    Existen secuencias genéticas que podrían asociarse a un mejor desempeño atlético, tales como los polimorfismos I/D en el gen ACE y R577X en el gen ACTN3, que se relacionan a fenotipos de mayor fuerza muscular y resistencia. El propósito de este trabajo fue iniciar un estudio exploratorio en dos polimorfismos, ACE (rs1799752) y ACTN3 (rs1815739) para determinar su presencia y evaluar si existe asociación con variables antropométricas y de rendimiento según la especificidad/especialidad deportiva en 37 atletas de fondo y fuerza-velocidad de Coldeportes. Se tomó sangre total para extracción de ADNg, cuantificación y amplificación por PCR convencional para genotipificar atletas y controles. Se encontró asociación moderada entre el polimorfismo del gen ACE y los grupos de fuerza-velocidad, resistencia y grupo control; sin embargo, el polimorfismo del gen ACTN3 no tuvo una asociación significativa. Los resultados de este estudio podrían contribuir a la caracterización molecular de atletas y optimizar las metodologías de identificación talentos deportivos a temprana edad.There are genetic sequences that could be associated with better athletic performance, such as the I/D polymorphisms in the ACE gene and R577X in the ACTN3 gene, which are related to phenotypes of greater muscular strength and endurance. The purpose of this work was to initiate an exploratory study in two polymorphisms, ACE (rs1799752) and ACTN3 (rs1815739) to determine their presence and assess whether there is an association with anthropometric and performance variables according to sport specificity / specialty in 37athletes of background and strength-speed of Coldeportes. Whole blood was taken for extraction, quantification and amplification of cDNA by conventional PCR to genotyping athletes and controls. A moderate association was found between the ACE gene polymorphism and the force-speed, resistance and control group; however, the polymorphism of the ACTN3 gene did not have a significant association. The results of this study could contribute to the molecular characterization of athletes and optimize methodologies to identify sports talents at an early age

    El rotavirus es la principal causa de diarrea en niños menores de 5 años de edad y en países en vía de desarrollo ocasiona 453 000 millones de muertes al año. Aunque existen actualmente en el mercado dos vacunas de rotavirus vivos atenuados (Rotarix ™) y (RotaTeq ™) que disminuyen los síntomas asociados a la infección, se ha observado que estos no desaparecen completamente y aún se presentan hospitalizaciones debidas a la infección por rotavirus. Por tal motivo en el Laboratorio de Biología Molecular de Virus de la Universidad Nacional de Colombia se buscan alternativas terapéuticas que permitan la reducción de la infección, mediante el estudio de moléculas celulares como ROS, NF-kB, PPAR-γ y COX-2 las cuales inducirían una respuesta inflamatoria en la célula huésped como mecanismo de defensa ante el ataque del virus. Se ha encontrado que el estímulo de PPAR-γ con el fármaco agonista Pioglitazona y el tratamiento con antioxidantes como N-acetilcisteína (NAC) reduce la infección, sin embargo no se ha determinado si este efecto es debido a la acción directa de los fármacos sobre el virus o a la activación de PPAR-γ y por consiguiente de vías pro-inflamatorias. Este trabajo tuvo por objetivo determinar el efecto de los agonistas de PPAR-γ Pioglitazona, Tiazolidinediona, Rosiglitazona, Ácido linoléico y Ácido docahexaenóico y del Ácido retinoico sobre la expresión de PPAR-γ, IκB, NF-κB y COX-2 usando como modelo celular vellosidades aisladas de ratón adulto ICR infectadas con rotavirus ECwt, las cuales fueron aisladas, infectadas y luego tratadas o no con los agonistas de PPAR-γ y el ácido retinoico. Tanto la infección, como la expresión de las proteínas celulares se evaluó utilizando inmunocitoquímica, inmunofluorescencia, Western Blot y ELISA de captura. En todos los casos, los niveles de expresión de proteína se compararon con sus correspondientes controles no tratados. Se determinó que la infección por rotavirus en vellosidades aisladas de ratón ICR aumenta la expresión de PPAR-γ, NF-κB, IκB, COX-2 y Hsc70 y que los agonistas de PPAR-γ y el Ácido retinoico disminuyen la infección causada por rotavirus en vellosidades aisladas de ratón ICR y modifican la expresión (retornando a sus niveles basales) de las proteínas PPAR-γ, NF-κB, IκB, COX-2 y Hsc70.Rotavirus