Città per la gente. Intervista a Jan Gehl
In the interview Jan Gehl traces some fundamental stages of his training and his decades of research and professional activity. From the study trip in Italy in the early sixties’ the Danish architect's main concern was to think about space and its ability to influence the quality of life of those who live in it. Ingrid Mund, psychologist and wife, the Anglo-Swedish architect Ralph Erskine and the city of Copenhagen were the starting points for a reflection on the city and the way it is used by people. Modernity has neglected the needs of citizens, their attitudes and desires. The Danish architect has worked all over the world to recover this error. From Melbourne and Perth to the recent proposals for New York, he applied the "Public Space / Public Life" method, which is a survey that is based on numbers and concrete data and that listens to the needs of citizens, offering adequate answers remembering that “ life follows form ". With a thought that always comes back to where it all started: Copenhagen, a pioneering city in the enhancement of public space as a space for people, which continues to be a global reference on the role that streets can take in terms of improvement the quality of life
Fil: Capuano, Vicente Conrado. Fil: Capuano, Vicente Conrado. Design of an electrophoretic module for protein separation
In this paper we discuss the model, theory and design optimization of an electrophoretic separation system configured as a split flow thin fractionation, which can be used for protein separation from milk samples as part of an integrated analytical system for detection of contaminants or pathogens. In particular, we describe the design and the fabrication strategy to achieve optimal separation and a sample-processing throughput in the order of ml/min
Continuous extraction of proteins with a miniaturized electrical split-flow cell equipped with suspended splitters fabricated by dry film lamination
This article describes the development of an electrically driven Split-Flow Thin microsystem suitable for continuous-flow protein purification or concentration. We used a technology based on dry film resist, Ordyl SY 355, to realize 3D suspended structures in a microchannel (150 μm), including patterned Cr/Pt electrodes. The device developed for this research is the first electrical miniaturized dry film SPLITT system including suspended splitters and represents a good miniaturization effort of the ideal SPLITT cell, thus expanding the possibilities of realizing split-flow fractionation microfluidic network based on dry film technology. A suitable design and fabrication process for the realization of such a kind of device is reported. The purification/concentration performances of the system have been tested by infusing a Bovine Serum Albumin solution (0.1 weight percentage) through the inlets and evaluating the protein content at the outlets after the application of a pulsed electric potential. Experimental conditions have been changed during the testing phase, by varying the applied potential and the duty cycle of the square wave and the flow rate to find suitable conditions for protein extraction. The best-achieved purification/concentration rate was around 60%, measured as concentration difference between the two output channels normalized on the input concentration, thus confirming the viability of device for protein enrichment or purification for sample preparation. The device has a potential for integration in micro-TAS, thanks to its reduced dimensions, for applications involving protein analysis or bio-sensing in matrices including proteins interfering with the measurement
A Miniaturized SPLITT System for On-Line Protein Separation
The development of a microfluidic module for on-line protein separation is presented in this paper. The device is based on the SPLITT (Split flow Thin fractionation) technique and the separation of proteins is electrically driven by means of two platinum electrodes included in the structure of the microdevice. The microfluidic network is realized by means of a thin dry film structure, Ordyl SY 355, (thermo-compression lamination), laminated on three levels, which is patterned through a photolithographic technique. The device has been tested with a Bovine Serum Albumin (BSA) solution, through absorbance measurements with a spectrophotometer, with best achieved separation at the outlet of 40%, measured as relative concentration unbalance at output channels
A continuous flow microelectrophoretic module for protein separation
In the framework of our research work, we are developing a microfluidic system that can be used as a pre-treatment device for the sample preparation in analytical microsystems, as biosensors, for instance. Sample pre-treatment phase is usually envisaged before chemical and biological analysis. For example, if we want to perform contaminants detection in a complex matrix as milk, we must consider that the sample consists in a solution of butterfat globules and water with dissolved carbohydrates and protein complexes. If we want a bio-sensor to perform a label free detection of some target-contaminants in milk, interferents in the matrix must be removed. Proteins are an important part of these interferents, and their concentration needs to be reduced, in order to avoid fouling and consequently the inhibition of the sensor, or false positive results in the analysis. We have designed and fabricated a SPLITT flow thin fractionation device for protein separation that is now under testing with a BSA solution
