    Analysis of chromatin structure reveals the connection between sQTLs and the splicing of distant genes

    Gene expression and regulation with or without alternative splicing are key factors for cells to properly function. Distant splicing quantitative trait loci (distant sQTLs) are genomic mutations that impact the alternative splicing patterns of far-away genes. Nevertheless, the procedures causing a distant sQTL to regulate the alternative splicing of genes are not well defined. Higher resolution chromosome conformation capture experiments like Micro-C or Hi-C together with an expanding number of sQTL datasets on humans help us in understanding the spatial processes governing distant sQTL relationships at a genome-wide scale. In this study, we focus on analyzing whether spatial closeness helps in regulating sQTL-gene interactions over high-order chromatin topological domain structure, which is inferred from chromosome conformation experiments. We discover larger-scale chromatin shape to be in line with sQTL associations. In detail, sQTLs are generally spatially near their splicing genes in 3D, they frequently appear near topologically associating domain (TAD) and frequently interacting region (FIRE) boundaries, and are favorably related to genes over TADs and FIREs. Additionally, we discover that inside-domain sQTLs accompanied by functional regulatory elements, including enhancers and promoters, are spatially closer than all inside-domain sQTLs. This result suggests that spatial closeness between sQTLs and their distant splicing genes obtained from chromatin’s TAD structure has major importance in regulating alternative splicing and thus in gene regulation. Our results are robust across different experiments such as Hi-C and Micro-C, different TAD inference methods, different Hi-C binning resolutions, different alternative splicing events, and once we control for eQTLs, which are shown to be spatially close to their genes. © The Author(s) 2025.TÜBİTAKPublisher versio

    Identifying the time of a step change with multivariate single control charts

After a signal is generated by the simultaneously used control charts or a single control chart, add-on change point procedure estimates the time of the change. In this study, multivariate joint change point estimation performance for simultaneous monitoring of both location and dispersion is compared under the assumption that various single charts are used to monitor the process. The change detection performance for several structural changes for the mean vector and covariance matrix is also discussed.     Evaluating the Anchorflow Suture Technique Versus Conventional Continuous Suturing in Vaginal Cuff Closure During TLH: A Multicenter Retrospective Analysis

    Background and Objectives: Total laparoscopic hysterectomy (TLH) is a commonly performed gynecological procedure. Vaginal cuff closure significantly impacts operative time and outcomes. This study evaluates the newly developed Anchorflow Suture (AFS) technique compared to conventional continuous suturing in terms of efficiency and safety. Materials and Methods: A multicenter retrospective cohort study was conducted at two tertiary referral hospitals involving 208 women who underwent TLH for benign indications. Women were divided into two groups based on vaginal cuff closure technique: AFS and continuous suturing. Demographic characteristics, surgical parameters, and postoperative outcomes were analyzed using appropriate statistical tests, with a significance level of p < 0.05. Results: No significant differences were found between groups in age, BMI (body mass index), gravida, parity, or menopausal status. Vaginal cuff closure time was significantly shorter with AFS (10.26 ± 2.3 min) compared to continuous suturing (13.36 ± 2.8 min, p < 0.001). Operative time was shorter in the AFS group, though not statistically significant (p = 0.15). Both techniques demonstrated similar safety profiles, with no cases of vaginal cuff dehiscence and comparable rates of granulation tissue formation, bleeding, and urinary incontinence. The AFS group showed a slightly lower incidence of postoperative bleeding (five cases vs. three cases). Conclusions: The AFS technique significantly reduces vaginal cuff closure time and demonstrates a comparable safety profile to continuous suturing. This method enhances surgical efficiency without increasing complications. Further prospective studies are needed to evaluate its long-term effects on pelvic floor integrity, sexual function, and surgeon proficiency

Yet it is an unspoken truth that Kosovo needs a compromise with Serbia to receive international recognition. Turkey can contribute to bridging relations between Belgrade and Pristina by identifying ways of cooperation on technical issues concerning the living standards of all Kosovar citizens without getting entangled on the issue of recognition. Turkey can play a complimentary and even crucial role that could actually ease the task of the European Union between Serbia and Kosovo

    Energy Efficient Feature Extraction for Single-Lead ECG Classification Based On Spiking Neural Networks

    Cardiovascular diseases are the leading cause of death in the devel- oped world. Preventing these deaths, require long term monitoring and manual inspection of ECG signals, which is a very time consum- ing process. Consequently, a wearable system that can automatically categorize beats is essential.Neuromorphic machines have been introduced relatively recently in the science community. The aim of these machines is to emulate the brain. Their low power design makes them an optimal choice for a low power wearable ECG classifier.As features are crucial in any machine learning system, this thesis aims at proposing an energy efficient feature extraction algorithm for ECG arrhythmia classification using neuromorphic machines. The feature extraction algorithm proposed in this thesis consists of the merger of a low power feature detection and a feature selection algorithm. Also, different network configurations have been investigated to achieve classification using an LSM architecture. The resulting system can accurately cluster seven beat types, has an overall classification rate of 95.5%, and consumes an estimate of 803.62 nW.Electrical Engineering | Circuits and System

    The purpose of this thesis is to explain the process of policy change in public financial management in Turkey. It looks into the interaction between endogenous and exogenous factors affecting the evolution of policy principles and programmes and uses a policy transfer approach. The author compares two periods: the adjustment-oriented (1980-1999) and accession-oriented (1999 onwards) reform eras. Considering the IFIs and the EU as proxies for globalization and Europeanization, the thesis also examines the impact of these institutions on the evolution of the good governance agenda to draw generalizable conclusions regarding the relationship between globalization and Europeanization. The main findings of the study are: first, accession-oriented reform enabled the policy transfer process and resulted in structural changes in public financial management in line with good governance principles. Although conditionality had been more coercive under adjustment-oriented reform, change had remained limited due to the ability of endogenous actors to dominate the scope and pace of change. Secondly, although economic crises triggered reform initiatives in both periods, the crucial factors accounting for transformative policy change in the second period were a) the consolidation of a reformist advocacy coalition, b) the strengthening of civil society and market forces vis-à-vis the state and c) the emergence of the EU as an anchor for change. Finally, the thesis demonstrates that the good governance agenda is an area of ideational and empirical policy convergence between globalization and Europeanization.