is the principal cause of diarrhea children under 5-year-old minor of age and in developing countries it causes 453 000 million deaths per year. For such a motive in the Laboratory of Molecular Biology of Virus of the National University of Colombia there are looked therapeutic alternatives that allow reduction of the infection, trough the study of cellular molecules like ROS, NF-kB, PPAR-γ And COX-2 which would induce an inflammatory response in the host cell as mechanism of defense before the attack to the virus. It has be found that stimulation of PPAR-γ with the medicament agonista Pioglitazona and the treatment with antirust as N-acetilcisteína (NAC) reduces the infection, nevertheless it has not decided if this effect is due to the direct action of the medicaments on the virus or to PPAR's activation and consequently of pro-inflammatory routes. The objective of this Word has been determine the effect of PPAR's agonist Pioglitazona, Tiazolidinediona, Rosiglitazona, linoleic acid on the expression of PPAR-γ, IκB, NFκB and COX-2 use as cellular model villi of adult mouse ICR infected with rotavirus ECwt, which were isolated, infected and then treated or not with PPAR's agonistas and the retinoic acid. The infection and the expression of the cellular proteins was evaluated using inmunocitochemistry, inmunofluorescence, Western Blot and ELISA of capture. In all the cases, the levels of expression of protein were compared with his corresponding controls not agreements. It was determined thah rotavirus infection on insolated villi of ICR adult mouse increases the expression of PPAR-γ, IκB, NFκB, COX-2 and Hsc70.     Standardization of a microspectrophotometric method for the determination of the enzymatic activity of Phosphomanomutase 2 in a control sample from Bogotá Colombia

    La deficiencia en la glicosilación PMM2-CDG (CDG Ia) es una enfermedad autosómica recesiva, causada por el daño de la enzima citoplasmática fosfomanomutasa 2 (E.C.5.4.2.8), la cual cataliza la conversión de manosa-1-fosfato a manosa-6-fosfato, indispensable para la N-glicosilación. Esta enfermedad es de orden multisistémica caracterizado por fallos neurológicos, digestivos, nefrológicos, hepáticos, hematológicos y cardíacos. En Colombia no hay laboratorios clínicos que realicen la prueba enzimática para el diagnóstico como tampoco hay estudios que reporten intervalos de actividad enzimática, por lo cual el objetivo de este estudio fue establecer los valores de referencia de la actividad específica de la PMM2. El método diagnóstico microespectrofotométrico fue estandarizado con 50 muestras de controles voluntarios, para ello se extrajeron los leucocitos mediante el método dextrán-heparina a partir de sangre total, se sonicaron para extraer la enzima de interés y se cuantificó el contenido de proteína por el método de Folin-Lowry. Para determinar la integridad de las muestras se usó como control la enzima lisosomal N-Acetilgalactosamina-6-sulfatosulfatasa. Los análisis estadísticos permitieron comprobar que no existen diferencias significativas entre género ni edad al encontrar un valor-p 0,12 y 0,6537 respectivamente, bajo un nivel de significancia de 0,05. Se encontró un valor de actividad específica en el límite superior mayor a lo reportado con respecto a otras investigaciones. El valor de referencia de actividad enzimática determinado en este estudio fue de 6,546-48,023 nmol/h. mg de proteína. Los valores de actividad obtenidos en este grupo control permitirán iniciar el diagnóstico de pacientes deficientes de PMM2 desde edad neonatal y contribuir así con el sistema de salud colombiano, como también dar un manejo acertado por parte del personal médico, como de la familia y así mejorar la calidad de vida de estos pacientes.Deficiency in glycosylation PMM2-CDG (CDG Ia) is an autosomal recessive disease, caused by damage of the cytoplasmic enzyme phosphomanomutase 2 (E.C.5.4.2.8), which catalyzes the conversion of mannose-1-phosphate to mannose-6 -phosphate, essential for N-glycosylation. This disease is multisystemic in nature characterized by neurological, digestive, nephrological, hepatic, hematological and cardiac failures. In Colombia there are no clinical laboratories that perform the enzymatic test for diagnosis, nor are there studies that report intervals of enzymatic activity, so the objective of this study was to establish the reference values of the specific activity of PMM2. The microspectrophotometric diagnostic method was standardized with 50 samples of voluntary controls, for this the leukocytes were extracted by the dextran-heparin method from whole blood, sonicated to extract the enzyme of interest and the protein content was quantified by the method of Folin-Lowry. To determine the integrity of the samples, the lysosomal enzyme N-Acetylgalactosamine-6-sulfatosulfatase was used as a control. Statistical analyzes allowed us to verify that there are no significant differences between gender or age when finding a p-value 0.12 and 0.6537 respectively, under a significance level of 0.05.     