Conditioning of hiPSC-derived cardiomyocytes using surface topography obtained with high throughput technology
No full textSurface functionalization of polymers aims to introduce novel properties that favor bioactive responses. We have investigated the possibility of surface functionalization of polyethylene terephthalate (PET) sheets by the combination of laser ablation with hot embossing and the application of such techniques in the field of stem cell research. We investigated the response of human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) to topography in the low micrometer range. HiPSC-CMs are expected to offer new therapeutic tools for myocardial replacement or regeneration after an infarct or other causes of cardiac tissue loss. However, hiPSC-CMs are phenotypically immature compared to myocytes in the adult myocardium, hampering their clinical application. We aimed to develop and test a high-throughput technique for surface structuring that would improve hiPSC-CMs structural maturation. We used laser ablation with a ps-laser source in combination with nanoimprint lithography to fabricate large areas of homogeneous micron- to submicron line-like pattern with a spatial period of 3 µm on the PET surface. We evaluated cell morphology, alignment, sarcomeric myofibrils assembly, and calcium transients to evaluate phenotypic changes associated with culturing hiPSC-CMs on functionalized PET. Surface functionalization through hot embossing was able to generate, at low cost, low micrometer features on the PET surface that influenced the hiPSC-CMs phenotype, suggesting improved structural and functional maturation. This technique may be relevant for high-throughput technologies that require conditioning of hiPSC-CMs and may be useful for the production of these cells for drug screening and disease modeling applications with lower costs.Fil: Cortella, Lucas R. No full textConselho Nacional de Pesquisa e Desenvolvimento Científico e Tecnológico - CNPqThis paper proposes a technical-pedagogic approach on the music dynamics played on the trumpet by using four pieces by the Brazilian composer Matheus Bitondi for solo trumpet in chamber music - duet setting. The research, study and practice of music expression tools can aid on the performance of a piece. The present compositions were selected through a bibliographical research finding excerpts where dynamic changes occur. Later, trumpet exercises from published trumpet methods were suggested in addition to other exercises
created and adapted by the author. The discussed dynamic variations were classified and the excerpts were recorded and made available through QR code enabling the reader to listen and see the recording’s sound wave patterns. In addition to the proposals and recordings this research presents a historical content on the music dynamics for trumpet in the Portuguese
language, a synthesis of the research done on the topic and provides the score for the four works by Bitondi.Este trabalho propõe uma abordagem técnico-pedagógica relacionada à dinâmica no trompete, utilizando-se de quatro obras do compositor brasileiro Matheus Bitondi (1979-), para trompete solista em música de câmara – formação duo. Nota-se que a pesquisa, estudo e prática dessa ferramenta da expressividade podem auxiliar na performance de uma obra musical. Por meio de uma pesquisa bibliográfica, foi realizada a seleção das obras, dos trechos os quais ocorrem as variações de intensidade e, posteriormente, foram sugeridos exercícios encontrados em publicações para trompete, adicionando-se outros que foram elaborados e adaptados pelo autor. As variações de dinâmicas discutidas foram classificadas e os trechos gravados e disponibilizados para escuta por meio de QR code, que possibilita ao leitor, ouvir e visualizar o espectro da gravação. Além das propostas e gravações, esta pesquisa apresenta um conteúdo histórico sobre a dinâmica no trompete em língua portuguesa, uma síntese das pesquisas sobre a dinâmica no trompete no Brasil e disponibiliza as partituras de quatro obras de Bitondi

Jornalismo Participativo, Letramento Midiático e Mediações: a manifestação de competências midiáticas em jovens universitários do ABC paulista
No full textDocumentos apresentados no âmbito do reconhecimento de graus e diplomas estrangeirosEsta dissertação se propõe a analisar como as competências midiáticas se manifestam em jovens universitários de 18 a 25 anos, participantes ou não de conteúdos jornalísticos. Para tal, utilizamos uma metodologia qualitativa, que constitui um questionário para seleção de perfis – seguindo o critério da heterogeneidade – e entrevistas semiestruturadas. A análise vai ao encontro dos objetivos da dissertação, problematizando os tipos de participação no jornalismo, levando em consideração a circulação e disseminação de conteúdo. Resgata-se as origens do conceito de jornalismo participativo, enquadrando-o a partir da formação de um novo ecossistema midiático na contemporaneidade, como advogam Anderson, Bell e Shirky. Apresenta-se o letramento midiático e a aquisição de competências como uma das saídas para que o indivíduo seja mais ativo e participativo em conteúdos jornalísticos, assim como Celot e Pérez-Tornero. Entretanto, nossa abordagem também inclui a Teoria das Mediações de Jesus Martin-Barbero no processo, pontuando que o sujeito negocia os sentidos dos conteúdos jornalísticos com o contexto que o circunda. Verifica-se que a família, os amigos, namorados(as) e outras mediações são variáveis importantes para a aquisição das competências midiáticas e devem ser considerados ao analisar como elas se manifestam nos indivíduos

A RELAÇÃO ENTRE CONSCIÊNCIA MORAL E ORDEM POLÍTICA NA OBRA “DE CIVITATE DEI” DE SANTO AGOSTINHO
No full textO presente artigo concentra-se na obra “De Civitate Dei”, apresenta primeiramente um panorama histórico da obra e seu objeto de análise, situando o escrito de Agostinho no contexto da queda do Império Romano e na resposta que o autor dá aos acontecimentos históricos. Percebemos que a solução apontada pelo Santo Doutor possui um cunho eminentemente moral e só em um segundo momento político, tratando da vida virtuosa dos cidadãos mediante uma leitura da história sob um critério ético-moral. Agostinho opera uma transposição da teoria da vontade do âmbito subjetivo para o histórico, aborda a moralidade do homem mediante dos conceitos fundamentais caritas e cupiditas, os dois amores que correspondem à cidade terrestre e a cidade celeste. Palavras-chave: Moral; Estado; caritas; civitas. Abstract: This article focuses on the work "De Civitate Dei," first presents a historical overview of the work and its object of analysis, situating the writing of Augustine in the context of the fall of the Roman Empire and the response that the author gives to events Historic. We realized that the solution proposed by the Holy Doctor has an eminently moral and just die in a second political moment, addressing the virtuous life of citizens through the reading of the story from an ethical-moral criterion. Augustine operates a transposition of the subjective theory of the will to the historical context, discusses the morality of man through the basic concepts cupiditas and caritas, the two loves that match earthly city and the heavenly city.
