Characterization of age-related changes within the lysine acetylation pattern of immune effector cells
Im Laufe der Zeit ist der menschliche Organismus mehr Krankheitserregern ausgesetzt. Dadurch verringert sich die Reserve an naiven T-Zellen. Deacetylase-Inhibitoren (HDACis) wurden zur Behandlung einer Vielzahl an Krankheiten. Deshalb ist es unabdingbar, das Zusammenspiel zwischen Acetylierung und Alterung innerhalb von T-Zellen zu untersuchen. Zu diesem Zweck wurden CD8+-T-Zellen von jungen und alten Blutspendern isoliert und mit HDACi behandelt. Die funktionellen Auswirkungen auf T-Zellen Aktivierung wurden anschließend mittels biomolekularer Vorgehensweisen beurteilt. Es wurde eine Korrelation zwischen der HDACi-Behandlung und der Aktivierung von T-Zellen festgestellt. Während eine Erhöhung der Acetylierung die Apoptoserate der Zellen nicht stört, verändert sie das Cytoskelett und die Signalproteine.Over time, the human organism is exposed to pathogens, which reduces the reserve of naïve T cells. Deacetylase inhibitors (HDACis) are used to treat a wide range of conditions. Therefore, this work studies the interplay between acetylation and aging within T cells. To this end, CD8+ T cells were isolated from young and old donors and treated with HDACi. The functional effects on T cell activation were then evaluated through biomolecular approaches. To better understand the intracellular outcome of acetylation, high-throughput transcript analysis and protein expression profiles were also performed, along with the relative protein and transcript expression of proteins participating in the signaling pathways involved in the activation and differentiation of T cells. A correlation was found between HDACi treatment and T cell activation. While an increase in acetylation does not disturb the apoptosis rate of the cells, it alters the cytoskeleton and signaling proteins
Integrated Analysis of Energy, Material and Time Flows in Manufacturing Systems
AbstractEnvironmental objectives (e.g. energy and resource demand, emissions, waste) become increasingly relevant for manufacturing companies in addition to traditional economic objectives (e.g. throughput time, output). Currently, different methods and tools are available to address those objectives individually, such as value stream mapping (economic), material and energy flow analysis/MEFA as well as Life Cycle Assessment/LCA (environmental). However, there is a lack on approaches that bring together benefits of those tools and allow simultaneous consideration of all objectives. Against this background, a methodology is developed to analyses the energy, material and time flows of manufacturing systems in an integrated manner. The proposed method is exemplary applied to the case of an Australian manufacturing company

Dynamic Modular Architecture for Product Lifecycle
AbstractA module is a set of components with interfaces selected in order to help designers address ilities or non-functional system requirements. Consequently, the boundaries of a module do not necessarily coincide with those dictated by functional decomposition. Modularization usually makes the architecture more complex due to additional interfaces and redundancies that have negative consequences on system performance. As a result, modularization is accompanied by a trade-off between non-functional and functional requirements. Additionally the system lifecycle consists of several phases, each characterized by different activities and goals. Systems may benefit from different modular architectures in the different lifecycle phases. This paper presents a dynamic modular architecture methodology, where the modular architecture changes over the different product lifecycle phases. An example of a relatively simple mechanical system - a bicycle – is presented to illustrate the implementation of the methodology
Optimierung der Uterotomie-Wundheilung durch Intervention im Rahmen der Re-Sectio caesarea
Fragestellung: Kann intraoperativer Ultraschall der uterinen Narbenregion und Resektion im Rahmen der Re-Sectio caesarea die Anzahl an Doppelnarben vermindern und die Narbenarchitektur verbessern? Methoden: Probandinnen zur 1. und 2. Sectio bildeten respektive die Kontrollgruppe (CG) 1 bzw. 2. Die Interventionsgruppe (IG) erhielt zusätzlich einen intraoperativen Ultraschall sowie die Resektion der Narbenregion. Nach 6-9 Monaten erfolgte ein verblindeter Vaginalultraschall zur Beurteilung der Narbenanzahl und relativen Myometriumsdicke. Ergebnisse: Von 156 Rekrutierten wurden 124 nachuntersucht und116 Probandinnen analysiert. In der CG 1 wurde zu 100% 1 Narbe detektiert. In der CG 2 lagen in 42,9% Doppelnarben vor, welche in der IG signifikant auf 7,1% (p=0.002) gesenkt werden konnte. Für die relative Myometriumsdicke bei primären Sectiones zeigte sich eine signifikante Verbesserung (p<0.001) in der zweiseitigen ANOVA-Analyse zwischen der CG 2 und der IG. Zusammenfassung: Durch die o.g. Intervention kann die Anzahl an uterinen Doppelnarben reduziert und die Narbenarchitektur nach Re-Sectio caesarea verbessert werden.Objective: Can the number and architecture of uterine cesarean scars be improved by an intraoperative ultrasound guided identification of uterine scar tissue and its resection during repeat cesarean section. Methods: Participants with 1 and 2 Cesareans were enrolled in Control group 1 and 2 respectively. The interventiongroup received additionally an intraoperative ultrasound and resection of the detected scar tissue. 6-9 months postoperative a blinded transvaginal ultrasound was performed to evaluate the number and the deficiency ratio of the uterine scars. Results: 156 participants were recruited and 116 included in the analysis. In control group 1 in 100% one scar was detected. Participants of the control group 2 had double scarring in 42,9% of all cases in comparison to 7,1% (p= 0.002) in the intervention group. The two way ANOVA analysis showed a significant improvement (p < 0.001) in deficiency ratio between control group 2 and interventiongroup after adjustment for primary cesarean sections. Conclusion: The intervention leads to a reduction of uterine double scarring and improves the scar architecture after repeat Cesarean section
Current Status, Future Expectations and Mitigation Potential Scenarios for China's Primary Aluminium Industry
AbstractOver the past three decades, China has undergone a strong economic growth, largely industry-driven. The rise of consumption resulted in increasing material requirements like steel, cement, plastic and aluminium. Regarding aluminium, the in-use stock increased to 58.9kg/capita in 2009, from around 8.5kg/capita in 1989 and 19.4kg/capita in 1999. China's role in the aluminium industry is crucial. On its own, it produces around half of the world's primary aluminium output, destined for both domestic consumption and international export markets. However China's domestic bauxite reserves are limited and at current static exploitation would last for only 18 more years. Considering the low quality of its bauxite and the young and relatively low in-use stock level, China has to rely mainly on primary production, by heavily depleting its bauxite resources and by importing foreign bauxite and alumina. Primary aluminium production takes however a high environmental toll. This paper evaluates the effect of changes in: energy efficiency due to the technological level of both electricity and aluminium production, quality of resources and share of secondary and primary production; on the environmental impact due to the Chinese primary aluminium sector, by means of forecasting scenarios and mitigation potentials
