Isoflavoni e proteina in soia. Valutazione di varietà in differenti condizioni di gestione agronomica.
The soybean can to be a top crop for the health environment. It's a crop which fixes the atmospheric nitrogen in his tissues. The nitrogen is fixed by nitrogenase-reductase to produce high biological value Organic compound. A part of these substances remains in the soil and can to enrich the soil fertility. (organic-N availability).In the Padua University Experimental Farm, a research work has been realized for to evaluate the nutraceutical compounds production of the most important soybean varieties diffused in Italy. The nutraceutical compounds examined are isoflavones and proteins.
The isoflavones and the soybean proteins are the positive effects on the human health if they are firmly integrated in the diet. (Cancer prevention, menopause symptom alleviation, reduction of the cholesterol accumulation) (Lacombe S et al 2000).
This work has concerned the study of the 35 and 28 soybeans varieties during the 2005. 2006 and 2007. The varieties are cultivated in two different ways:
a) biological way
b) traditional way
The production of the isoflavones are been evaluated in the different plant parts (germ and cotyledon) with a simple HPLC analysis method. A variability coefficient >27% has been found. A different varieties vocation for the soyfood productions has been underlined.
Some varieties are good for the nutraceutical production (high contents in isoflavone compound), some varieties are good for the low content isoflavone production (baby meal).
The agronomic variables have influenced the isoflavone concentration in the soybean seeds In the 2006 and 2007 a second experiment has been realized for to study the irrigation effect, the nitrogen input effect, the soil type effect, on the isoflavone and proteins production. For this study two different varieties has been chosen: Ales (high isoflavone concentration low protein concentration) and Nikir (high protein concentration and low isoflavone concentration). These varieties are grown in the water optimal availability condition and they have compared with a control non irrigated.
By the results is it possible to see witch the ETM condition determine a major isoflavone concentration,(+11%) in the seeds in comparison with the control non irrigated and fertilized.
This effect is more evident in the seeds produced on the stem ramification
Estimation of cotyledon isoflavone abundance by a grey luminance-based model in variously hilum-coloured soybean varieties
BACKGROUND: The nutraceutical uses of soybean (Glycine max L. Merr.) have received increasing attention in recent years, due to the therapeutic effects of high seed isoflavone concentrations against heart disease, cancer and menopausal symptoms.
RESULTS: We found a close correlation between seed isoflavone abundance and hilum colour in a set of 17 contrasting soybean varieties. Image analysis of the hilum greylevel pattern allowed us to identify a power model which approximates total cotyledon isoflavone concentrations (TCIC) at 65–71% by the normalised modal grey level. Higher TCIC levels were assigned to darker hilum varieties and vice versa within a variety-dependent response. Optimisation of the algorithm required correction for a few specific varieties falling in the intermediate 1.1–1.5 mg/g TCIC range, which were over-estimated by the model, perhaps due to variations in hilar optical properties related to the geometric features of both hilum and seed.
CONCLUSION: In view of its easy, low-cost detection, seed hilum colour is a useful phenotypic trait in soybean for rapid evaluation of isoflavone abundance in food uses and for improving specific nutraceutical breeding programmes
Effetti dell’irrigazione sulla produzione di semente di soia a destinazione nutraceutica
L’inserimento della soia negli avvicendamenti colturali italiani può parzialmente rispondere ad una crescente esigenza di salvaguardia dell’ambiente e di crescente domanda di prodotti nutraceutici.
Negli anni 2012 e 2013 è stata svolta una sperimentazione allo scopo di individuare, per alcune varietà particolarmente interessanti per caratteristiche nutraceutiche, i protocolli di coltivazione adatti alla produzione di semente da destinare alla semina, con migliorata attitudine alla concentrazione di isoflavoni.
La sperimentazione si è svolta in vaso in ambiente controllato (2012) e all’aperto (2013). Nel primo anno sono state coltivate tre varietà (Dekabig-optimize, Pacific, Sponsor) sottoposte a tre livelli di irrigazione (capacità di campo, 50% A.D., 10% A.D.). Nel 2013 i semi delle tre varietà raccolti nella sperimentazione precedente sono stati coltivati con apporti idrici naturali in quattro diversi terreni (argilloso, medio limoso, sabbioso e torboso), al fine di valutare l’effetto del trattamento agronomico del primo anno sulla seconda generazione. La concentrazione di isoflavoni nei cotiledoni è stata quantificata tramite metodica HPLC.
In regime di irrigazione intermedio (50% A.D.) si è ottenuta la massima concentrazione di isoflavoni, mentre con apporti irrigui alla capacità di campo l’elevata dimensione dei semi ha avuto un effetto diluizione. L’apporto idrico in prima generazione ha avuto un effetto significativo sulle piante coltivate in seconda riproduzione. Si è potuto infatti accertare che, la coltivazione in presenza di una elevata disponibilità idrica (mantenimento alla capacità di campo) influenza positivamente il contenuto di isoflavoni in seconda generazione per tutte le varietà in sperimentazione, particolarmente nei terreni medio-limoso e argilloso.
Si può quindi concludere che adeguati apporti irrigui, anche attraverso sistemi ad elevata efficienza come la manichetta forata, siano condizione necessaria per l’ottenimento di semente certificata da destinare ad una filiera soia ad uso nutraceutico. In relazione alle caratteristiche morfologiche dei semi ottenuti in terreno torboso – di elevate dimensioni e con contenuti intermedi di isoflavoni – similmente a quanto osservato nel primo anno di sperimentazione con elevati apporti irrigui, è lecito attendersi che la produzione di semente in questo tipo di terreno possa soddisfare le esigenze specifiche di questo tipo di filiera
Effetto ammendante del biochar in maiscoltura
Il biochar è il sottoprodotto risultante dalla pirolisi di biomasse vegetali a temperature molto elevate e limitata disponibilità di ossigeno. L’interesse crescente verso questa modalità di sfruttamento energetico delle biomasse ha accresciuto gli studi sul possibile reimpiego di tale materiale. Il biochar potrebbe essere, infatti, favorevolmente impiegato come ammendante dei terreni agrari in virtù del suo apporto di carbonio e nutrienti, anche se alcune sue caratteristiche chimiche (es. presenza di metalli pesanti, pH, salinità) ne potrebbero limitarne l’utilizzazione agronomica.
