The feasibililty of a less invasive method to assess endometrial maturation--comparison of simultaneously obtained uterine secretion and tissue biopsy
Objective: to compare the assessment of endometrial maturation parameters in endometrial secretion samples obtained by a novel minimally invasive technique with those assessed in tissue biopsies. Design: prospective study. Setting: university hospital. Population: healthy female volunteers attending a gynaecological outpatient clinic. Methods: endometrial secretion fluid and tissue sampling 5 days after a spontaneous ovulation assessed with ultrasound. Main outcome measures: progesterone (P) receptor, Ki-67 expression and the Noyes criteria were used to date endometrial biopsies. In the endometrial fluid samples, glycodelin A (GdA), leukaemia inhibitory factor (LIF) and P levels were analysed, and protein content and electrophoresis patterns were determined. RESULTS: All data were correlated to estradiol (E2) and P serum concentrations. The dating according to histology and immunohistochemical staining patterns correlated significantly with GdA levels (r=0.376, P=0.048) in endometrial fluid samples as well with serum levels of E2 (r=0.568, P=0.001) and P (r=0.408, P=0.023). No correlation was observed between tissue dating and LIF levels and protein content in endometrial fluid samples. Conclusions: the measurement of GdA in endometrial secretion samples may provide a less invasive method for assessing endometrial maturation in potential conception cycles without disrupting implantation.<br/

Impact of ovarian stimulation on mid-luteal endometrial tissue and secretion markers for receptivity
The objective of this study was to investigate the effect of ovarian stimulation for IVF on endometrial secretion and tissue markers of receptivity in the mid-luteal phase. In 10 oocyte donors, endometrial secretions and biopsies were sampled 5 days after spontaneous ovulation and oocyte retrieval in consecutive cycles. Four subjects received progesterone in the luteal phase of the stimulated cycles. Mid-luteal endometrial maturation in the stimulated cycle was compared with the spontaneous cycle, by histological dating, Ki-67, oestrogen receptor (ER) and progesterone receptor (PR) expression, secretion levels of leukaemia inhibitory factor (LIF), glycodelin A (GdA) and progesterone, and protein profile. No significant differences in histological markers, expression of Ki-67, PR, ER, secretion protein profiles or concentrations of LIF, GdA, or progesterone were observed when comparing natural with stimulated cycles. Progesterone supplementation of stimulated cycles was associated with significantly lower Ki-67 (P = 0.03) and ER (P = 0.04) expression compared with the non-supplemented stimulated cycle. In this pilot study, ovarian stimulation was not demonstrated to alter the studied markers of endometrial maturation in the mid-luteal phase
Messung der Hell-Dunkel-Adaptation und ihre Beeinflussung durch die extrazelluläre Calciumkonzentration : elektrophysiologische Messungen am Ventralnerv Photorezeptor von Limulus polyphemus
Method: A set-up for intra- and extracellular electrophysiological measurements on the Limulus ventral nerve photoreceptor was built up. A suction electrode method was developed to record the extracellular currents of a single photoreceptor cell at different membrane areas. Simultaneously the voltage response (receptor potential) could be measured intracellularly. 1) The time course of dark adaptation following light adaptation by a bright 1 or 5 s illumination (4,4 10 photons cm s at 543 nm) was measured by two different methods: a) were determined by measuring the amplitude of the intracellularly recorded receptor potential in response to test stimuli of constant intensity and duration in dependence on the dark adaptation time. b) were determined by measuring the sensitivity increase in dependence on the dark adaptation time by determining the light intensity necessary to evoke a criterion response amplitude of the receptor potential. The experimental data for the dark adaptation time t and the light intensity I were fitted by two exponential functions I = and by two power functionsI = , respectively. The better fit - that means the higher r -values (correlation coefficient) could be obtained by the power functions (fig. 18). 2) The time course of dark adaptation was characterized by two different phases, a fast and a slow one. The first rather fast increase of sensitivity after light adaptation ( = 5,5 s; s = ± 0,6 s, n = 16) was followed by a second slower phase ( = 287 s; s = ± 44 s, n = 16). In the double logarithmic plot two different slopes could be determined. The exponent characterizing the first slope was 3,5 (s = ± 0,4; n = 16) in the average and characterizing the second slope was 1,0 (s = ± 0,1; n = 13) in the average. 