The role of NOTCH2 gene in human malignant glial brain tumours
Background: Glioblastoma multiforme (GBM), astrocytoma (A) and oligodendroglioma (OG)
are the neoplasms of the glial lineage in the Central Nervous System (CNS). Among them, GBM
occurs at the highest frequency and shows the shortest patient median survival time of some 10
months as compared for instance to the survival time of OG of about 10 years. Genetically, OG
differs from GBM by the frequent combination of loss of heterozygosity (LOH) on chromosomes
1p and 19q, which is associated with more favourable prognosis in OG patients. However, the
clinical significance of LOH on 1p in other glioma subtypes remained unknown.
Methods and Results: We identified a subgroup of GBM with LOH on centromeric chromosome
1p together with longer survival. The minimally lost area(s) in both GBM and OG converged at
the NOTCH2 locus on 1p11 and positively correlated with prognosis in GBM as well as in OG
patients. Comparison between gene expression of NOTCH2 and the genetic status at the NOTCH2
locus on chromosome 1p11 supported the hypothesis of a loss of function alteration of NOTCH2
in tumours. However, many GBMs do not display deletions at the NOTCH2 locus on 1p11 and do
express the NOTCH2 gene. Abundant expression of components of canonical NOTCH signaling
in these tumors and a positive correlation between NOTCH2 transcripts with the target gene
HES-1 (P=0.0001) indicated that functional NOTCH signaling in glioma is mainly driven by
NOTCH2. In addition, we defined TNC, the gene for the cell migration factor tenascin-C as a
novel target gene for NOTCH signaling. We further showed that activation of NOTCH signaling
was indeed promoting TNC-dependent glioma cell motility. Thus, together with the ability to
increase proliferation, canonical Notch signaling turned out to be critical for glioma progression.
We also found that non-canonical Notch signaling was associated with the maintenance of
tumorigenic potential of the GBM cells in soft agar culture. In addition, Notch2 had a prosurvival
effect on GBM cells by upregulating anti-apoptotic proteins Bcl-2 and Mcl-1,
independently of the canonical pathway. Finally, defective degradation pathway of Notch
receptors in GBM cells led to slow receptor turnover, thereby providing additional contribution to
the oncogenic function of Notch2.
Conclusion: This study identified aberrant multi-facetted oncogenic behaviours of Notch
proteins, in particular of Notch2, in GBM. This provided a molecular basis for the higher
aggressiveness of Notch2-positive GBM compared to Notch2-negative GBM or OG, and
suggested Notch2 as a sensible target for new therapeutic approaches against GBM

Disfluency in dialogue:an intentional signal from the speaker?
Disfluency is a characteristic feature of spontaneous human speech, commonly seen as a consequence of problems with production. However, the question remains open as to why speakers are disfluent: Is it a mechanical by-product of planning difficulty, or do speakers use disfluency in dialogue to manage listeners' expectations? To address this question, we present two experiments investigating the production of disfluency in monologue and dialogue situations. Dialogue affected the linguistic choices made by participants, who aligned on referring expressions by choosing less frequent names for ambiguous images where those names had previously been mentioned. However, participants were no more disfluent in dialogue than in monologue situations, and the distribution of types of disfluency used remained constant. Our evidence rules out at least a straightforward interpretation of the view that disfluencies are an intentional signal in dialogue

When do special interests run rampant ? disentangling the role in banking crises of elections, incomplete information, and checks and balances
The author investigates the political determinants of government decisions that benefit special interest groups - especially government decisions to deal with banking crises. He finds that the better informed the voters, the more proximate elections, and the larger the number of political veto players ( conditional on the costs to voters of relevant policy decision), the smaller the government's fiscal transfer are to the financial sector and the less likely the government is to exercise forbearance in dealing with insolvent financial institutions. The results suggest that policies thatmight be appropriate for mitigating banking crises in the United States might be less effective in settings where voters are less informed, where elections are less competitive, and where there are fewer veto players, because in these settings checks and balances are missing. These policies include: a) Disseminating information about the costs of inefficient government decisions. b) Improving the structure of legislative regulatory oversight. c) Intervening early in insolvent banks. The author concludes that the more veto players there are, the less likely policies are to favor special interest groups (contrary to previous views). Moreover, the closer the elections, the less likely policies are to favor special interest groups.

Cyclin D1 immunohistochemical expression and somatic mutations in canine oral melanoma
Canine oral melanoma (COM) is the most frequent tumour with oral localization in dogs. Copy number gains and amplifications of CCND1, a gene coding for Cyclin D1, are the most frequent chromosomal aberrations described in human non-UV induced melanomas. Twenty-eight cases of COM were retrieved from paraffin-blocks archives. A total of 4 μm thick sections were immunostained with an antibody against human Cyclin D1 and Ki-67. Cyclin D1 and Ki-67 expressions were scored through two counting methods. DNA was extracted from 20 μm thick sections of formalin-fixed paraffin-embedded blocks. Pathological and surrounding healthy tissue was extracted independently. Cyclin D1 immunolabelling was detected in 69% (18/26) while Ki-67 was present in 88.5% (23/26) of cases. Statistical analysis revealed correlation between two counting methods for Cyclin D1 (r = 0.54; P = .004) and Ki-67 (r = 0.56; P = .003). The correlation found between Ki-67 and Cyclin D1 indexes in 16/26 cases labelled by both antibodies (r = 0.7947; P = .0002) suggests a possible use of Cyclin D1 index as prognostic marker. Polymerase chain reaction analysis on CCND1 coding sequence revealed the presence of nine somatic mutations in seven samples producing synonymous, missense and stop codons. Since none of the single-nucleotide polymorphisms was found to be recurrent, it is suggested that overexpression of Cyclin D1 may be the consequence of alterations of CCND1 upstream regions or other genetic aberrations not detectable with the methodology used in this study. Future studies are needed to verify the potential use of Cyclin D1 index as prognostic indicator and to highlight the molecular events responsible for Cyclin D1 overexpression in COMs
