No full textBorn in Clinton, Iowa, in 1985, Salvatore Sal Augustine Giunta decided to enlist at age 17, while working in a Subway sandwich shop. He joined the Army in November 2003. He attended Infantry One Station Unit Training and the Basic Airborne Course at Fort Benning, Ga., before being assigned to the 173rd Airborne Brigade Combat Team, Vicenza, Italy, in 2004. Promoted to the rank of staff sergeant in 2009, Giunta completed two combat tours to Afghanistan totaling 27 months of deployment. On Oct. 25, 2007, while conducting a patrol as team leader, Giunta and his team were navigating through the treacherous terrain of Afghanistan\u27s Korengal Valley when they were ambushed by a well-armed and well-coordinated insurgent force. While under heavy enemy fire, Giunta immediately sprinted toward cover and engaged the enemy. Seeing that his squad leader had fallen and believing that he had been injured, Giunta exposed himself to withering enemy fire and raced toward his squad leader, helped him to cover and administered medical aid. While administering first aid, Giunta\u27s body armor was struck by enemy fire. Without regard to the ongoing fire, he engaged the enemy before prepping and throwing grenades, using the explosions for cover in order to conceal his position. Attempting to reach additional wounded fellow soldiers who were separated from the squad, Giunta and his team encountered a barrage of enemy fire that forced them to the ground. The team continued forward and upon reaching the wounded soldiers, Giunta realized that another soldier was still separated from the element. Giunta then advanced on his own initiative. As he crested the top of a hill, he observed two insurgents carrying away an American soldier. He immediately engaged the enemy, killing one and wounding the other. Upon reaching the wounded soldier, he began to provide medical aid as his squad caught up and provided security. For his extraordinary gallantry, Giunta was awarded the Medal of Honor, making him the first living recipient of the Medal of Honor for service in Iraq or Afghanistan, the first living service member to be awarded the Medal of Honor since the Vietnam War and the eighth service member to receive the nation\u27s highest military decoration for valor in Iraq and Afghanistan. Giunta\u27s other military decorations include the Bronze Star, Purple Heart, Army Commendation Medal with oak leaf cluster, Army Achievement Medal, National Defense Service Medal, two Army Good Conduct Medals and the Global War on Terrorism Service Medal. Giunta retired from the Army in June 2011, and in 2012 authored Living with Honor. He currently lives in Colorado with his wife, Jennifer, and daughter

No full textIl saggio di Santo Giunta riflettere sul rapporto tra restauro e rifunzionalizzazione di un edificio storico, focalizzando l’attenzione su un’opera di Pasquale Culotta a Raccuja (ME). Il saggio è pubblicato in un libro, curato da Calogero Marzullo, che nasce dalla volontà di rileggere alcune riflessioni su ricerche e sperimentazioni di diversi autori che dialogano tra loro per tentare di definire il valore del progetto di architettura. Nel caso di studio in esame il progetto non è solo la rispondenza funzionale di elementi riconoscibili tra le proposte strutturali e culturali, ma tiene conto delle forme della vita contemporanea, di una socialità interagente e di confine. La qualità emotiva di una attività progettuale di Pasquale Culotta assorbe sguardi diversi e punti di vista molteplici che sono in grado di integrare le esigenze sociali delle persone con le condizioni d’origine a cui si vuole riportare il manufatto storico. È un complesso sistema di rapporti che lega queste parti fra significati e differenze in un’attività sistemica che mira alla definizione di parti con soluzioni strutturanti, attraverso nuovi servizi e ambiti riconoscibili. L’architetto, tra le proposte strutturali e culturali, interpreta per innesti puntuali le forme dentro il costruito. Il progetto deve ricostruire la propria ragion d’essere a partire dall’esistente non ignorando una sensibilità nei confronti delle tecniche artigianali, applicate in particolare agli ambiti spaziali per le attività culturali.The text by Santo Giunta reflect on the relationship between restoration and re-functionalization of a historic building, focusing attention on a work by Pasquale Culotta in Raccuja. It is published in a book, edited by Calogero Marzullo, which arises from the desire to reread some reflections on research and experimentation of different authors who interact with each other to try to define the value of the architectural project.
In the case study in question, the project is not only the functional correspondence of recognizable elements between the structural and cultural proposals, but takes into account the forms of life contemporary, of an interacting and borderline sociality. The emotional quality of Culotta's design activity absorbs different gazes and multiple points of view that are able to integrate the social needs of people with the conditions of origin to which the historical artefact is to be brought back. It is a complex system of relationships that links these parts between meanings and differences in a systemic activity that aims to define parts with structuring solutions, through new services and recognizable areas. Among the structural and cultural proposals, the architect interprets the forms within the built structure through punctual grafting. The project must reconstruct its raison d'etre starting from the existing one, not ignoring a sensitivity towards craft techniques, applied in particular to spatial areas for cultural activities
Riflessioni sui confini del giudizio di legittimità costituzionale a partire dall’“ordinanza Cappato”
No full textLo scritto si concentra su alcune specifiche questioni che emergono dall’ordinanza della Corte costituzionale n. 217 del 2019. In primo luogo, viene valutata l’idoneità della soluzione proposta dalla Corte a conseguire gli obiettivi indicati nell’ordinanza. In secondo luogo, vengono prese in esame le soluzioni alternative – già consolidatesi nella giurisprudenza costituzionale – che si sarebbero potute scegliere ai medesimi fini. Da ultimo, vengono avanzate proposte de jure condendo che consentirebbero al Giudice costituzionale di modulare gli effetti delle proprie decisioni ed evitare il ricorso a tecniche decisorie – quale quella utilizzata nel c.d. “caso Cappato” – che sembrano travalicare i confini dei “poteri di gestione del processo costituzionale”.The paper focuses on some specific aspects arising from the order of the Constitutional Court No. 217 of 2019. Firstly, the study assesses the suitability of the solution proposed by the Court for achieving the objectives defined in the order. Secondly, the Author considers the alternative solutions – already consolidated in the constitutional case law – which the Constitutional Court could have chosen for reaching the same purposes. Lastly, the paper put forward proposals de jure condendo, in order to allow the Constitutional Judge to modulate the effects of its decisions and to avoid the use of types of decision – such as the one used in the s.c. “Cappato case” – which seem to go beyond the boundaries of “management powers of the constitutional trial”

