Seismic risk of infrastructure systems with treatment of and sensitivity to epistemic uncertainty
No full textModern society’s very existence is tied to the proper and reliable functioning of its Critical Infrastructure (CI) systems. In the seismic risk assessment of an infrastructure, taking into account all the relevant uncertainties affecting the problem is crucial. While both aleatory and epistemic uncertainties affect the estimate of seismic risk to an infrastructure and should be considered, the focus herein is on the latter. After providing an up-to-date literature review about the treatment of and sensitivity to epistemic uncertainty, this paper presents a comprehensive framework for seismic risk assessment of interdependent spatially distributed infrastructure systems that accounts for both aleatory and epistemic uncertainties and provides confidence in the estimate, as well as sensitivity of uncertainty in the output to the components of epistemic uncertainty in the input. The logic tree approach is used for the treatment of epistemic uncertainty and for the sensitivity analysis, whose results are presented through tornado diagrams. Sensitivity is also evaluated by elaborating the logic tree results through weighted ANOVA. The formulation is general and can be applied to risk assessment problems involving not only infrastructural but also structural systems. The presented methodology was implemented into an open-source software, OOFIMS, and applied to a synthetic city composed of buildings and a gas network and subjected to seismic hazard. The gas system’s performance is assessed through a flow-based analysis. The seismic hazard, the vulnerability assessment and the evaluation of the gas system’s operational state are addressed with a simulation-based approach. The presence of two systems (buildings and gas network) proves the capability to handle system interdependencies and highlights that uncertainty in models/parameters related to one system can affect uncertainty in the output related to dependent systems
Reevaluating the function of a transcription factor: MBF-1 as a sea urchin chromatin organizer ?
No full textThe Zinc-finger MBF-1 factor is involved in the expression of the early histone genes during devel-opment of the sea urchin embryo (1, 2). In spite of being a transcription activator, the DNA-binding domain of MBF-1 shares high sequence similarity with that of the chromatin organizer CTCF of vertebrates and drosophila (3). On the other hand, extensive in silico analysis failed to identify the sea urchin CTCF ortholog (4). This led us to speculate that MBF-1 somehow could have co-opted the function of CTCF during evolution of the echinoderms. Since in vertebrates CTCF binds Hox chromatin, to support our hypothesis, we first identified high-score putative binding sequences for CTCF/MBF-1 within the single sea urchin Hox gene cluster. Moreover, we observed the full evolu-tionary conservation of these binding sites in S. purpuratus and P. lividus species. Worth of men-tion, by chromatin immunoprecipitation (ChIP) assay, we detected the occupancy of MBF-1 on hox11/13-a, -b, and -c regulatory sequences at distinct stages of development. As expected from the binding of an activator, we found that the association of MBF-1 to the cis-regulatory sequences of both hox11/13-a and -b genes relates to the transcriptional status of these genes. Strikingly, we also mapped the physical binding of MBF-1 to hox11/13-c, which is instead not expressed during em-bryogenesis. Altogether, these observations indeed suggest the possibility that MBF-1, besides be-ing a transcription activator, could also function as a general chromatin organizer. To further support this hypothesis, we are planning ChIP-seq experiments to identify the association of MBF-1 to the sea urchin chromatin at a genome-wide level.

Köpek testislerinde hacim hesaplaması için Cavalieri metodu uygulanabilirliği
No full textAmaç: Sunulan bu çalışmada bir nesnenin hacminin hesaplanması için tarafsız ve etkin bir metot olan Cavalieri metodunun invitro olarak köpek testisi üzerine uygulanabilirliği araştırılmıştır. Gereç ve Yöntem: Çalışmada 5 adet yetişkin köpekten alınan testisler kullanıldı. Arşimet prensibi ile hacimleri hesaplanan testislerin daha sonra Cavalieri metodu uygulanarak hacimleri hesaplandı ve sonuçlar karşılaştırıldı. Bulgular: Yapılan ölçümler neticesinde sağ ve sol testisler için hacim ortalama olarak Arşimet prensibince 18.72 cm3 , Cavalieri metoduyla da 18.25 cm3 olarak bulunmuştur. İki metod ile elde edilen sonuçlar arasında istatistiksel olarak farklılık bulunmadı (p<0.05). Öneri: Hacim hesaplamada endirekt yöntem olan su taşırma prensibi (Arşimet prensibi) küçük testislerde hacim hesaplamasında “kapiller etki” probleminden dolayı hata verirken Cavalieri metodu nesnenin yapısından bağımsız olarak çalışır ve en karmaşık şekli olan nesnelerin hacimlerini bile kolayca hesaplayabilir. Elde edilen sonuçlar Cavalieri metodunun köpek testisinin hacminin hesaplanmasında kullanılabileceğini göstermiştir.Aim: Aim of this research was to investigate the capability of Cavalieri principle (unbiased and efficient method using to volume measurement) for the in vitro estimation of the volume of the dog. Materials and Methods: Five male adult dog testes were used. First, testis volumes were measureament by water displacement method (Archimedes principle), then cavalieri method used to estimate the volume of same testis. Afterwards, the results of these methods were compared. Results: The mean volume of the left and right testis together was found 18.72 cm3 according to Archimedean principle and 18.25 cm3 according to Cavalieri principles. Statistical difference was not found (p<0.05). Conclusion: The direct volume measurement method “water displacement” could not be Performed on the small testis because of the “capillary effect”. However Cavalieri principle runs free from the shape and orientation of the material. The study showed that Cavalieri principle was capable to measure the testis volume
