Sviluppo e validazione di un nuovo sistema diagnostico rapido per l’isolamento e la valutazione dell’antibiotico suscettibilità di Mycoplasma spp. in medicina veterinaria
Mycoplasmas are recognized as a cause of many and important infections in veterinary medicine, resulting in different clinical signs, poorly responsive to antimicrobial therapy. Their isolation should be considered in the differential diagnosis and, because an accurate identification is crucial in an effective therapeutic program, new techniques can be useful to overcome the long cultivation period required. Moreover, efficacious post-therapeutic outcomes need a timely diagnosis combined with an appropriate selection of antibacterial agents. The aims of this work were to: -develop a new diagnostic system for the rapid and semi-quantitative detection of Mycoplasma species in veterinary medicine, directly from clinical specimens; -validate the performances of the new system; -optimize the validity of the antibiotic susceptibility testing evaluating the Minimal Inhibition Concentration of a panel of nine antibiotics used in veterinary medicine

Validity and reliability of the Eating Problem Checklist
The aim of this study was to validate the latest version of the Eating Problem Checklist (EPCL), a tool designed to assess eating-disorder behaviours and psychopathology in patients with eating disorders, session-by-session. The EPCL was completed at baseline by participants with eating disorders (n = 161) and a healthy control group (n = 379) and then administered session-by-session in a subgroup of 75 participants with eating disorders. The EPCL demonstrated good internal consistency, test-retest reliability, and concurrent and criterion validity, and principal axis analysis of the session-by-session data identified two factors ('eating concerns' and 'body image concerns') that accounted for 51.3% of the variance. Furthermore, session-by-session analysis indicated that the EPCL is able to identify specific weekly improvements and/or deterioration in eating-disorder psychopathology. These findings suggest that the EPCL is a valid and reliable self-report questionnaire that provides relevant clinical information regarding weekly changes in eating-disorder behaviours and psychopathology in patients with eating disorders
A simplified FE model for the non-linear analysis of container stacks subject to inertial loads due to ship motions
On board ships, cargo-related failure conditions are strongly dependent on cargo and lashing systems. This study presents a procedure for achieving an effective FE structural model of a container stack for carrying out the numerical simulations with the aim to define a numerical model that accurately simulates the actual deformation of a stack, making it as simple as possible in order to decrease the computational cost of the simulations. Using this model, a series of numerical analyses can be carried out for a huge amount of loading conditions obtained by non-linear numerical simulations of ship motions
Exocrine and endocrine functional reserve in the course of chronic pancreatitis as studied by maximal stimulation tests
Thirty patients suffering from chronic alcoholic pancreatitis (18 calcified) were entered into a study of exocrine and endocrine pancreatic function based on two maximal stimulation tests, namely the secretin-cerulein test and the glucagon test with serum assays of C peptide. The glucagon test was also performed in 19 control subjects. In addition, 10 chronic pancreatitis patients and nine controls were subjected to an oral glucose tolerance test (OGTT) with serum insulin determinations. C peptide basal values were decreased only in patients with severe pancreatic exocrine insufficiency (P < 0.001), while DELTA-C peptide values were also reduced in patients with moderate exocrine insufficiency (P < 0.001). Lipase output correlated very well with DELTA-C peptide values (P < 0.001). While serum insulin levels during OGTT and C peptide basal values showed no significant differences between the chronic pancreatitis and control groups, DELTA-C peptide values were significantly reduced in chronic pancreatitis patients (P < 0.02). Both endocrine and exocrine function are impaired in chronic pancreatitis, as demonstrated by maximal tests, even in early stages of the disease
Resistance to activated protein C in healthy women taking oral contraceptives
Resistance to activated protein C (APC) is at present considered the most frequent laboratory abnormality in patients with deep-vein thrombosis. An increased risk for venous thrombosis is associated to the use of oral contraceptives (OC). We studied APC sensitivity in 50 healthy women taking OC and in 50 healthy controls, matched for age, smoking habit, educational and social levels, and the main biochemical routinary parameters. Subjects with a personal or familial history of thrombosis and also with chronic or acute diseases were excluded. Protein C, protein S, antithrombin III and lupus anticoagulant activity (LAC) were also evaluated. Increased fibrinogen and protein C levels, decreased protein S, and shortened PT and APTT were also observed in women taking OC.APC sensitivity ratio (APC-SR) was significantly lower in the OC group than in a control group (2.6 +- 0.38 v 2.81 +- 0.35, P lt 0.01). Seven of eight women with APC ratio ltoreq 2 (APC resistant) were OC users: the difference of prevalence was statistically significant (chi-squared test, P lt 0.05). Only two out of eight women were found heterozygous for the Leiden factor V mutation. Two APC-resistant women without the Leiden mutation subsequently discontinued OC and both then normalized their APC-SR. We conclude that acquired factors, i.e. oral contraceptives, may play an important role in determining plasma APC resistance
