What remains of a personal library is much more than a catalogue of titles. My father, Bernardo Secchi, had a vast and stratified library which, as well as books and notes, is a rich deposit of research themes and trains of thought. Starting over from the library means revealing the figure of the author as a reader: not just an alter ego for the designer, the planner or the researcher. Through the library we trace and discover different paths: they will tell us about the author’s relationship with the academic disciplines, with other authors, with the construction of planning’s specific discourse and with the multitude of diverse knowledges which are unavoidably implicated. The library is, thus, much more than a metaphor of knowledge: its construction and composition correspond in a concrete and revelatory way to the unfolding of intellectual labour, in its making, in its digression and ultimately in style
Studies on analogues of succinic semialdehyde as substrates for succinate semialdehyde dehydrogenase from rat brain
The methyl ester of succinic semialdehyde (SSA) was examined as a substrate for succinate semialdehyde dehydrogenase (SSADH) from rat brain. It was found that the ester can be oxidized by the enzyme. Values of K(m) for SSA-Me were higher than for those for SSA, and for this substrate the enzyme showed a substrate- dependent inhibition. This finding suggests that the carboxylate group of SSA is not essential in the process of inhibition of SSADH by the substrate. Cyclopropyl analogues of SSA, cis- and trans-1-formyl-cyclopropan-2-carboxylic acids, were also individually tested as substrates of SSADH. Only the trans isomer was found to be oxidized to the corresponding dicarboxylic acid; it inhibited the enzyme in the same range of concentrations as SSA. The above data suggest that, as for γ-aminobutyric acid, SSA is present in an unfolded, transoid conformation at the active site of SSADH

Growth hormone secretion in relation to plasma lactate and glucose concentrations during a maximal treadmill exercise test in horses
Detection of desamido forms of purified bovine growth hormone
Tryptic peptide mapping and amino acid sequencing were used for the identification of desamido forms of bovine growth hormone. The major desamido forms of bovine growth hormone resulted from deamidation of asparagine in position 13, 148 and glutamine 140. These forms did not seem to be produced by prolonged treatment of bovine growth hormone in tryptic hydrolysis
Gas Chromatographic analysis of neutral monosaccharides as their O-pentafluorobenzyloxime acetates
In order to develop a new derivatization procedure for the gas chromatographic analysis of sugars, O-(2,3,4.5.6-pentafluorobenzyl)hydroxylamine was reacted with the neutral monosaccharides most commonly present in glycoproteins. The resulting oximes were subsequently acetylated, and the resulting volatile derivatives can be separated on both packed and capillary columns. The method appeared to be rapid, simple and reliable. As an application, the neutral sugars content of some known glycoproteins was analyzed
HPLC determination of D-amino acid oxidase activity
A new procedure for the assay of D-amino acid oxidase activity has been developed. alpha-Ketoisovaleric acid, derived from D-valine, was estimated by high-performance liquid chromatography after reaction with o-phenylenediamine to give the corresponding quinoxalinol derivative. alpha-Ketovaleric acid was used as an internal standard to ensure the reproducibility of the method. As an example of application, kidney cortex homogenates were analyzed for their D-amino acid oxidase activity. The advantages of the presented procedure for the determination of the enzymatic activity in biological samples compared with previously reported procedures are discussed
Identification of freshwater fish commercially labelled "perch" by isoelectric focusing and two-dimensional electrophoresis
Isoelectic focusing (IEF) and 2-dimensional electrophoresis (2-DE) were used to distinguish 4 freshwater fish species which are sold under the generic label of "perch": Perca fluviatilis (European perch), Lates niloticus (Nile perch), Stizostedion lucioperca (European pike perch) and Morone chrysops x saxatilis (sunshine bass). These species have different com. values but are easily "interchangeable" because they are sold already filleted, in view of the numerous bones of the whole fish. IEF of the water-sol. proteins extd. from fish muscle resolved in species-specific patterns. Intra-species polymorphism was low, and did not concern the bands identified as characteristic of the species. As well, 2-DE maps showed numerous species-specific protein spots. Interestingly, while none of the IEF bands was common to all 4 species, several major 2-DE spots were similar. Therefore, IEF of water sol. sarcoplasmic proteins is sufficient to unambiguously discriminate among the 4 species considered. Anal. by 2-DE, which has a higher resoln. power but it is more expensive and time consuming, may be applied to obtain further knowledge of the proteome of poorly characterized species

The significance of Aquaporins in placenta and fetal membranes : the regulation of amniotic fluid production and composition during canine pregnancy
Although cats are induced ovulators, the relationship between the day of breeding, the number of matings and the likelihood of ovulation and conception have not been extensively investigated. In this experiment, cats were mated either once or three times on day 1 or day 5 of oestrus to study the incidence of the LH surge, ovulation and conception rates. The percentage ovulating and the conception rates after a single mating on day 1 of oestrus were 60% (6/10) and 33.3% (2/6), respectively, and for cats mated once on day 5 of oestrus were 83.3% (10/12) and 40% (4/10), respectively. When cats were mated three times on day 1 of oestrus, the ovulation rates and conception rates were 70% (7/10) and 85.7% (6/7), respectively, and for those mated three times on day 5 of oestrus were 100% (10/10) and 100% (10/10), respectively. The concentration of LH did not increase in non-ovulating cats, and cats that were mated three times had LH concentrations that were numerically higher than those that were mated once. Litter size was neither related to the day of mating nor to the number of matings. Although an increase in the number of matings on day 1 of oestrus produced a numerically larger LH surge, it did not increase the ovulation rate, suggesting that plasma oestradiol concentrations were not sufficiently elevated to induce a high pituitary response to mating stimulation. The conception rate after a single mating was low, suggesting that the number of sperm per mating was not sufficient. These results suggest that mating more than once in the middle of oestrus is required to improve ovulation rates and conception rates in cats
