Local fabric density in L2 vertebral body bone by high contrast resolution x-ray microtomography

Many x-ray microtomography.(XMT) studies of human vertebral bone samples have been conducted, but none with high quality, high contrast resolution methodology, which necessarily requires long integration times and, therefore, stable samples. We accessed a collection of 69 ~2mm thick, parasagittal sections of L2 vertebral bodies which were preserved - by embedding in PMMA - under the aegis of the BioMed 1 European [long before Brexit] project [Bone quality in osteoporosis]. Prior studies using quantitative backscattered electron [qBSE-SEM] imaging had shown a wide spread of local mineralisation density values when sampled at one cubic micrometer [1 fl] resolution, but only in 2D section planes. To acquire 3D data, we used the QMUL MuCAT2 TDI microtomography system at 30 micron voxel size, 90kV, typically 72h scan time, each scan corrected for beam-hardening and calibrated with a multi-metal calibration carousel: Linear Attenuation Coefficient (LAC) accuracy better than 2%. Results shown are expressed in LAC (cm-1), equivalence to assumed HA concentration. Analysis using ImageJ Fiji. To avoid partial volume artefacts, we stripped, in 3D, one voxel from all free bone surfaces. We compared distributions for the whole bone slab and regions selected to contain only internal trabecular bone, avoiding cortices and end plates. Both showed extreme ranges, but whole slab values contained a higher proportion of highest density voxels. These results are in complete agreement with the earlier qBSE-SEM data which showed the highest values in calcified cartilage in end plates and calcified ligamentous inclusions and Sharpey fibre bone rich regions in cortices, NB the anterior cortex. 'Bone' in the vertebral body is not one tissue but has a variety of fabrics and fabric densities. David Zimmer Christmas letter
This Christmas letter written November 30, 1999, by David Zimmer is titled "Season's Greetings from the last of the Red-Hot-Santas!" It features an illustration of Santa Claus with a guitar, and a summary of Zimmer's year.
David Zimmer (1929-2005) was born in Harrisburg, Ohio. He enlisted in the U.S. Army and served for two years during the Korean War at the Brooke Army Medical Center in San Antonio, where he performed in drag for wounded soldiers. After the war, he returned to Ohio. Zimmer performed as Dolly Divine, a name inspired by the song "Hello Dolly." In 1964, he established the Berwick Ball with Orn Huntington, another important early gay activist in Central Ohio. The Ball began as a formal Halloween costume ball that provided a safe space to gather and enjoy drag shows for the gay community each year; over the years, it grew into an annual Halloween tradition and an important fundraiser for the AIDS movement and other charities. During the 1970s, Zimmer was also known for hosting lavish parties at his Harrisburg home. In 1989, he moved to the German Village area of Columbus where he remained active in the community. During the 1990s, Zimmer continued to perform in and out of drag and commissioned costume designer Dick Frank to make elaborate outfits. Zimmer worked for Huntington National Bank for 39 years and was a member of the Harrisburg United Methodist Church, Veterans of Foreign Wars and the German Village Society

Prospective isolation of human bone marrow stromal cell subsets: a comparative study between Stro-1-, CD146- and CD105-enriched populations
Stro-1 has proved an efficacious marker for enrichment of skeletal stem and progenitor cells although isolated populations remain heterogeneous, exhibiting variable colony-forming efficiency and osteogenic differentiation potential. The emerging findings that skeletal stem cells originate from adventitial reticular cells have brought two further markers to the fore including CD146 and CD105 (both primarily endothelial and perivascular). This study has compared CD146-, CD105- and Stro-1 (individual and in combination)-enriched human bone marrow stromal cell subsets and assessed whether these endothelial/perivascular markers offer further selection over conventional Stro-1. Fluorescent cell sorting quantification showed that CD146 and CD105 both targeted smaller (2.22% ± 0.59% and 6.94% ± 1.34%, respectively) and potentially different human bone marrow stromal cell fractions compared to Stro-1 (16.29% ± 0.78%). CD146+, but not CD105+, cells exhibited similar alkaline phosphatase-positive colony-forming efficiency in vitro and collagen/proteoglycan deposition in vivo to Stro-1+ cells. Molecular analysis of a number of select osteogenic and potential osteo-predictive genes including ALP, CADM1, CLEC3B, DCN, LOXL4, OPN, POSTN and SATB2 showed Stro-1+ and CD146+ populations possessed similar expression profiles. A discrete human bone marrow stromal cell fraction (2.04% ± 0.41%) exhibited positive immuno-labelling for both Stro-1 and CD146. The data presented here show that CD146+ populations are comparable but not superior to Stro-1+ populations. However, this study demonstrates the critical need for new candidate markers with which to isolate homogeneous skeletal stem cell populations or skeletal stem cell populations which exhibit homogeneous in vitro/in vivo characteristics, for implementation within tissue engineering and regenerative medicine strategies