3) The influence of the extracellular Ca-concentration on the dark adaptation process was investigated. Only the first phase of dark adaptation was strongly dependent on thechanged extracellular Ca-concentration - in accordance with the Ca-hypothesis (Lisman and Brown, 1972) - while the second phase of dark adaptation was nearly Ca-independent (s. Tab. 11). According to this effect the coefficients and of the power function were changed: The exponent was decreased to 1,7 (s = ± 0,5; n = 6) when the external Ca-concentration was lowered from10 mmol/l to 250 mol/l (see for example fig. 22); was increased to 4,7 (s = 0,5; n = 3) raising the extracellular Ca-concentration to 40 mmol/l (see for example fig. 24). The factor characterizing the sensitivity for t = 1 s varies greatly from experiment to experiment; is decreased - corresponding to a greater sensitivity - to about 5 % in the average by lowering the extracellularCa-concentration and strongly increased by a factor of 70 by raising the extracellular Ca-concentration
Einflüsse ubiquitär vorkommender Xenobiotika auf humane immortalisierte endometriale Epithelzellen (hTERT-EEC B37)
Die in dieser Arbeit verwendeten Umweltkontaminanten, Dioxine und polychlorierte Biphenyle sind ubiquitär anzutreffen, sie entstehen vor allem in den industrialisierten Ländern. Aufgrund ihrer guten Lipidlöslichkeit, sowie ihrer hohen Halbwertszeit kummulieren sie in der Nahrungskette. Im Rahmen dieser chronischen Exposition gelten sie als karzinogen und lösen in Säugetieren multiple Entwicklungs- und Differenzierungstörungen aus, auch eine Rolle bei der Genese der Endometriose wird diskutiert. Zu unterscheiden ist die akute Intoxikation mit den Folgen einer Chlorakne und dem möglichen „wasting-syndrome“, welche nicht Bestandteil dieser Arbeit war. Ein großer Teil der zellulären Wirkungen der Dioxine werden über den sogenannten „Dioxin-Rezeptor“, den Arylhydrocarbonrezeptor (AhR) gesteuert. Bisherige Untersuchungen dieser zellulären Abläufe erfolgten ausschließlich an malignen, transformierten Tumorzellen. Ziel dieser Arbeit war, die humane endometriale Epithelzelllinie hTERT-EEC B37 und deren Reaktionen auf eine Exposition mit Dioxin und polychlorierten Biphenylen (PCB) zu untersuchen. Die hTERT-EEC B37 sollten durch Expositionsversuche und nachfolgenden Untersuchungen auf Transkriptions- und auf Translationsebene als in vitro Zellmodell charakterisiert werden. Die Funktionalität des rezeptorgebundenen Signaltransduktionsweges sollte mit Hilfe von transienten Transfektionen untersucht werden. Es zeigte sich eine Induzierbarkeit der klassischen AhR-Zielgene, vor allem von CYP 1A1, allerdings in unterschiedlicher Stärke durch die verschiedenen PCB. Auch ließ sich eine Wechselwirkung mit Östrogenen beobachten. Im Rahmen der durchgeführten Proliferations- und Apoptoseversuche zeigten sich die Umweltkontaminanten geringfügig proliferationshemmend, nicht jedoch apoptosefördernd. Die hTERT-EEC B37 stellten sich im Rahmen dieser Arbeit als ein durchaus geeignetes Zellmodell zur weiteren Untersuchung AhR-vermittelter Effekte heraus. Ziel zukünftiger Arbeiten sollte es sein, die in vivo stattfindenden Wechselwirkungen zwischen endogenen Botenstoffen und den exogenen Xenobiotika zu eruieren.von Marco Polo Peic
Einfluss eines experimentell induzierten Typ-1-Diabetes mellitus auf endometriale Faktoren während der Frühschwangerschaft am Modell des Kaninchens
Frauen mit Typ-1-Diabetes mellitus sind subfertil. Eine Ursache dafür ist eine gestörte embryo-maternale Kommunikation bereits in der Präimplantationsphase. Ziel dieser Arbeit war es, endometriale Faktoren zu detektieren, welche durch einen maternalen Diabtes verändert sind. Als Reproduktionsmodell diente das Kaninchen. Diesem wurde durch die Chemikalie Alloxan ein experimenteller Diabetes mellitus induziert. Im ersten Teil wurden die Faktoren des Insulin-IGF-Systems untersucht. Zur Detektion weiterer endometrialer Faktoren erfolgte im zweiten Teil ein Next Generation Sequencing, wobei zahlreiche durch Diabetes und Gravidität veränderte Faktoren und Signalwege gefunden wurden. Im dritten Teil konnte ich erstmals Protocadherin 15 im Endometrium näher charakterisieren. Die Ergebnisse zeigen, dass es bereits in der Präimplantationsphase durch einen maternalen Diabtes zu Veränderungen endometrialer Faktoren kommt.Woman with type-1 diabetes mellitus are subfertile. Aetiological could be a disrupted embryo-maternal communication already during the preimplantation period. Aim of this dissertation was to find endometrial factors affected by maternal diabetes. We employed the rabbit as reproduction model. Diabetes mellitus was induced by alloxan. First I was investigating the impact of diabetes on factors of the insulin-IGF-system. Then I did a next generation sequencing to search for further endometrial factos influenced by diabetes and pregnancy. In the third part I characterized for the first time Protocadherin 15 in endometrium of human and rabbit.