Los defectos de glicosilación tipo Ib (CDG-Ib) son de herencia autosómica recesiva, se presenta por la deficiencia de la enzima citoplasmática fosfomanosa isomerasa (PMI, EC 5.3.1.8). Se caracteriza por fibrosis hepática que conlleva a un fallo hepático, hipoglucemia con hiperinsulinemia, coagulopatías con trombosis y accidentes cerebro vascular en algunos casos, hemorragias, enteropatía con pérdida proteica y desnutrición por diarrea intratable. El objetivo de este estudio fue determinar valores de referencia de la actividad enzimática específica de la PMI en una población colombiana para realizar el diagnóstico de este defecto metabólico. Se estandarizó un método diagnóstico micro espectrofotométrico a partir de 32 muestras, de las cuales se aislaron leucocitos por el método dextran-heparina de sangre completa, luego se lisaron por sonicación, se cuantificó el contenido de proteína por el método Folin-Lowry y la actividad enzimática fue medida mediante tres reacciones acopladas en la cual actividad específica es directamente proporcional a la producción de NADPH+H+. Se realizó para el grupo control análisis estadísticos descriptivos y de normalidad por género y edad con el programa XLSTAT. Los resultados muestran que no existe diferencia significativa de actividad enzimática por género al encontrar un p-valor 0,3954, como tampoco por edad, al hallar un p-valor 0,6788. Se encontró que el valor de actividad específica es mayor a lo reportado con respecto a otros estudios 551-2416 nmol/h. mg de proteína. Este estudio permitirá iniciar el diagnóstico de pacientes deficientes de la PMI de forma temprana y oportuna, para así evitar complicaciones relacionadas con esta enfermedad, ya que ofrece un tratamiento fácil y a bajo costo, al poder suplementar manosa en forma oral. Esto beneficiará al paciente y a su familia mejorando su calidad de vida y al sistema de salud colombiano al generar una relación positiva costo-beneficio, lo que hace que sea una posible enzima candidata para pruebas de tamizaje neonatal.The glycosylation defects type Ib (CDG-Ib) are of autosomal recessive inheritance, this desease is presented by the deficiency of the cytoplasmic phosphomannose isomerase enzyme (PMI, EC 5.3.1.8). It is characterized by hepatic fibrosis that leads to hepatic failure, hypoglycemia with hyperinsulinemia, coagulopathies with thrombosis and cerebrovascular accidents in some cases, hemorrhage, enteropathy with protein-losing and malnutrition due to intractable diarrhea. The objective of this study was to determine reference values of PMI specific enzyme activity in a sample colombian to diagnose this metabolic defect. We analized 32 samples, of which leukocytes were isolated by the dextran-heparin method of whole blood,It was lysed by sonication, the protein content was quantified by the Folin-Lowry method. A micro spectrophotometric diagnostic method was standardized and activity enzymatic activity was measured by three coupled reactions in which specific activity is directly proportional to the production of NADPH+H+. Descriptive and normality statistical analyzes by gender and age were performed for the control group with the XLSTAT program. The results show that there is not significant difference in enzymatic activity by gender on finding a p-value of 0.3954, nor by age p-value of 0.6788. We found that the specific activity value is higher than values reported with respect to other studies 551-2416 nmol / h. mg protein. This study will allow to initiate the diagnosis of patients deficient of the PMI in an early aged and thereby avoid complications related to this disease.This disorder offers an easy treatment and it is low cost with oral mannose supplementation.     Analysis of the elimination of urinary debris from cancer patients treated with the oncolytic virus Wt1-5.