Nel corso del 2014 presso l’azienda agraria sperimentale Lucio Toniolo dell’Università di Padova a Legnaro, sono stati studiati gli effetti dell’applicazione di biochar derivante da biomasse legnose al dosaggio dell’1% (circa 26 t/ha) incorporato nei primi 20 cm di terreno con l’erpicatura poco prima della semina di due ibridi di mais (classe 700), caratterizzati da una diversa sensibilità allo stress idrico.
Il biochar possedeva pH alcalino, una moderata conducibilità elettrica ed un contenuto di metalli pesanti inferiore ai limiti di legge previsti per i fertilizzanti organici, ad eccezione del Ni (135 vs. 100 mg kg-1 ammessi). Nel terreno ammendato è stato osservato un aumento della biodisponibilità di Ni, ma anche di Cu e Zn a cui è tuttavia corrisposta una riduzione della concentrazione di Cu e Zn nei residui colturali e invarianza nella granella; sono invece diminuiti sensibilmente i valori di P e S in entrambe le strutture vegetali.
La stabilità dei parametri vegetativi (es. altezza delle piante, contenuto di clorofilla fogliare) e produttivi (resa, peso specifico, tenore proteico e di amido) conseguenti all’applicazione del biochar, ne suggerisce un valido impiego in maiscoltura. Il miglioramento dell’accrescimento radicale, che è stato osservato nell’ibrido più sensibile allo stress idrico, suggerisce di approfondire lo studio sull’efficacia di questo ammendante anche in regimi di limitata disponibilità idrica, cosa che non è stato possibile accertare nel corso del 2014 a causa dell’elevata piovosità estiva
Soybean isoflavone patterns in main stem and branches as affected by water and nitrogen supply
Several isoflavones, such as genistein, daidzein, and their glucosyl and malonyl forms were detected
by HPLC–UV in cotyledons, together with glycitein forms in the hypocotyl of two 1-maturity class contrasting
soybean varieties (Ales with high isoflavone and low protein concentrations vs. Nikir, showing
opposite behaviour). Cultivation involved drip irrigation and rainfed conditions, factorially combined
with two levels of nitrogen supply (0 and 100 kg ha−1 at R1–R3 stages) at Legnaro (NE Italy) for two years
(2006–2007). Yield components and hypocotyl and cotyledon isoflavone concentrations were measured
in stems and branches separately.
Ales was confirmed as having a higher total cotyledon isoflavone concentration (TCIC) than Nikir, with
higher fractions of total daidzein in cotyledons and total glycitein in hypocotyl. In 2007, a 40% decline in
total hypocotyl isoflavone concentration (THIC) was observed compared with 2006, due to a higher daily
range temperature regime during seed filling.
Irrigation increased seed yield and protein, but its effect on TCIC was negligible. Irrigation reduced THIC
(∼8%) in the drier year (2007), perhaps due to greater hypocotyl weight (dilution effect) and lower fungal
disease attack. TCIC and THIC were generally increased by the unusual nitrogen fertilisation practice (TCIC
in year 2: 1.85 vs. 1.58 mg g−1), especially in Nikir.
The most important finding was a 24.4% cotyledon (1.54 vs. 1.96 mg g−1) and 4.2% hypocotyl (14.9 vs.
14.3 mg g−1) greater isoflavone concentration in seeds of branches compared with those of the main stem,
and this may have a practical application in sowing density.
It is currently concluded that nitrogen fertilisation improves the nutraceutical properties of soybean,
although its effect depends on the activity of the isoflavone biosynthetic pathway and the concurrent
extent of seed bulging
Nitrogen fertilization and irrigation compulsory effect on soybean (Glycine max L. Merr) isoflavone
Intraspecific variability for soybean cotyledon isoflavones in different cropping and soil condidtions.
Cotyledon isoflavones, such as genistein, daidzein and their glucosyl and malonyl forms, were detected by HPLC-UV in 18 0- and 1-maturity class soybean varieties, cultivated under organic and conventional farming (3 years) and various soils (2 years) at Legnaro, NE Italy. Among varieties, the overall coefficient of variation for total cotyledon isoflavone concentration (TCIC) was 22%, and cluster analysis revealed three variety groups. Group L had 1.22 mg g(-1) TCIC (low) and 40.3% proteins: group H, mainly including black-hilum varieties, 2.10 mg g(-1) (high) and 39.2%, and group 11.77 mg g(-1) (intermediate) and 39.4%. Isoflavone profiles were similar in groups L and H (55% genistein and 45% daidzein aglycone equivalents), whereas group I had increased genistein (61%), which is recognised to have lower intestinal bioavailability (nutraceutics). Under organic management, a 2% protein increase (all groups) was associated with a 5-15% variety-dependent decrease - in groups H and I - and stable TCIC in group L Isoflavone accumulation was positively affected by rainfall and limited daily range temperature regimes during pod filling, and hail damage (year 2005). In the Nikir variety (maturity class 1), grown in four lysimeter soils, TCIC was negatively correlated with soil C/N ratio, partially explaining the worse TCIC in organic cultivation. Based on this relationship, TCIC followed the order clay-loam > silty-loam > organic > sandy, with a 64% difference between extremes
