has always posed a challenge to translators as they translate the various forms of humour that exist in text, as the ability to discern what is humourous in the culture of the target language would require an understanding of the said culture. With a focus on the translation of exaggeration and puns as the chosen type of humour portrayal, this paper sets out the discuss the challenges faced by translators when they are translating humour written by the author, taking into account the cultural references in the source text that is native to the society of the source language, and the strategies to be applied when translating said cultural references. The choice of strategy employed in the translation of the extract in this paper is the theory of functional equivalence, which according to Newmark (1988), is replacing a cultural word or reference in the source language with its equivalent in the target language, with the usage of transcription or “borrowing”, which according to Harvey (2000) is a reproduction or transliteration of the original term in certain cases where the cultural reference in the humour is understood by the readers of the translated text. In conclusion, the understanding of culture, whether that of the culture of the source text, or the culture that is being translated into, is important in the translation of humour. It is important for the translator to understand the culture of the society portrayed in the source text as that is the first step to recognising the humour portrayed by the author of the original text. The understanding of the culture of the society of the target reader is important as well as the translator would need to identify and have knowledge of what kind of humour is acceptable in the target reader’s society, as well as to know boundaries of what topics might be taboo or forbidden by the authorities, and to take note not to cross these boundaries.Master of Art

    Inspired by local author Danielle Lim’s English language biography of her uncle’s struggle with schizophrenia and her mother’s painful journey as both his and their mother’s caregiver, this capstone project explores translation strategies involved in presenting the complexities of the little-known Schizophrenia, as well as in converting Singlish and dialects to the Chinese language. Taking a functionalist perspective of translation, the Chinese rendition of The Sound of Sch: A Mental Breakdown, A Life Journey hopes to raise public awareness about mental illnesses in its target text readers, and to reduce social stigmatisation of the mentally ill in that process. The translation process involves managing the author’s intent, the function of the target text, textual coherence and fidelity, as well as achieving a balance between foreignisation and domestication. Through comparing the local version with that of a Taiwanese translator, differences in the strategies and perspectives involved will also be identified and discussed, shedding light on how best to create a target text that is both faithful to the source text and yet not without the translator’s unique individuality.     A Pilot Study of Maternal Songwriting to Support Bonding During Pregnancy: “I Could Feel Her Rise and Fall With My Breath While Singing”

    IntroductionAttachment is a lasting psychological and emotional bond that impacts a child's social, emotional, and cognitive development. Maternal-fetal attachment is a precursor to this bond that is regarded as a predictor for mother-infant relating postbirth. Although previous studies have investigated use of music interventions to support maternal-fetal attachment, no study to date has investigated the use of maternal songwriting to support this bond during pregnancy.MethodsFour pregnant women were recruited to this pilot study. Each woman participated in 2 songwriting workshops with the aim of writing an original song to their fetus. Following the workshops, experiences of using songwriting for the purposes of supporting bonding were investigated through individual semistructured interviews and analyzed using thematic content analysis.ResultsAnalysis of semistructured interviews with pregnant women revealed 3 main themes: song singing promotes physical connection with the fetus; songwriting encourages open communication with the fetus; and songwriting can create a musical legacy within families.DiscussionTo the best of our knowledge, this study is the first of its kind that investigated maternal songwriting to support bonding during pregnancy. Findings indicated that songwriting has the potential to support such attachment due to its capacity to enhance physical connection and open communication with the fetus, while also creating a musical legacy that can be used and passed on among other family members. Overall, songwriting may support pregnant women to draw on their inner resources to create such a bespoke musical gift to their unborn child. Further research around this topic is warranted

The release of her latest creative fiction works The Liu Woman and The Yang Woman, both centered on “Crimes of Passion” proves to be a pivotal change in her writing style. Male authors tend to defame and at times, demonize, women in their writing, reflecting the unfair treatment of women in a patriarchal society. As a female author, Zhang’s first novel exudes a strong sense of feminism; I can relate her work to Hélène Cixous’ “Écriture Féminine”. She applies the deconstruction theory in her analysis of female criminals and their fates, in a bid to redefine women and lend a voice to the once muted women in the history of literature.Bachelor of Art

    &apos;Hong Kong Story&apos; has always been an euphemism of &apos;Hong Kong&apos;s history&apos; in enunciating the characteristic of local history. In the colonization era, Hong Kong history was indeed a colonial history because started from the very early period, the historical archives were written by either the missionaries or aristocracies, which laden with dominant European discourse.The changes in the political and cultural environment because of the 1997 Handover had invoked a blatant identity searching process among Hong Kong writers. After Hong Kong has entered into the post-transitional period, local writers started to re-investigate the history of Hong Kong. The reappropriation of history is the common theme shared by Shi Shu Ching&apos;s Hong Kong Trilogy and Dung Kai Cheung&apos;s &apos;The Rise and Fall of Wing Sing Street&apos; and The Atlas: The Archeology of an Imaginary City.The thesis argues that through constant re-narrating Hong Kong history in writing Hong Kong story, not only points to a recognized need in Hong Kong culture to search for a new identity, a new self-consciousness that can claim a different history than what has been instituted in the past colonial discourse and the appropriation of the PRC national discourse at the turn of 1997, but also gives the writer themselves a chance to construct a differentiated and unique identity; because retrieving history in writing fictions is the indispensable for any conception of self or subjectivity.</p