    La viroterapia oncolítica ha surgido como una estrategia prometedora en la investigación contra el cáncer. En este estudio, se evaluó la eliminación del virus oncolítico Wt1-5 en la orina de 5 pacientes con diferentes tipos de cáncer tratados con este virus, mediante la detección de proteínas virales así como su capacidad para infectar células U937 en cultivo. Para ello, se recolectaron muestras de orina en diferentes tiempos post-aplicación (24-504 h), analizándose la presencia de partículas virales mediante SDS-PAGE, Western Blot, inmunofluorescencia, citometría de flujo y ELISA de captura. Se determinó la presencia de proteínas virales en detritus urinario, indicando la excreción del virus por esta vía. Además, se observó que parte del virus eliminado mantiene su capacidad infectiva, al lograr la infección de células U937 in vitro. Los resultados mostraron que la mayor cantidad de partículas virales en orina se detectó entre las 24-96 horas post-aplicación, con una reducción progresiva a partir de las 72 horas, completándose la eliminación en aproximadamente 21 días. La inmunofluorescencia y la citometría de flujo evidenciaron variaciones en la infección celular según el tiempo de recolección de la orina, sugiriendo una eliminación viral dinámica. El ELISA de captura sugiere la presencia de proteínas virales libres, indicando que parte del virus es eliminado en forma de fragmentos no infecciosos. Se concluye que el virus oncolítico Wt1-5 se excreta en la orina y que parte de él conserva su capacidad infectiva, lo que sugiere la necesidad de evaluar su eliminación y persistencia en fluidos biológicos.Oncolytic virotherapy has emerged as a promising strategy in cancer research. In this study, the elimination of the oncolytic virus Wt1-5 in the urine of five cancer patients treated with this virus was evaluated by detecting viral proteins and assessing its ability to infect U937 cells in culture. Urine samples were collected at different post-application times (24–504 h), and the presence of viral particles was analyzed through SDS-PAGE, Western Blot, immunofluorescence, flow cytometry, and capture ELISA. Viral proteins were detected in urinary debris, indicating virus excretion through this route. Additionally, part of the eliminated virus retained its infective capacity, as it successfully infected U937 cells in vitro. Results showed that the highest amount of viral particles in urine occurred between 24–96 hours post-application, with a progressive reduction starting at 72 hours, and complete elimination at approximately 21 days. Immunofluorescence and flow cytometry revealed variations in cellular infection depending on urine collection time, suggesting dynamic viral shedding. The capture ELISA indicated the presence of free viral proteins, suggesting that part of the virus is eliminated as non-infectious fragments. It is concluded that the oncolytic virus Wt1-5 is excreted in urine and that part of it retains infective capacity, highlighting the need to evaluate its elimination and persistence in biological fluids.Universidad Nacional de Colombi

    Determination of interaction with rotavirus WTEW heat shock protein in cytoplasmic membrane cells KATO III

    En el laboratorio de Biología Molecular de Virus de la Universidad Nacional de Colombia se está investigando si rotavirus WWM, TRUY, Wt1-5, O-ECwt y WTEW (generados en el laboratorio), pueden utilizarse como virus oncolítico. Trabajos previos sugieren que los aislamientos rotavirales infectan las líneas celulares tumorales Sp2/0- Ag14 (mieloma murino), U937 (linfoma histiocitico), REH (leucemia linfocítica aguda), PC3 (adenocarcinoma grado IV de próstata). MDA y MCF-7 (adenocarcinoma de mama) Este trabajo tuvo por objetivo determinar si hay o no interacción del rotavirus WTEW con las proteínas de choque térmico Hsp90, Hsp70, Hsp60, Hsp40 durante la entrada a la línea tumoral KATO III (carcinoma de estómago). Para esto se aislaron fracciones enriquecidas de membranas citoplasmáticas, se incubaron con rotavirus o se bloquearon con anticuerpos específicos anti-Hsp (completos o la porción F(ab’)2) y se adicionó rotavirus MOI 8. La interacción se observó mediante técnicas de ELISA de captura, coinmunoprecipitación y Western Blot. Para determinar el papel funcional de las HSPs, las células se incubaron con anticuerpos F(ab’)2 y luego con rotavirus por 12h a 37°C. El rotavirus se incubó con proteínas HSPs completas solubles durante 1h a 37°C y luego el complejo HSPs-rotavirus se adicionó a las células por 12h a 37°C. La infección en ambos casos se evaluó mediante