The impact of chemical fixation on the microanatomy of mouse brain tissue
Chemical fixation using paraformaldehyde (PFA) is a standard step for preserving cells and tissues for subsequent microscopic analyses such as immunofluorescence or electron microscopy. However, chemical fixation may introduce physical alterations in the spatial arrangement of cellular proteins, organelles and membranes. With the increasing use of super-resolution microscopy to visualize cellular structures with nanometric precision, assessing potential artifacts - and knowing how to avoid them - takes on special urgency.We addressed this issue by taking advantage of live-cell super-resolution microscopy that makes it possible to directly observe the acute effects of PFA on organotypic brain slices, allowing us to compare tissue integrity in a ‘before-and-after’ experiment. We applied super-resolution shadow imaging to assess the structure of the extracellular space (ECS) and regular super-resolution microscopy of fluorescently labeled neurons and astrocytes to quantify key neuroanatomical parameters.While the ECS volume fraction and micro-anatomical organization of astrocytes remained largely unaffected by the PFA treatment, we detected subtle changes in dendritic spine morphology and observed substantial damage to cell membranes. Our experiments show that PFA application via immersion does not cause a noticeable shrinkage of the ECS in brain slices, unlike the situation in transcardially perfused animals where the ECS typically becomes nearly depleted.In addition to the super-resolved characterization of fixation artefacts in identified cellular and tissue compartments, our study outlines an experimental strategy to evaluate the quality and pitfalls of various fixation protocols for the molecular and morphological preservation of cells and tissues
Super-Resolution Microscopy Opens New Doors to Life at the Nanoscale
Super-resolution fluorescence microscopy holds tremendous potential for discovery in neuroscience. Much of the molecular machinery and anatomic specializations that give rise to the unique and bewildering electrochemical activity of neurons are nanoscale by design, ranging somewhere between 1 nm and 1 μm. It is at this scale where most of the unknown and exciting action is and where cell biologists flock to in their dreams, but it was off limits for light microscopy until recently. While the optical principles of super-resolution microscopy are firmly established by now, the technology continues to advance rapidly in many crucial areas, enhancing its performance and reliability, and making it more accessible and user-friendly, which is sorely needed. Indeed, super-resolution microscopy techniques are nowadays widely used for visualizing immunolabeled protein distributions in fixed or living cells. However, a great potential of super-resolution microscopy for neuroscience lies in shining light on the nanoscale structures and biochemical activities in live-tissue settings, which should be developed and harnessed much more fully. In this review, we will present several vivid examples based on STED and RESOLFT super-resolution microscopy, illustrating the possibilities and challenges of nano-imaging in vivo to pique the interest of tech-developers and neurobiologists alike. We will cover recent technical progress that is facilitating in vivo applications, and share new biological insights into the nanoscale mechanisms of cellular communication between neurons and glia

Romanian Yoga School MISA and States of Enlightenment
The diploma thesis "MISA Yoga School and the States of Enlightenment" deals with the historical background and some aspects of spiritual teachings of MISA school established in 1990 in Bucharest. The first part gives a thorough overview of MISA's short history and proposes its division into consecutive periods. The crucial role of the founder and spiritual head, Gregorian Bivolaru, is assessed. In the second part, particular yogic principles characteristic for MISA are covered from both theoretical and practical perspective. The selected secondary sources and authentic experiences of yoga practitioners (including the author himself) are referred to. The work comprises four chapters supplemented with interviews with chosen yogis, the bibliography, the list of relevant websites and a brief English summary. The thesis has an ambition to stimulate further research in the given field

Misa Lingkungan yang Berdaya Ubah Bagi Persaudaraan Umat Beriman Menurut Kevin W. Irwin dan Paus Yohanes Paulus II
This article aims at examining the effects of misa lingkungan on the ecclesial life of parishioners. The question is whether misa lingkungan has any transformative power for building a communion of parishioners? As a theoretical concept, the author takes some ideas developed by Kevin W. Irwin in his book Models of the Eucharist, as well as some of the main ideas of Pope John Paul II's encyclical Ecclesia de Eucharistica. Based on these theological concepts, the author conducted interviews and questionnaires aimed at the parishioners of St. Aloysius Gozaga Parish, Mlati, Semarang Archdiocese. He tries to find out how far the adage lex orandi – lex credendi – lex vivendi influenced the understanding and the living of the parishioners. The author concludes that the Eucharist that is believed, lived, and celebrated has a positive impact on parishioners. The fruits of the Eucharist celebration were seen in their spirit of brotherhood and solidarity for those in need
