Pharmacogenetics of ophthalmic topical β-blockers
Glaucoma is the second leading cause of blindness worldwide. The primary glaucoma risk factor is elevated intraocular pressure. Topical β-blockers are affordable and widely used to lower intraocular pressure. Genetic variability has been postulated to contribute to interpersonal differences in efficacy and safety of topical β-blockers. This review summarizes clinically significant polymorphisms that have been identified in the β-adrenergic receptors (ADRB1, ADRB2 and ADRB3). The implications of polymorphisms in CYP2D6 are also discussed. Although the candidate-gene approach has facilitated significant progress in our understanding of the genetic basis of glaucoma treatment response, most drug responses involve a large number of genes, each containing multiple polymorphisms. Genome-wide association studies may yield a more comprehensive set of polymorphisms associated with glaucoma outcomes. An understanding of the genetic mechanisms associated with variability in individual responses to topical β-blockers may advance individualized treatment at a lower cost
The "Herkules" - novel of Andreas Heinrich Bucholtz.
Bucholtz "Herkules"-novel is an extensive, rambling work of 1920 quarto pages and barely readable nowadays. It was very popular for the first 130 years, however, and went through seven editions and adaptations. The present study tries to account for the work's initial popularity, but at the same time emphasises that "Herkules" has many features which make it strangely antiquated even for the century in which it was written. This study further attempts to show how "Herkules" is composed of many heterogeneous elements which had accumulated from antiquity onwards. It examines some of the principal components of these literary traditions apparent in the form, content and style of the work, demonstrating how they are exploited by the author for purposes of edification, entertainment and emotive effect("prodesse", "delectare" and "movere") and to what extent they are transmuted by the cultural, social and confessional background of the author's upbringing and profession. Chapter I reviews the judgements passed on "Herkules", from the clearly positive ones of contemporaries like Rist and Schottel down to the mainly negative ones of the principal modern historians of literature. It draws attention to several shortcomings inherent in such appraisals and suggests a re-examination of certain elements in "Herkules" which hark back to the literary traditions of previous ages. Chapter II proposes a re-appraisal of the work in another respect, vis: in the framework of the concept of literary art then obtaining, stressing in particular the artefact quality and the factors of edification, entertainment and emotive effect. Chapters III end IV examine the work's links with popular late-medieval and 16th century literature and with the late-Greek novel respectively. Chapter V seeks to explain the diversity of styles in "Herkules" by the 17th century modification of a three-style technique ultimately traceable to classical antiquity, and suggests a co-ordination with the "prodesse-delectare-movere" function and the Humanist-Protestant-burgher milieu factors
Mutations in CPAMD8 Cause a Unique Form of Autosomal-Recessive Anterior Segment Dysgenesis
Anterior segment dysgeneses (ASDs) comprise a spectrum of developmental disorders affecting the anterior segment of the eye. Here, we describe three unrelated families affected by a previously unclassified form of ASD. Shared ocular manifestations include bilateral iris hypoplasia, ectopia lentis, corectopia, ectropion uveae, and cataracts. Whole-exome sequencing and targeted Sanger sequencing identified mutations in CPAMD8 (C3 and PZP-like alpha-2-macroglobulin domain-containing protein 8) as the cause of recessive ASD in all three families. A homozygous missense mutation in the evolutionarily conserved alpha-2-macroglobulin (A2M) domain of CPAMD8, c.4351T>C (p. Ser1451Pro), was identified in family 1. In family 2, compound heterozygous frameshift, c.2352_2353insC (p.Arg785Glnfs(∗)23), and splice-site, c.4549-1G>A, mutations were identified. Two affected siblings in the third family were compound heterozygous for splice-site mutations c.700+1G>T and c.4002+1G>A. CPAMD8 splice-site mutations caused aberrant pre-mRNA splicing in vivo or in vitro. Intriguingly, our phylogenetic analysis revealed rodent lineage-specific CPAMD8 deletion, precluding a developmental expression study in mice. We therefore investigated the spatiotemporal expression of CPAMD8 in the developing human eye. RT-PCR and in situ hybridization revealed CPAMD8 expression in the lens, iris, cornea, and retina early in development, including strong expression in the distal tips of the retinal neuroepithelium that form the iris and ciliary body, thus correlating CPAMD8 expression with the affected tissues. Our study delineates a unique form of recessive ASD and defines a role for CPAMD8, a protein of unknown function, in anterior segment development, implying another pathway for the pathogenicity of ASD
A Systematic Review of Online Sex Addiction and Clinical Treatments Using CONSORT Evaluation
Researchers have suggested that the advances of the Internet over the past two decades have gradually eliminated traditional offline methods of obtaining sexual material. Additionally, research on cybersex and/or online sex addictions has increased alongside the development of online technology. The present study extended the findings from Griffiths’ (2012) systematic empirical review of online sex addiction by additionally investigating empirical studies that implemented and/or documented clinical treatments for online sex addiction in adults. A total of nine studies were identified and then each underwent a CONSORT evaluation. The main findings of the present review provide some evidence to suggest that some treatments (both psychological and/or pharmacological) provide positive outcomes among those experiencing difficulties with online sex addiction. Similar to Griffiths’ original review, this study recommends that further research is warranted to establish the efficacy of empirically driven treatments for online sex addiction