inmunocitoquímica. Finalmente se incubaron las células con el rotavirus WTEW por 1 h a 4°C y luego se le adicionaron los péptidos sintéticos de cada Hsp por 1h a 4°C dejando por 12 h a 37°C en cámara de CO2. La infección se evaluó por ELISA de captura. Se determinó que existe interacción entre rotavirus WTEW con las HSPs de manera directa y funcional ya que al bloquear o alterar dicha unión se disminuye la infección, lo que sugiere que el rotavirus WTEW interacciona con las HSPs de la línea KATO III facilitando el ingreso del rotavirus a la célula.In the Molecular Biology of Virus located at Universidad Nacional de Colombia, the research to determine whether rotavirus WWM, Wt1-5, O-ECwt and WTEW (generated in the laboratory) can be used as oncolytic virus. Preliminary research suggests that rotaviral isolates infected tumor cell lines Sp2/0-Ag14 (murine myeloma), U937 (histiocytic lymphoma), REH (Acute Lymphocytic Leukemia), PC3 (prostate adenocarcinoma grade IV). MDA and MCF-7 (breast adenocarcinoma). This study was aimed to determine whether there was or not WTEW rotavirus interaction with heat shock protein Hsp90, Hsp70, Hsp60, Hsp40 during the entry to the tumor cell line KATO III (gastric carcinoma). For this, fractions enriched with cytoplasmic membranes were isolated, incubated with rotavirus or blocked with specific anti-Hsp (complete or portion F(ab')2) and was added rotavirus MOI 8. The interaction was observed by capture ELISA, co-immunoprecipitation and western blotting. To determine the functional role of HSPs, the cells were incubated with antibody F(ab')2 and then with rotavirus for 12 h at 37 °C. Rotavirus was incubated with soluble proteins HSPs complete for 1 h at 37 °C and then the HSPs rotavirus complex was added to the cells for 12 h at 37 °C. In both cases the infection was assessed by immunocytochemistry. Finally cells rotavirus WTEW incubated for 1 h at 4 °C and then were added the synthetic peptides each Hsp for 1 h at 4 °C leaving 12 h at 37 °C in CO2 chamber. Infection was evaluated capture ELISA. It was determined that there is interaction between the rotavirus WTEW and HSPs was direct and functional manner as to block or alter binding said infection is decreased, suggesting that interacts rotavirus WTEW and HSPs of facilitating the entry of rotavirus to the cell KATO III line

    Los trastornos de comportamiento están caracterizados por la persistencia de síntomas como las agresiones y dificultades en las relaciones familiares, sociales y académicas; las enfermedades relacionadas con estos trastornos en niños, niñas y adolescentes colombianos, son el principal motivo de consulta en salud mental escolar; estudios recientes han establecido asociaciones de algunos marcadores genéticos con trastorno de comportamiento como lo son, el trastorno de hiperactividad y déficit de atención (TDAH), el trastorno obsesivo compulsivo (TOC), el trastorno del espectro autista (TEA). De acuerdo con lo anterior, nuestra investigación tuvo como objetivo realizar un análisis genético molecular y estadístico a partir del estudio de asociación de seis marcadores genéticos (MOBP (rs864643), GPC6 (rs7995215), BTBD3 (rs6131295), DRD4 (VNTR EXON III 48pb), HTTLPR (VNTR Exón XV 44pb) y SLC1A1 (rs301434)). Por lo cual, se analizó una muestra de 50 pacientes infantiles colombianos de 6 a 16 años, con diagnóstico de trastornos de comportamiento, los cuales fueron comparados con 50 controles. El análisis genético se llevó a cabo por PCR (Reacción en cadena de la polimerasa) convencional y por RT-PCR en tiempo real (Sonda TaqMan). El análisis estadístico por medio del programa PLINK, permitió realizar una determinación del equilibrio de Hardy-Weinberg, establecer el p-value y Odd Ratio de asociación de las variantes de estudio. Los resultados obtenidos permitieron concluir que no hay asociación de ninguno de los marcadores analizados con los trastornos de comportamiento infantiles, ya que presentaron p-valores, p > 0,05. Por otro lado, todos los marcadores se encontraron en equilibrio de Hardy Weinberg, excepto SLC1A1. Sin embargo, todos los marcadores presentaron un Odd Ratio mayor a 1 lo que permitió inferir que existe la probabilidad de una asociación significativa con trastorno de comportamiento como TDAH, TOC y TEA; este estudio permitirá el análisis y estudio de marcadores genéticos, y se recomienda ampliar la muestra de población de estudio y así obtener resultados más significativos.Behavioral disorders are characterized by the persistence of symptoms such as aggression and difficulties on family, social relationship, and academics. The