Finland che, al pari della sent. n. 170/2014 della Corte costituzionale italiana, si occupa del bilanciamento tra l’interesse dello Stato (in questo caso finlandese) alla conservazione del paradigma eterosessuale del matrimonio e l’interesse di una coppia (divenuta) omosessuale a mantenere il vincolo matrimoniale, validamente contratto prima del cambio di sesso di uno dei coniugi. La Corte EDU ritiene che subordinare il riconoscimento del mutamento di sesso alla conversione del matrimonio in unione registrata non violi né l’art. 8 né l’art. 14 CEDU, dal momento che la protezione offerta alle coppie omosessuali è del tutto analoga a quella riconosciuta alle coppie sposate

Centromere Chromosome Orientation Fluorescent in situ Hybridization (Cen-CO-FISH) Detects Sister Chromatid Exchange at the Centromere in Human Cells
No full textHuman centromeres are composed of large tandem arrays of repetitive alpha satellite DNA, which are often sites of aberrant rearrangement in cancers (Mitelman et al., 1997; Padilla-Nash et al., 2001). To date, annotation of the human centromere repetitive sequences remains incomplete, greatly hindering in-depth functional studies of these regions essential for chromosome segregation. In order to monitor sister chromatid exchange happening at the centromere (C-SCE) due to recombination and mutagenic events, I have applied the Chromosome-Orientation Fluorescence in situ Hybridization (CO-FISH) technique to centromeres (Cen-CO-FISH) in human cells. This hybridization-based method involves (1) the incorporation of nucleotide analogs through a single round of replication, (2) enzymatic digestion of the newly synthesized DNA strand and (3) subsequent hybridization of single-stranded probes, in absence of a denaturation step. The resulting signal allows to differentially label each sister chromatid based on the 5’−3’ directionality of the DNA and to score aberrant staining patterns indicative of C-SCE. The Cen-CO-FISH method applied to human centromeres revealed that human centromeres indeed undergo recombination in cycling cells resulting in C-SCE, and centromere instability is enhanced in cancer cell lines and primary cells undergoing senescence (Giunta and Funabiki, 2017). Here, I present the detailed protocol of the preparation, experimental procedure and data acquisition for the Cen-CO-FISH method in human cells. It also includes a conceptual overview of the technique, with examples of representative images and scoring guidelines. The Cen-CO-FISH represents a valuable tool to facilitate exploration of centromere repeats
