    Morphologic and functional correlates of synaptic pathology in the cathepsin D knockout mouse model of congenital neuronal ceroid lipofuscinosis

    Mutations in the cathepsin D (CTSD) gene cause an aggressive neurodegenerative disease (congenital neuronal ceroid lipofuscinosis) that leads to early death. Recent evidence suggests that presynaptic abnormalities play a major role in the pathogenesis of CTSD deficiencies. To identify the early events that lead to synaptic alterations, we investigated synaptic ultrastructure and function in presymptomatic CTSD knockout (Ctsd) mice. Electron microscopy revealed that there were significantly greater numbers of readily releasable synaptic vesicles present in Ctsd mice than in wild-type control mice as early as postnatal day 16. The size of this synaptic vesicle pool continued to increase with disease progression in the hippocampus and thalamus of the Ctsd mice. Electrophysiology revealed a markedly decreased frequency of miniature excitatory postsynaptic currents (mEPSCs) with no effect on paired-pulse modulation of the evoked excitatory post synaptic potentials in the hippocampus of Ctsd mice. The reduced mEPSCs frequency was observed before the appearance of epilepsy or any morphologic sign of synaptic degeneration. Taken together, these data indicate that CTSD is required for normal synaptic function and that a failure in synaptic trafficking or recycling may bean early and important pathologic mechanism in Ctsd mice; these presynaptic abnormalities may initiate synaptic degeneration in advance of subsequent neuronal loss

    Phenotypic reversal of the <i>btn1</i> defects in yeast by chloroquine: A yeast model for Batten disease

    BTN1 of Saccharomyces cerevisiae encodes an ortholog of CLN3, the human Batten disease gene. We have reported previously that deletion of BTN1, btn1-Δ, resulted in a pH-dependent resistance to d-(−)-threo-2-amino-1-[p-nitrophenyl]-1,3-propanediol (ANP). This phenotype was caused by btn1-Δ strains having an elevated ability to acidify growth medium through an elevated activity of the plasma membrane H+-ATPase, resulting from a decreased vacuolar pH during early growth. We have determined that growing btn1-Δ strains in the presence of chloroquine reverses the resistance to ANP, decreases the rate of medium acidification, decreases the activity of plasma membrane H+-ATPase, and elevates vacuolar pH. However, an additional effect of this phenotypic reversal is that activity of plasma membrane H+-ATPase is decreased further and vacuolar pH is increased further as btn1-Δ strains continue to grow. This phenotypic reversal of btn1-Δ can be considered for developing a therapy for Batten disease

    Maintenance and Update Frequency: notPlannedStatement: This Batten Fault Zone Gravity Survey (P201780), gravity point data contains ground gravity point data for the NTGS Batten Fault Zone Gravity Survey (P201780) survey acquired for Northern Territory of Australia (Northern Territory Geological Survey). This dataset contains a total of 7475 point data values. The data is located in NT and were acquired in 2017. The point located data were collected in grids layout at a station spacing between 500 and 2000 metres. Terrain corrections were calculated using the INTREPID Geophysics software package. The processed data are checked by GA geophysicists using standard methods for assessing quality to ensure that the final data are fit-for-purpose. All data are provided in EPSG:4283 coordinates, Australian Height Datum (AHD) and gravity datum of AAGD07. The units are degrees, meters, and micrometres per second squared, respectively. Reference: Intrepid Geophysics, http://www.intrepid-geophysics.com.Gravity data measures small changes in gravity due to changes in the density of rocks beneath the Earth's surface. The data collected are processed via standard methods to ensure the response recorded is that due only to the rocks in the ground. The results produce datasets that can be interpreted to reveal the geological structure of the sub-surface.     Wiley College and the Literacy Project

    In the years after the Civil War until the 1930s, blacks in America undertook a literacy project -- a vast effort, long-lived, undertaken not by the power, authority, or bureaucracy of government, but instead by blacks acting on their own, unaware of similar efforts in a thousand other places. In the project, Wiley College in Marshall, Texas, a Historically Black College, assumed the obligation to develop leaders, equip teachers, and contribute in any way possible to Negro literacy. Scholars of black colleges have lumped all black colleges together and assumed that what was true about one college was true of all. While this paper does not argue that Wiley College is a representative case study, its story is so different from widely accepted narratives about black colleges that the paper supports the need for historians to take a fresh look at accepted narratives. Commonly held narratives state that the two crucial elements of Negro college success were overall direction by northern church denominations in the 1800s and financial support from northern foundations in the early decades of the twentieth century; they deny agency to leaders and supporters of HBCs like Wiley College. The narrative I trace shows people of Wiley, its President, Matthew W.     Characterization of a cross-reactive linear epitope in human genogroup I and bovine genogroup III norovirus capsid proteins

    The Southampton norovirus (SV) capsid protein was expressed as VLPs by recombinant baculoviruses in insect cells and was used to immunize mice for the production of monoclonal antibodies (mAbs). One mAb, CM54, showed broad cross-reactivity to genogroup I (GI) noroviruses, but was not reactive to GII capsid proteins. Interestingly mAb CM54 reacted to a bovine norovirus capsid protein. Immunoblot analysis indicated the binding site for CM54 was located in the shell domain between amino acid residues 102–225 of the SV capsid protein. The epitope was mapped to high resolution using a peptide array and was located to the sequence LEDVRN at amino acid residues 162–167. Alignment of norovirus capsid protein sequences confirmed the epitope sequence was common to particular groups of human and bovine noroviruses. Modeling of the epitope onto the recombinant NV capsid protein revealed it was located to the inner surface of the shell domain. <br/

    This is Burt's reprint of what seems to have been a Putnam original. I have the Putnam edition listed also under 1905? The text and illustrations seem exactly the same throughout, but the typesetting of the text results in the book's finishing on 278 rather than 272. As I write there, most of the twenty-nine selections are indeed fairy tales, heavy on magic, demons, fairies, and angels. One poor Brahman after another runs into long, episodic adventures full of prophecies and beautiful princesses. The best examples might be Lambikin (22) and his protective drum, Punchkin (27), and The Soothsayer's Son (88). In the midst of these fairy tales, there are Jatakas and Kalila and Dimna materials like The Cruel Crane Outwitted (59) and TT (125). There are also Aesopic materials, like The Crane and the Lion (not wolf, 1), The Gold-giving Serpent (140), and DLS (184).