diseases related to these disorders in Colombian children and teenagers are the main reason for consultation in mental health at schools. Recent studies have established associations of some genetic markers with the behavioral disorder such as attention deficit hyperactivity disorder (ADHD), obsessive-compulsive (OCD), autistic spectrum (ASD). In concordance with the above, we were how aim to perform a molecular and statistical genetic analysis from the association study of six genetic markers (MOBP (rs864643), GPC6 (rs7995215), BTBD3 (rs6131295), DRD4 (VNTR EXON III 48pb), HTTLPR (VNTR Exon XV 44pb) and SLC1A1 (rs301434)). Therefore, we analyzed a sample of 50 Colombian children from 6 to 16 years old, with a diagnosis of behavioral disorders, which were compared with 50 controls. The genetic analysis was carried out by conventional PCR (polymerase chain reaction) and real-time RT-PCR (TaqMan probe). The statistical analysis by means of the PLINK program allowed us to make a determination of the Hardy-Weinberg equilibrium, establish the p-value and Odd Ratio of association of the study variants. The results obtained permitted us to conclude that there is no association of any of the analyzed markers with childhood behavior disorders, since they presented p-values, p> 0.05. On the other hand, all markers were found in Hardy Weinberg equilibrium, except SLC1A1. However, all markers presented an Odd Ratio greater than 1, which enabled us to infer that there is a probability of a significant association with the behavioral disorder such as ADHD, OCD, and ASD.     La Leishmaniasis cutánea es producida principalmente por el parásito Leishmania braziliensis. La OMS estima que se presentan entre 0,7 y 1,3 millones de casos nuevos al año por lo tanto se ha convertido en un problema de salud pública. Para su estudio este trabajo se planteó como objetivo el de caracterizar in silico 3 candidatos a transportadores de NAD+ en Leishmania braziliensis mediante análisis de estructura primaria, predicción de estructura secundaria y obtención de modelos predictivos de la estructura terciaria, con los que se identificaron 19 residuos conservados en los 3 candidatos de los cuales dos residuos están relacionados con el transporte de nucleótidos de adenina, asimismo se halló una alta probabilidad de presentar una regulación por fosforilación/desfosforilación pero no por acetilación/desacetilación. Adicionalmente se comprobó la presencia del dominio de la familia de “Transportadores mitocondriales” caracterizado por 6 hélices α transmembranales, las cuales también fueron reconocidas. Mediante los modelos de estructura terciaria se comprobó una alta identidad con el transportador mitocondrial de ADP/ATP de la especie Bos taurus lo cual sugiere una posible homología funcional y por lo tanto la capacidad de transportar nucleótidos de adenina específicamente NAD+ como en la translocasa de ATP/ADP de Clamidia trachomatis. Adicionalmente se comprobó la presencia de los genes de los 3 candidatos con tamaños de 1.089, 1.014 y 957 pb en el genoma de Leishmania braziliensis por métodos experimentales y se obtuvieron proteínas recombinantes para cada uno de ellos. Estos resultados constituyen evidencia sólida para continuar con estudios que permitan comprobar su funcionalidad.Cutaneous Leishmaniasis is mainly caused by the parasite Leishmania braziliensis. The OMS estimates that occur between 0.7 and 1.3 million new cases per year therefore it has become a public health problem. For its study, this paper aimed to characterize in silico 3 candidates to NAD+ transporters in Leishmania braziliensis by primary structure analysis, secondary structure prediction and obtaining predictive models of tertiary structure, with which 19 residues conserved in the three candidates are identified, of them two residues are related to the transport of adenine nucleotides, also a high probability to present a regulation by phosphorylation/dephosphorylation but not by acetylation/deacetylation was found. Additionally, the presence of the domain of the family of "Mitochondrial Carriers" characterized by α helices transmembrane 6 was checked. By models from tertiary structure high identity was found with ADP/ATP mitochondrial transporter of the Bos taurus specie, suggesting a possible functional homology and therefore the ability to transport adenine nucleotides specifically NAD+ as ATP/ADP translocase of Chlamydia trachomatis. In addition, the presence of the genes of the three candidates with sizes of 1089, 1014 and 957 bp was checked in the genome of Leishmania braziliensis by experimental methods and recombinant proteins for each were obtained.     Expression of sodium / iodine symporter (NIS) in cells derived from colon cancer: analysis of their effect on cellular glycolytic metabolism

    La proteína NIS es una glicoproteína localizada en la membrana basal de los tirocitos, que cataliza un transporte activo de yodo al interior de la célula. En los humanos dicha proteína se expresa, además de la tiroides, en las glándulas mamarias en periodos de lactancia y gestación, en glándulas salivales, en la mucosa gástrica, estómago, en cáncer de mama y en los ovarios. Estudios anteriores han demostrado que la transfección de NIS en células derivadas de diferentes tipos de cáncer permite la utilización de radioisótopos de yodo como una herramienta efectiva para el diagnóstico y tratamiento de este tipo de patologías, de forma similar a como se utiliza actualmente para el diagnóstico y tratamiento de patologías tiroideas. Sin embargo, para una óptima utilización de este nuevo tipo de tratamientos es importante comprender cómo la introducción de esta proteína puede afectar el metabolismo de las células cancerosas. En la Unidad TIRO-MATOs (Facultad de Medicina, Universidad de Niza, Francia), se desarrolló una línea de células de carcinoma de colon (HT29) que expresan de forma estable el gen nis, (HT29-NIS). A partir de esta línea celular, en el presente trabajo se evaluó el efecto de la transfección de la proteína NIS sobre el metabolismo glucolítico de las células HT29 sometidas a condiciones de hipoxia y normoxia, comparando la viabilidad y proliferación, y la captación de glucosa entre las células HT29 y HT29-NIS por medio de la sonda de glucosa 2-NBDG. También en este estudio se analizó la expresión de algunos genes implicados en la vía glucolítica celular como: GLUT-1, HK-1 y PK-M2 mediante RT-PCR. Los resultados determinaron que la viabilidad de las dos líneas celulares no se ve afectada significativamente por el tiempo de cultivo. Respecto a la captación de glucosa se encontró que el metabolismo de las células es heterogéneo, pero que al exponer las células a 24 horas de hipoxia este metabolismo tiende a homogenizarse ya que la mayor proporción de las células tiende a captar la misma concentración de glucosa, por otro lado se encontró que la línea celular HT29-NIS capta una mayor concentración de glucosa que la línea celular HT29-WT tanto en hipoxia como en normoxia. En cuanto a la transcripción de los genes analizados se observó un aumento en la expresión de cada uno de ellos en ambas líneas celulares luego del tratamiento con hipoxia, además que las células transfectadas con NIS presentan un aumento mayor de transcripción de los genes respecto a las células silvestres.NIS protein is a glycoprotein located in the basement membrane of thyrocytes, which catalyzes an active iodide transport into the cell. In humans this protein is also thyroid, mammary gland in pregnancy and lactation periods in salivary glands in the gastric mucosa, stomach, breast and ovarian expressed. Previous studies have shown that transfection of NIS in cells derived from different cancers allows the use of radioisotopes of iodine as an effective tool for diagnosis and treatment of such diseases, similar to as is currently used for diagnosis and treatment of thyroid diseases. However, for optimal use of this new type of treatment it is important to understand how the introduction of this protein may affect the metabolism of cancer cells. In the SHOT-Matos (Faculty of Medicine, University of Nice, France) unit, a cell line of colon carcinoma (HT29) stably expressing the nis gene was developed (HT29-NIS). From this cell line, in this study the effect of transfection of the NIS protein on the glycolytic metabolism of HT29 cells under normoxic and hypoxic conditions was assessed by comparing the viability and proliferation, and glucose uptake between HT29 and HT29-cells using NIS probe 2-NBDG glucose. Also in this study the expression of some genes involved was analyzed in cell glycolytic pathway as GLUT-1, HK-1 and PK-M2 by RT-PCR. The results determined that the viability of the two cell lines is not significantly affected by the culture time. Regarding glucose uptake was found that the metabolism of cells is heterogeneous, but at 24 hours after exposing cells hypoxia this metabolism tends to be homogenized as the larger proportion of the cells tend to capture the same glucose concentration, on the other hand it was found that the cell line HT29-NIS captures a greater glucose concentration that the cell line HT29-WT both normoxia and hypoxia.
